Oral argan oil attenuates gastric mucosal injury and shapes macrophage effector functions in a preclinical ethanol-induced ulcer model.

BACKGROUND: Macrophages are central to mucosal defense and key regulators of tissue homeostasis and inflammation, representing important targets for immunomodulatory strategies in gastric injury. This study investigated the modulatory effects of argan oil on macrophage effector functions, focusing on lipid droplet (LD) dynamics and macrophage extracellular trap (METosis)-associated cellular events under basal and ethanol-induced gastric ulcer conditions. METHODS: In a preclinical randomized ethanol-induced gastric ulcer model, peritoneal resident macrophages (PRMϕs), a well-established primar

Mechanism of Lipi Jiangzhuo Decoction in Improving Metabolic Dysfunction-Associated Steatohepatitis Through the PERK/PINK1/GPx4 Pathway.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is characterized primarily by hepatocyte lipoapoptosis and hepatic inflammation, frequently developing from overweight/obesity. To date, no specific therapeutics exist to reverse MASH. Although resmetirom has been approved in some regions, patients in many Asian countries, including China, still lack access to approved pharmacotherapy for MASH. Lipi Jiangzhuo decoction (LPJZD) is a promising traditional Chinese medicine formula for MASH. However, to date, there have been no comprehensive studies clarifying its potential mechan

FABP4-dependent fatty acid oxidation-fueled mitochondrial ROS induces the mobilization of cellular iron and facilitates Trypanosoma cruzi proliferation in murine adipocytes.

Fatty acid-binding protein 4 (FABP4) is a cytosolic lipid chaperone predominantly expressed in adipocytes. It has been shown that Trypanosoma cruzi targets adipose tissues and resides in adipocytes. However, how T. cruzi manipulates adipocytes to redirect nutrients for its benefit remains unknown. Here, we uncover the role of FABP4 in facilitating T. cruzi infection in murine 3T3-L1 adipocytes. We demonstrate that pharmacological or genetic inhibition of FABP4, carnitine palmitoyltransferase I (CPT-1), or fatty acid oxidation (FAO) abrogates the intracellular growth of T. cruzi in adipocytes.

Metabolic Reprogramming by Time-Restricted Feeding Confers Resistance to PM2.5-Induced Metabolic Disturbances in Adipose Tissue.

Recent research indicates that exposure to fine particulate matter (PM2.5) not only impairs the respiratory system but also disturbs adipose tissue homeostasis, contributing to rising obesity rates and systemic metabolic disturbances. Time-restricted feeding (TRF), a dietary approach that confines daily food consumption to specific time windows, has demonstrated significant potential in preventing and managing metabolic disorders. However, it remains unclear if TRF can alleviate PM2.5-induced adipose tissue dysfunction, and the mechanisms involved require investigation. We established a PM2.5-

Hepatic Overexpression of a Novel Soluble Endogenous Transforming Growth Factor β Type II Receptor-Immunoglobulin G1 Fc Fusion Protein (TβRII-SE/Fc) Attenuates Early Stage Metabolic Dysfunction-Associated Steatotic Liver Disease in Rats.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease. Due to its complex pathophysiology, treatment options remain limited. Transforming growth factor β (TGF-β) is a pleiotropic cytokine involved in the regulation of liver fibrosis and metabolism. Previously, we have demonstrated the antifibrotic effect of TβRII-SE/Fc delivered by a lentiviral vector (Lv) in a liver fibrosis model. TβRII-SE/Fc is a fusion protein composed of the soluble isoform of the TGF-β type II receptor (TβRII-SE) and the human IgG-Fc domain. In this study, we evaluat

Suppression of PKM2 by Leonurine Attenuates Alcoholic Hepatitis Through Regulating Glycolytic Reprogramming.

Alcoholic hepatitis (AH) is a liver disease caused by long-term heavy drinking. In the early stages, it usually manifests as fatty liver, which can then develop into liver fibrosis and cirrhosis. Kupffer cells are liver resident macrophages and the effect of glycolytic reprogramming on the polarization of macrophages plays an important role in hepatitis, which maybe a possibility therapeutic target for treating AH. Leonurine (Leo) is a natural alkaloid from Leonurus japonicus Houtt. and has been commonly used to ameliorate metabolic disorders. However, the protective effects and underlying mec

Time-restricted eating promotes sustained fat loss in Drosophila.

While current therapeutics restricting calorie intake, such as GLP-1 agonists, induce fat loss for many people, they are ineffective for others and concerns remain about their long-term effects on health, particularly loss of lean muscle mass. Moreover, many quickly regain fat if they stop treatment. In contrast, time-restricted eating does not restrict calorie intake but instead restricts the time window for eating and prevents obesity in mice and humans. Here we investigated the effects of intermittent T ime- R estricted F eeding (iTRF), which extends lifespan and delays markers of aging, on

Environmental endocrine disruptors activate CBX2 to disrupt cholesterol homeostasis in PCOS.

Polycystic ovary syndrome (PCOS) is a common endocrine disorder influenced by genetic and environmental factors, yet the molecular mechanisms linking endocrine-disrupting chemicals (EDCs) to ovarian dysfunction remain unclear. Here, we establish a letrozole-induced PCOS-like rat model and integrate bulk RNA-seq, single-cell RNA-seq of human granulosa cells (GCs), and functional validation to identify CBX2 as a critical epigenetic regulator connecting EDC exposure to PCOS pathogenesis. Transcriptomic profiling revealed dysregulated cell cycle progression, disrupted cholesterol homeostasis, and

4-phenylbutyric acid attenuates hepatic ER stress, pyroptosis, and NAFLD in adult rat offspring exposed to a high-fat, high-fructose diet during perinatal and adulthood.

BACKGROUND: Perinatal exposure to high-fat and/or high-fructose diets affects offspring metabolic programming, increasing the risk of metabolic disorders such as non-alcoholic fatty liver disease (NAFLD). In addition, re-exposure to these diets during adulthood exacerbates metabolic disturbances, indicating an interaction between early- and later-life diets in disease development. Therefore, the present study was designed to assess the effects of high-fat, high-fructose (HFF) diet exposure during the perinatal period and adulthood on hepatic endoplasmic reticulum (ER) stress, pyroptosis, and N

High-pressure homogenised rice bran nanofibers as stabilisers in pickering emulsions: Interfacial behaviour, emulsification efficiency and oxidative stability.

Rice bran nanofibers (RBNFs), derived from rice bran (RB), were investigated as sustainable, food-grade stabilizers for Pickering emulsions (PEs). RBNFs were extracted via sequential chemical treatments followed by high-pressure homogenization (HPH), designed to enhance their surface activity and interfacial performance. After 5 HPH passes at 150bars, the nanofibers exhibited an average diameter of 32 nm and significantly improved rearrangement kinetics at air-water, and oil-water interfaces (-43.67 x 10-4s-1 and -20.6 x 10-4s-1, respectively), indicating a higher affinity for interfacial rear

Oleogel fabrication from Lactobacillus-stabilized emulsion templates: The role of bacterial surface properties in network stabilization mechanisms.

Lactobacillus cells, owing to their combined interfacial stabilizing and potentially postbiotic functionalities, have emerged as promising natural Pickering stabilizers. However, the relationship between bacterial surface properties, emulsion-template stability, and oleogel formation remains poorly understood. In this study, eight Lactobacillus strains with distinct surface properties were employed to subsequently construct Pickering emulsion templates and oleogels. Principal component analysis revealed distinct relationships among bacterial surface properties and emulsion-template characteris

Novel Potential Markers of Metabolic Dysfunction-Associated Steatohepatitis Prone to Hepatocellular Carcinoma.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH), the progressive phenotype of metabolic dysfunction-associated steatotic liver disease (MASLD), is well recognized for its increased risk of progressing to cirrhosis and hepatocellular carcinoma (HCC). However, early diagnosis and identification of the MASH patients with a tendency toward malignancy HCC remain a significant challenge. METHOD: In previous studies, we established a novel conditional inducible HRAS gene expression murine model with normal diet and lifestyle conditions, which exhibited 100% HCC incidence and reca

Fetal sex shapes maternal immune adaptation: placental extracellular vesicles differentially reprogram the phenotype, metabolism, and function of circulating monocytes.

INTRODUCTION: Maternal immune adaptation during pregnancy is orchestrated by dynamic signals from the uterine microenvironment, including placental extracellular vesicles (pEVs) released into maternal circulation. EVs have emerged as key mediators of this crosstalk; however, their role in sex-specific immune modulation remains incompletely defined. Here, we investigated whether pEVs derived from term placentas induce sex-dependent changes in the phenotype, metabolism, and function of human monocytes. METHODS: pEVs were isolated from 13 term uncomplicated placentas (six male-derived, M-pEVs, a

Exercise-diet intervention ameliorates but fails to fully reverse obesity-induced ovarian dysfunction: evidence spanning folliculogenesis to embryonic development.

Obesity, a globally prevalent chronic disease, disrupts systemic homeostasis and impairs female fertility, yet the mechanisms linking adipose dysfunction to ovarian reserve remain unclear. Using high-fat diet-induced obese C57BL/6 mouse models (HFD) and exercise-diet intervention models (SE group), we systematically evaluated obesity-associated reproductive deficits. Histomorphological analysis revealed that HFD mice exhibited ovarian atrophy, increased atretic follicles, and reduced primordial/antral follicle counts, which were partially restored by SE intervention. TEM demonstrated lipid dro

Microglial glycolytic reprogramming in alzheimer's disease: association with impaired phagocytic function and altered vascular proximity.

BACKGROUND: Alzheimer's disease (AD) is characterized by chronic neuroinflammation alongside amyloid-beta plaque and phosphorylated tau (p-Tau) tangle accumulation. Microglia, as resident immune cells, undergo glycolytic reprogramming that may exacerbate inflammation and impede toxic protein clearance. Specifically, the glycolytic enzyme pyruvate kinase M2 (PKM2) drives proinflammatory microglial phenotypes linked to neurodegeneration. This study investigates how PKM2-mediated microglial glycolytic reprogramming and inflammatory responses alongside Aβ/p-Tau clearance in human AD brains. METHO

Validated Stability-Indicating RP-HPLC Method for Nirmatrelvir in Self-Emulsifying Drug Delivery Systems: Formulation Characterization and Permeability Enhancement.

This study presents a novel analytical and formulation strategy to enhance the oral delivery and quality assessment of Nirmatrelvir, a poorly water-soluble antiviral agent. A self-emulsifying drug delivery system (SEDDS) was developed using Labrafac MC 60, ethanol, and Transcutol HP (55:25:20), resulting in a nanoemulsion with a droplet size of 145.23 ± 3.23 nm, low polydispersity index (0.189 ± 0.023), and high transmittance (98.97 ± 0.25%). The formulation exhibited rapid emulsification (< 90 s) and significantly improved permeability, achieving a fivefold increase (Papp: 4.20 × 10-6 cm/s) a

Analysis of effector/memory regulatory T cells from arrhythmogenic cardiomyopathy patients identified IL-32 as a novel player in ACM pathogenesis.

Arrhythmogenic cardiomyopathy (ACM) is an inherited cardiac disorder that causes sudden cardiac death and progressive heart failure. Besides fibro-fatty replacement and myocyte degenerative changes, inflammatory patchy infiltrates are found in myocardial histological analysis of ACM patients. Inflammatory cells could actively participate in ACM pathogenesis, contributing to the alteration of cardiac microenvironment homeostasis, thus triggering disease evolution. In order to characterize the immune-derived mediators involved in ACM pathogenesis, peripheral blood mononuclear cells from ACM pati

Methylation of lncSHGL promotes adipocyte differentiation by regulating miR-149/Mospd3 axis.

Obesity poses significant health risks and can negatively impact an individual's quality of life. The human obesity phenotype results from the differentiation of pre-adipocytes into adipocytes, which leads to hypertrophy and hyperplasia in adipose tissue. The molecular mechanisms by which long non-coding RNAs (lncRNAs) modulate adipocyte differentiation, a process implicated in obesity development, remain poorly characterized. A lncRNA which suppressed the hepatic gluconeogenesis and lipogenesis (lncSHGL) was newly identified. Our research aims to elucidate the functional role and mechanistic

Essential oil of oregano (Origanum vulgare L.) reduces infection and proliferation of Toxoplasma gondii in BeWo cells with induction of autophagy and death of tachyzoites through a mechanism similar to necrosis.

Toxoplasmosis poses a global health threat, ranging from asymptomatic cases to severe, potentially fatal manifestations, especially in immunocompromised individuals and congenital transmission. Prior research suggests that oregano essential oil (OEO) exhibits diverse biological effects, including antiparasitic activity against Toxoplasma gondii. Given concerns about current treatments, exploring new compounds is important. This study was to assess the toxicity of OEO on BeWo cells and T. gondii tachyzoites, as well as to evaluate its effectiveness in in vitro infection models and determine its

Carvacrol-loaded emulsions stabilized by soy protein isolate/chitosan oligosaccharide conjugates improved the quality of refrigerated sea bass (Lateolabrax maculatus).

In this study, the soybean protein isolate / chitosan oligosaccharide (SPI/COS) conjugates were developed using glycosylation and used as a stabilizer of carvacrol-loaded emulsions for refrigerating sea bass (Lateolabrax maculatus). The results of Fourier transform infrared spectroscopy and fluorescence spectroscopy showed that COS modified the secondary and tertiary structure of SPI by covalent binding. The SPI/COS conjugate with a mass ratio of SPI and COS of 4:1 (SPI/COS-4) exhibited high glycosylation degree, great water solubility and better emulsification properties. Carvacrol-loaded emu
