Innate and Adaptive Immunity in Giant Cell Arteritis.

Autoimmune diseases can afflict every organ system, including blood vessels that are critically important for host survival. The most frequent autoimmune vasculitis is giant cell arteritis (GCA), which causes aggressive wall inflammation in medium and large arteries and results in vaso-occlusive wall remodeling. GCA shares with other autoimmune diseases that it occurs in genetically predisposed individuals, that females are at higher risk, and that environmental triggers are suspected to beget the loss of immunological tolerance. GCA has features that distinguish it from other autoimmune disea

Neutrophil-associated plasma proteomics identifies HDAC1 as a baseline biomarker of immune tolerance during immunosuppressant withdrawal after pediatric liver transplantation: a single-center cohort study.

BACKGROUND: In pediatric liver transplantation (pLT), long-term immunosuppression (IS) contributes to infection risk and chronic toxicity, yet IS minimization or withdrawal requires balancing rejection risk. Practical biomarkers for baseline risk stratification at the start of planned withdrawal remain scarce. This study investigated whether baseline neutrophil-associated proteomic signatures and histone deacetylase 1 (HDAC1) levels are associated with IS withdrawal outcomes. METHODS: Within an institutional IS withdrawal program (n = 77), 59 recipients had evaluable outcomes by the follow-up

Transcriptome changes in leukocytes of dairy calves exposed to heat stress.

Heat stress impacts the health and immune function of dairy calves. Our objective was to conduct a longitudinal study to assess temporal alterations in the transcriptome of white blood cells from dairy calves exposed to heat stress. Eight Holstein bull calves (68.5 ± 1.37 kg of BW, aged 3.5 ± 0.5 weeks) were raised in individual pens following current commercial management practices. Calves were exposed to cyclic heat stress treatment (40°C ambient temperature from 0800 to 1900 h daily followed by 27°C) for 5 days. Blood samples were collected at 0, 1, and 5 days relative to treatment to condu

Endotheliopathy within the thromboinflammatory network of sepsis-induced coagulopathy.

BACKGROUND: Sepsis represents a life-threatening syndrome driven by dysregulated host responses in which inflammation, coagulation, and endothelial dysfunction intersect. Increasing evidence suggests that endotheliopathy is a dynamically interacting component linking systemic inflammation, immunothrombosis, and microvascular dysfunction. Rather than acting as a singular upstream trigger, endothelial dysfunction evolves in parallel with platelet, neutrophil, and monocyte activation, forming a self-amplifying thromboinflammatory network that drives organ failure. MAIN TEXT: The vascular endothe

Clonal Hematopoiesis of Indeterminate Potential (CHIP): A Model of Mutation-Driven Thromboinflammation.

Clonal hematopoiesis refers to the clonal expansion of hematopoietic stem and progenitor cells, driven by somatic mutations. Major mutated genes in clonal hematopoiesis include genes involved in epigenetic regulation including DNA methylation and/or chromatin modification (e.g., DNMT3A, TET2, and ASXL1), tumor suppressors (e.g., TP53), signal transduction (e.g., JAK2), and RNA splicing (e.g., SF3B1 and SRSF2). Clonal hematopoiesis includes clonal hematopoiesis of indeterminate potential (CHIP), clonal cytopenia of unknown significance (CCUS), and myelodysplastic syndromes/neoplasms (MDS). CHIP

ZIF-8-Based Nanomedicine for Bone Regeneration: A Critical Appraisal of Preclinical Osteoimmunomodulatory Evidence and Translational Readiness.

Zeolitic imidazolate framework-8 (ZIF-8)-based biomaterials are increasingly investigated for bone repair because they combine cargo loading, environment-dependent degradation, surface engineering, and bioactive Zn2+ release. However, recent reviews already catalogue the multifunctional design and orthopedic applications of ZIF-8, while the directness of evidence supporting specific osteoimmunomodulatory claims remains less clear. This structured narrative review critically appraises preclinical ZIF-8 literature using a predefined evidence-classification framework that separates direct ZIF-8 e

M-CSF priming enhances the efficiency and reproducibility of hiPSC-derived osteoclast differentiation and pharmacological responsiveness to anti-resorptive agents.

Human induced pluripotent stem cell (hiPSC)-derived osteoclasts offer a potentially scalable platform for studying osteoclast biology and anti-resorptive pharmacology. However, differentiation efficiency and inter-line reproducibility remain limited. To address this limitation, we investigated whether cytokine priming during the monocyte-to-macrophage transition is an important determinant of efficient and reproducible osteoclast differentiation from hiPSCs across diverse genetic backgrounds. Monocytes derived from three independent hiPSC lines were primed briefly with M-CSF, GM-CSF, or no cyt

Redefining Difficult-to-Treat Systemic Lupus Erythematosus: Biomarkers of Molecular Refractoriness Beyond Clinical Failure.

Difficult-to-treat systemic lupus erythematosus (D2T-SLE) remains a major unmet challenge in contemporary lupus care, yet it continues to be defined predominantly by clinical non-response rather than underlying biology. Current biomarkers largely quantify inflammatory burden, immune complex activity, or organ damage and do not reliably capture persistent activation of pathogenic pathways under therapy. Emerging multi-omics, single-cell, and longitudinal studies suggest that, in a subset of patients, apparent treatment failure may reflect incomplete attenuation of dominant immune circuits rathe

Host immune determinants of stromal vascular fraction graft survival: Toward a concept of SVF therapy resistance - A systematic narrative review.

Stromal vascular fraction (SVF)-based therapies and autologous fat grafting have emerged as promising regenerative strategies due to their pro-angiogenic, immunomodulatory, and trophic properties. However, despite encouraging preclinical and clinical findings, therapeutic outcomes remain highly heterogeneous, with marked variability in graft retention and functional efficacy between patients. Increasing evidence suggests that this variability cannot be explained solely by procedural factors or cellular composition, but may also depend on host-related immune and microenvironmental determinants.

Beyond the Neutrophil Count: Functional Profiling and Targeted Modulation of Neutrophils in Paediatric Care.

BACKGROUND: Neutrophils are the most abundant leukocytes in childhood and form a critical component of innate immunity, particularly in early life when adaptive responses are still maturing. In paediatrics, both quantitative and qualitative disturbances in neutrophil number or function shape susceptibility to infection and inflammatory tissue injury. Emerging data from neonatal, sepsis, and allergic cohorts show that paediatric neutrophils exhibit distinct effector profiles and regulatory roles compared with adults, positioning them as central mediators of child health and disease. Plasma Exosomal-Derived SERPINA1 and GNAI2 Downregulation as Potential Diagnostic Biomarkers of Kawasaki Disease with Coronary Artery Aneurysms.

Kawasaki disease (KD) with coronary artery aneurysms (CAAs) is currently the primary cause of childhood acquired heart disease with an unclear pathogenesis. We established five groups for the discovery of differentially expressed proteins (DEPs): healthy control, febrile control, KD without CAAs, KD with small and medium CAAs, and KD with giant CAAs (n = 8 in each group). The validation of selected DEPs was conducted in another five groups (n = 4 in each group). We conducted comprehensive bioinformatics analyses to elucidate the functional roles of the DEPs in the groups of KD with CAAs and KD

Inhalation of dust accumulated on personal computer fans in the office may cause pathological effects by disrupting the metabolic activity of alveolar macrophages.

Considering that computer fans draw in dust from the office air, and that printers and copiers are typically placed near computers, we collected dust that accumulated on computer fans from three commercial personal computer centers to predict the health effects of inhaled office dust (OD). OD contained various types of microplastics and non-protein particles, and several hydrophilic elements were bound to the OD. When aspirated OD via the pharynx for 90 days, blood levels of IP and K significantly decreased in male mice, but not in female mice. Meanwhile, the total number of pulmonary cells in

Evidence for HCO3- and NH3/NH4+-dependent pH regulatory mechanisms in the alkaline midgut of the sea urchin larva.

Alkaline digestive systems are well described for some insect species and their larval stages. More recently, larvae of the members of ambulacraria superphylum consisting of echinoderms and hemichordates were also discovered to have highly alkaline midguts (pH 9.5-10.5) with the underlying acid-base regulatory mechanisms largely unknown. Using pharmacological inhibition of acid-base transporters in conjunction with ion-selective microelectrode measurements and pH-sensitive dyes, we investigated intracellular and extracellular pH regulatory mechanisms of midgut epithelial cells of a sea urchin

Stage-Specific Proteomic Profiles in Dental Caries.

This study investigated the proteomic landscape of sound and carious coronal dentin to uncover the molecular signatures of host response, including tissue degradation, inflammation, and repair, across progressive stages of caries lesions. Dentin from deidentified human molars, grouped into 6 clusters of 3 teeth each, was pulverized to obtain ~1 g of tissue per cluster (n = 6). G1 and G2 protein extracts were obtained using guanidine before and after demineralization. Extracts from sound (S), distinct dentin caries (DDC), and extensive dentin caries (EDC) lesions were digested with trypsin and

Proinflammatory bone marrow niches and neutrophil activation are associated with TIGIT expression in multiple myeloma.

Multiple Myeloma (MM) is the second most frequent hematologic malignancy in adults in the Western world. The bone marrow microenvironment plays a critical role in MM progression by supporting malignant plasma cell (PC) survival, immune evasion, and proliferation. Among immune checkpoints, TIGIT has recently emerged as a relevant immunoregulatory molecule, capable of dampening cytotoxic responses and enhancing immune suppression. We investigated the expression of TIGIT in the bone marrow of newly diagnosed MM patients and its correlation with disease characteristics and microenvironmental featu

c-Myc Drives inflammation of the maternal-fetal interface, and neonatal lung remodeling induced by intra-amniotic inflammation.

Background: Intra-amniotic inflammation (IAI) is associated with increased risk of preterm birth and bronchopulmonary dysplasia (BPD), but the mechanisms by which IAI leads to preterm birth and BPD are poorly understood, and there are no effective therapies for preterm birth and BPD. The transcription factor c-Myc regulates various biological processes like cell growth, apoptosis, and inflammation. We hypothesized that c-Myc modulates inflammation at the maternal-fetal interface, and neonatal lung remodeling. The objectives of our study were 1) to determine the kinetics of c-Myc in the placent

Effects of the N-Butanol Extract of Pulsatilla Decoction on Neutrophils in a Mouse Model of Ulcerative Colitis.

Ulcerative colitis (UC) is a chronic inflammatory disease, the incidence of which is increasing worldwide. However, the etiology and pathogenesis of UC remains unclear. The n-butanol extract of Pulsatilla decoction (BEPD), a traditional Chinese medicine, has been shown to be effective in treating UC. This study aimed to explore the molecular mechanism underlying the effects of BEPD on UC, in particular its effects on neutrophil extracellular trap (NET) formation by neutrophils. High-performance liquid chromatography was used to determine the principal compounds of BEPD. UC was induced in mice

Age-dependent inflammatory response is altered in an ex vivo model of bacterial pneumonia.

BACKGROUND: Aging is associated with an increased incidence and mortality of Pseudomonas aeruginosa-induced pneumonias. This might be partly due to age-dependent increases in inflammatory mediators, referred to as inflamm-aging and a decline in immune functions, known as immunosenescence. Still, the impact of dysregulated immune responses on lung infection during aging is poorly understood. Here, we aimed to mimic inflamm-aging using ex vivo precision-cut lung slices (PCLS) and neutrophils - as important effector cells of innate immunity - from young and old mice and investigated the influence

Identification of co-expressed central genes and transcription factors in acute myocardial infarction and diabetic nephropathy.

BACKGROUND: Acute myocardial infarction (AMI) and diabetic nephropathy (DN) are common clinical co-morbidities, but they are challenging to manage and have poor prognoses. There is no research on the bioinformatics mechanisms of comorbidity, and this study aims to investigate such mechanisms. METHODS: We downloaded the AMI data (GSE66360) and DN datasets (GSE30528 and GSE30529) from the Gene Expression Omnibus (GEO) platform. The GSE66360 dataset was divided into two parts: the training set and the validation set, and GSE30529 was used as the training set and GSE30528 as the validation set. A

The mammosphere-derived epithelial cell secretome modulates neutrophil functions in the bovine model.

BACKGROUND: Innovative therapies against bacterial infections are needed. One approach is to focus on host-directed immunotherapy (HDT), with treatments that exploit natural processes of the host immune system. The goals of this type of therapy are to stimulate protective immunity while minimizing inflammation-induced tissue damage. We use non-traditional large animal models to explore the potential of the mammosphere-derived epithelial cell (MDEC) secretome, consisting of all bioactive factors released by the cells, to modulate host immune functions. MDEC cultures are enriched for mammary ste
