Mapping the alpha-subunit site photolabeled by the noncompetitive inhibitor [3H]quinacrine azide in the active state of the nicotinic acetylcholine receptor.

We have characterized the time-resolved labeling of a site on the Torpedo californica electrocyte acetylcholine receptor (ACHR) by the photoreactive noncompetitive inhibitor derivative quinacrine azide (QA). The dependence of [3H]QA labeling on acetylcholine (ACH) concentration and on time is consistent with the preferential labeling by [3H]QA of ACHR in the open state. The ACH-dependent [3H]QA labeling, which was associated predominantly with the alpha-subunit, was blocked by other noncompetitive inhibitors including quinacrine, chlorpromazine, proadifen, histrionicotoxin, and bupivacaine. al

High-Contrast PET Imaging of Vasopressin V1B Receptors with a Novel Radioligand, 11C-TASP699.

Vasopressin 1B receptors (V1BRs) are abundantly expressed in the pituitary, and in vivo PET of V1BRs was recently enabled by our development of a specific radioligand, 11C-TASP0434299, derivatized from pyridopyrimidin-4-one. Here, we identified a novel pyridopyrimidin-4-one analog, N-tert-butyl-2-[2-(6-11C-methoxypyridine-2-yl)-6-[3-(morpholin-4-yl)propoxy]-4-oxopyrido[2,3-d]pyrimidin-3(4H)-yl]acetamide (11C-TASP0410699, hereafter referred to as 11C-TASP699), as a potent V1BR radioligand producing a higher image contrast for the target than 11C-TASP0434299. Methods: In vitro properties of TASP

Purification and characterization of the glycoprotein hormone alpha-subunit-like material secreted by HeLa cells.

The protein secreted by HeLa cells that cross-reacts with antiserum developed against the alpha-subunit of human chorionic gonadotropin (hCG) has been purified approximately 30,000-fold from concentrated culture medium by organic solvent fractionation followed by ion exchange, gel filtration, and lectin affinity chromatography. The final preparation had a specific activity (by RIA) of 6.8 x 10(5) ng of alpha/mg of protein and appeared homogeneous by electrophoresis on reducing/denaturing polyacrylamide gels (SDS-PAGE). Amino acid analysis indicated that HeLa-alpha had a composition very simila

Syringol, a wildfire residual methoxyphenol causes cytotoxicity and teratogenicity in zebrafish model.

Natural toxicants, particularly methoxy phenols (MPs) generated by wildfire lignin, can accumulate in the environment, and cause serious health hazards in living organisms. Although the toxicity of MPs such as guaiacol and catechol has recently been described, there is minimal evidence of ecotoxicological effects of syringol. As a result, this study focuses on determining the toxicity by evaluating the cytotoxic and teratogenic effects of syringol in vitro and in vivo in human embryonic kidney (HEK-293) cells and zebrafish embryos, respectively. The ecotoxicity of syringol was predicted to be

Fusion-expressed CtxB-TcpA-C-CPE improves both systemic and mucosal humoral and T-cell responses against cholera in mice.

BACKGROUND: Development of an effective oral vaccine against Cholera, a life-threatening dehydrating diarrheal disease, proved to be a challenging task. To improve oral subunit vaccine immunogenicity and to prevent the state of oral tolerance, application of mucosal adjuvants might be a promising approach. In the present study, the CtxB-TcpA-C-CPE fusion was constructed in which CtxB and C-CPE were used as mucosal adjuvants and vaccine delivery system, respectively, to induce mucosal immune responses, and to improve the anti-toxin and anti-colonizing immunity against V. cholerae. Recombinant synthesis of insulin-like growth factor-binding protein-4 (IGFBP-4): Development, validation, and application of a radioimmunoassay for IGFBP-4 in human serum and other biological fluids.

Insulin-like growth factor-binding protein-4 (IGFBP-4), like the five other IGFBPs present in human serum, acts as a transport protein for insulin-like growth factor I (IGF-I) and IGF-II and modulates their biological effects. To investigate the role of IGFBP-4 in the physiology of the IGF system, we developed a sensitive RIA for IGFBP-4 employing, as antigen, tracer, and standard, recombinant human IGFBP-4 (rhIGFBP-4) expressed in Escherichia coli as a fusion protein with glutathione S-transferase and affinity purified with glutathione-derivatized resin. Antibody against the rhIGFBP-4 fusion

Structure and function studies of the cGMP-stimulated phosphodiesterase.

Studies of cGMP binding to both the native cyclic GMP-stimulated phosphodiesterase and to two unique isolated chymotryptic fragments lacking the catalytic domain suggest that the enzyme contains two noncatalytic cGMP-binding sites/homodimer. In the presence of high concentrations of ammonium sulfate, 2 mol of cGMP are bound/mol of cGMP-stimulated phosphodiesterase homodimer. Under these conditions, linear Scatchard plots of binding are obtained that give an apparent Kd of approximately 2 microM. The inclusion of 3-isobutyl-1-methylxanthine produces a curvilinear plot. In the absence of ammoniu

Development and diagnostic validation of a recombinant Hexon L1-based indirect enzyme-linked immunosorbent assay for detecting antibodies to fowl adenovirus serotype 8b in chickens.

Inclusion body hepatitis (IBH), caused by fowl adenoviruses (FAdVs), is a significant disease impacting poultry productivity. Recent data indicates that FAdV serotype 8b (FAdV-8b) is the predominant serotype associated with IBH cases in Thailand. Although the serum neutralization (SN) test is a commonly used method for detecting serotype-specific neutralizing antibodies, it has limitations in its routine application. This study developed and diagnostically validated an FAdV-8b-targeted indirect ELISA (iELISA) utilizing a recombinant protein of the FAdV-8b Hexon loop-1 region (rHexon8b), which

Recombinant L1R protein induces cross-neutralizing antibodies against capripoxviruses despite a subdominant humoral response.

Capripoxviruses, including the goatpox virus (GTPV), sheeppox virus (SPPV), and lumpy skin disease virus (LSDV), cause substantial economic losses in domestic ruminants in Asia, Africa, the Middle East, and Europe. The identification of viral antigens that elicit protective immunity, along with the characterization of antigen-specific humoral responses, is crucial for the rational development of vaccines and serodiagnostic assays. L1R is an essential membrane protein of the intracellular mature virion (IMV) and a known neutralizing antibody target for poxviruses. In this study, a truncated L1R

Characterization of 2-deoxyribose-5-phosphate aldolase from Bacillus altitudinis JYY-02.

HMG-CoA reductase inhibitors, commonly known as statins, effectively reduce low-density lipoprotein cholesterol levels in the blood, thereby playing a key role in the treatment of cardiovascular and cerebrovascular diseases. The biosynthesis of the compounds can be achieved via deoxyriboaldolase. In this study, a 672-bp gene coding 2-deoxyribose-5-phosphate aldolase (DERA) was identified from the genome of Bacillus altitudinis JYY-02 and overexpressed in Escherichia coli BL21(DE3). The recombinant DERA exhibited a molecular weight of 23.15 kDa and was purified by Ni-NTA affinity chromatography

In vitro and in silico characterization of Crocatin A from red betel leaves: targeting DNA gyrase B and DNA ligase of Enterococcus faecalis with ADMET-based druglikeness analysis.

Dental caries is a disease of tooth tissue that can lead to complications, with Enterococcus faecalis being one of the Gram-positive bacteria that plays a crucial role in the infection process. Red betel leaves (Piper crocatum Ruiz & Pav.) are known to contain various phytochemicals; however, the antibacterial potential of Crocatin A, one of its isolated compounds, against E. faecalis and its molecular targets has not been previously reported. This study hypothesized that Crocatin A exhibits antibacterial activity against E. faecalis and has the potential to inhibit DNA gyrase B and DNA ligase

α-Glucosidase Inhibitory Potential of Citrus reticulata Peel-Derived Flavonoids-A Prelude for the Management of Type 2 Diabetes.

α-Glucosidase inhibitors (AGIs) are compounds used to treat type 2 diabetes (T2D) by preventing the breakdown of dietary starch into monosaccharides, which reduces their absorption by the body and lowers blood glucose levels. AGIs often cause gastrointestinal issues such as diarrhea and flatulence due to excessive α-amylase inhibition, leading to excess residual starch reaching the colon and being fermented by microbes. There is a need to prospect for novel AGIs that are effective and have fewer adverse effects. This study investigated the potential of citrus-derived flavonoids as AGIs targeti

Expression and immunoassay assessment of immunomodulatory PanDR and SARS-CoV-2 receptor-binding domain antigen in bacteria host cell.

The gap in technical capacity and inadequate infrastructure for biomanufacturing limits progress in vaccine development and biomanufacturing in low- and middle-income countries (LMICs). Peptide subunit antigens offer a promising solution to producing cost-effective, safe and protective vaccines for SARS-CoV-2 and other emerging viral diseases. The receptor-binding domain (RBD) of the SARS-CoV-2 spike protein, which mediates viral entry through interaction with the human angiotensin-converting enzyme 2 (ACE2) receptor, represents a suitable model antigen for building and strengthening local cap

Biparatopic binding of ISB 1442 to CD38 in trans enables increased cell antibody density and increased avidity.

ISB 1442 is a bispecific biparatopic antibody in clinical development to treat hematological malignancies. It consists of two adjacent anti-CD38 arms targeting non-overlapping epitopes that preferentially drive binding to tumor cells and a low-affinity anti-CD47 arm to enable avidity-induced blocking of proximal CD47 receptors. We previously reported the pharmacology of ISB 1442, designed to reestablish synthetic immunity in CD38+ hematological malignancies. Here, we describe the discovery, optimization and characterization of the ISB 1442 antigen binding fragment (Fab) arms, their assembly to

Functional expression of five refolded recombinant variants of RBD from SARS-CoV-2 in Escherichia coli.

Production of the receptor binding domain, RBD, variants of SARS-CoV-2, has been necessary to understand their interaction with the human receptor ACE2 and to develop alternative strategies against COVID-19. Affordable bacterial production can aid in these efforts and offer alternatives to address the ongoing emergence of new variants. Expressing recombinant RBD in E. coli could provide a valuable alternative if the challenges related to disulfide bond formation, low solubility, and the absence of glycosylation capabilities are addressed in this heterologous expression system. In this study, f
