Microanalytical Mass Spectrometry with Super-Resolution Microscopy Reveals a Proteome Transition During Development of the Brain's Circadian Pacemaker.

During brain development, neuronal proteomes are regulated in part by changes in spontaneous and sensory-driven activity in immature neural circuits. A longstanding model for studying activity-dependent circuit refinement is the developing mouse visual system where the formation of axonal projections from the eyes to the brain is influenced by spontaneous retinal activity prior to the onset of vision and by visual experience after eye-opening. The precise proteomic changes in retinorecipient targets that occur during this developmental transition are unknown. Here, we developed a microanalytic

Philadelphia chromosome-negative myeloproliferative chronic neoplasms: is clonal hematopoiesis the main determinant of autoimmune and cardio-vascular manifestations?

In this article, we reviewed the possible mechanisms linking the clonal hematopoiesis of indeterminate potential (CHIP) to chronic myeloproliferative neoplasms (MPNs), autoimmune diseases (ADs), and cardiovascular diseases (CADs). CHIP is characterized by the presence of clonal mutations with an allelic frequency >2% in the peripheral blood without dysplasia, overt hematological neoplasms, or abnormalities in blood cell count. The prevalence may reach 20% of elderly healthy individuals and is considered a risk factor for myelodysplastic neoplasms and acute leukemia. In MPNs, CHIP is often asso

Identification of Neutrophil-Related Factor LCN2 for Predicting Severity of Patients With Influenza A Virus and SARS-CoV-2 Infection.

BACKGROUND: Influenza and COVID-19 are respiratory infectious diseases that are characterized by high contagiousness and high mutation and pose a serious threat to global health. After Influenza A virus (IAV) and SARS-CoV-2 infection, severe cases may develop into acute lung injury. Immune factors act as an important role during infection and inflammation. However, the molecular immune mechanisms still remain unclear. We aimed to explore immune-related host factors and core biomarker for severe infection, to provide a new therapeutic target of host factor in patients. METHODS: Gene expression

Microvesicles in plasma reflect coronary flow reserve in patients with cardiovascular disease.

High levels of microvesicles (MVs), a type of extracellular vesicles, are detected in several pathological conditions. We investigated the connection between coronary flow reserve (CFR), a prognostic clinical parameter that reflects blood flow in the heart, with levels of MVs and their cargo, from plasma of patients with cardiovascular disease. The PROFLOW study consists of 220 patients with prior myocardial infarction and measured CFR with transthoracic echocardiography. The patients were divided into high and low CFR groups. Plasma MVs were captured with acoustic trapping. Platelet- and endo

Longitudinal characterization of circulating neutrophils uncovers distinct phenotypes associated with disease severity in hospitalized COVID-19 patients.

Multiple studies have identified an association between neutrophils and COVID-19 disease severity; however, the mechanistic basis of this association remains incompletely understood. Here we collected 781 longitudinal blood samples from 306 hospitalized COVID-19 + patients, 78 COVID-19 âˆ' acute respiratory distress syndrome patients, and 8 healthy controls, and performed bulk RNA-sequencing of enriched neutrophils, plasma proteomics, cfDNA measurements and high throughput antibody profiling assays to investigate the relationship between neutrophil states and disease severity or death. We iden

Interactions of HIV and Antiretroviral Therapy With Neutrophils and Platelets.

Neutrophils are important components of the innate immune system that mediate pathogen defense by multiple processes including phagocytosis, release of proteolytic enzymes, production of reactive oxygen species, and neutrophil extracellular trap formation. Abnormalities of neutrophil count and function have been described in the setting of HIV infection, with the majority of antiretroviral agents (ARVs), excluding zidovudine, having been reported to correct neutropenia. Questions still remain, however, about their impact on neutrophil function, particularly the possibility of persistent neutro

The counter-intuitive role of the neutrophil in the acute respiratory distress syndrome.

INTRODUCTION: Neutrophils are the primary effectors of the innate immune system but are profoundly histotoxic cells. The acute respiratory distress syndrome (ARDS) is considered to be a prime example of neutrophil-mediated tissue injury. SOURCES OF DATA: The information presented in this review is acquired from the published neutrophil cell biology literature and the longstanding interest of the senior authors in ARDS pathogenesis and clinical management. AREAS OF AGREEMENT: Investigators in the field would agree that neutrophils accumulate in high abundance in the pulmonary microcirculation

Osteoprotegerin induces podosome disassembly in osteoclasts through calcium, ERK, and p38 MAPK signaling pathways.

Osteoclasts are critical for bone resorption and use podosomes to attach to bone matrix. Osteoprotegerin (OPG) is a negative regulator of osteoclast function that can affect the formation and function of podosomes. However, the signaling pathways that link OPG to podosome function have not been well characterized. Therefore, this study examined the roles of intracellular calcium and MAPKs in OPG-induced podosome disassembly in osteoclasts. We assessed the effects of the intracellular calcium chelator Bapta-AM, ERK inhibitor U0126, and p38 inhibitor SB202190 on OPG-treated osteoclast differenti

Isolation and characterization of stem cells from the human parathyroid gland.

OBJECTIVES: Somatic stem cells can be obtained from a variety of adult human tissues. However, it was not clear whether human parathyroid glands, which secrete parathyroid hormones and are essential in maintaining homeostasis levels of calcium ions in the circulation, contained stem cells. We aimed to investigate the possibility of isolating such parathyroid-derived stem cells (PDSC). MATERIALS AND METHODS: Surgically removed parathyroid glands were obtained with informed consent. Cell cytogenetics was used to observe chromosomal abnormalities. Surface phenotypes were characterized by flow cy

Serum MGP and ECP are commonly elevated in first-onset and recurrent ischemic stroke: a two-phase DIA proteomic study in a Chinese cold-region cohort.

BACKGROUND: Ischemic stroke (IS) has a high recurrence rate, yet effective biomarkers for distinguishing first-onset from recurrent IS remain limited. This study aimed to identify serum proteomic biomarkers commonly elevated in both first-onset and recurrent IS and to evaluate their ability to discriminate each disease state from healthy controls in a Chinese cold-region population. METHODS: The study was conducted in two phases. In the discovery phase, 10 patients with first-onset IS, 10 with recurrent IS, and 10 healthy controls were analyzed using Data-Independent Acquisition (DIA) proteom

Toward precision host-directed immunomodulation in community-acquired pneumonia: a proposed endotype-target-natural-product framework and a roadmap for validation in human lung organoids.

Community-acquired pneumonia (CAP) still causes high mortality despite effective antibiotics. Much of this death is driven by a dysregulated host immune response rather than by the pathogen itself, and antimicrobial resistance deepens the problem. Host-directed therapy has accordingly shifted toward precision immunomodulation, yet its benefit appears conditional on the inflammatory state. Stratification by a single inflammatory axis such as C-reactive protein (CRP) separates broad prognostic subgroups, but it does not indicate which effector pathway to target or which agent to use; we designat

Immune cell dynamics in myocardial infarction: mechanisms, therapeutic targets, and translational Perspectives.

BACKGROUND: Myocardial infarction (MI) triggers a complex and dynamic immune response involving both innate and adaptive immune cells. Among these, macrophages (Mφ), neutrophils, dendritic cells, and mast cells play essential roles in inflammation, tissue repair, and cardiac remodeling. Although inflammation is necessary for clearing necrotic tissue, dysregulated immune responses can exacerbate injury and lead to adverse cardiac outcomes. A comprehensive understanding of immune cell heterogeneity and functions in MI is critical for developing targeted therapeutic strategies. AIM OF REVIEW: Th

Peri-procedural Lidocaine Administration for Radiofrequency Ablation of Hepatocellular Carcinoma: A Pilot Study.

BACKGROUND AND AIMS: Radiofrequency ablation (RFA) is a curative option for early-stage hepatocellular carcinoma (HCC) when liver resection or transplantation is not a suitable option. This study evaluated whether a peri-procedural intravenous lidocaine infusion could reduce neutrophil extracellular trap formation (NETosis), neutrophil-to-lymphocyte ratio (NLR) and improve 1-year outcomes after RFA. METHODS: In this single-centre, randomized trial, 50 adult HCC patients undergoing RFA were allocated 1:1 to receive propofol-fentanyl sedation alone (PF group, n=25) or combined with intravenous

Structure-activity relationships and molecular mechanisms of natural products in sepsis-associated organ dysfunction: a narrative review.

BACKGROUND: Sepsis is defined as life-threatening organ dysfunction caused by a dysregulated host response to infection. Despite significant advancements in modern life-support technologies, the mortality rate of sepsis and its associated complications in the intensive care unit (ICU) remains unacceptably high. PROBLEM: The repeated clinical failure of traditional single-target therapies (e.g., anti-endotoxin agents) underscores an urgent need for systemic therapeutic strategies capable of simultaneously restoring the immune, metabolic, and coagulation networks. METHODS: This comprehensive n

Xuebijing injection in the treatment of COVID-19: An update on clinical studies, potentially active metabolites and mechanisms.

INTRODUCTION: Coronavirus disease 2019 (COVID-19) is an epidemic respiratory disease caused due to the infection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). In China, the National Health Commission of China announced that patients with COVID-19 who were treated with traditional Chinese medicines (TCMs) combined with antiviral drugs effectively alleviated their symptoms and recovered. Among these TCMs, Xuebijing (XBJ) injection plays an important role in the treatment of patients with COVID-19. However, this was a puzzle that what will be the clinical efficacy and safety of

Oral microbiome dysbiosis and innate immune dysregulation as determinants of oronasal fistula after primary cleft palate repair.

Oronasal fistula complicates 15-55% of primary cleft palate repairs, with recurrence rates approaching 43% after secondary closure, and global fistula rates have risen despite decades of iterative technical refinement, a trend that mechanical closure quality alone cannot explain. This narrative review synthesizes evidence from PubMed/MEDLINE, Scopus, and Web of Science from inception through April 2026 to argue that ONF is increasingly recognizable as a biologically mediated complication in which oral microbiome dysbiosis and innate immune dysregulation are primary, historically underrecognize

Seasonally dynamic ecosystems within offshore wind power monopile foundations support microbially influenced corrosion.

INRODUCTION: Microbially influenced corrosion (MIC) is a widespread problem in the maritime sector causingsevere economic damage. Foundation structures (monopiles) of offshore wind power systemsappear particularly prone to MIC. The hollow structures trap seawater and thus provide only limited exchange with the ambient waters. These special environmental conditions inside monopiles play a pivotal role for shaping the microbial community and respective biogeochemical processes. Currently, corrosion protection measures inside offshore monopiles only address electrochemical corrosion, likely resul

The "two-hit" storm: a hyper-inflammatory endotype in pediatric long COVID and its role in the severity of secondary bacterial pneumonia-a mechanistic review and clinical implications.

Following the COVID-19 pandemic, the clinical patterns of pediatric respiratory infections have undergone significant changes, with increasing attention on the immunological imprint left by Post-Acute Sequelae of SARS-CoV-2 infection (PASC), commonly known as Long COVID. A perplexing clinical phenomenon has been observed: some children with a history of Long COVID exhibit a disproportionately severe inflammatory response and extensive lung injury when encountering common community-acquired pneumonia, such as that caused by Mycoplasma pneumoniae or Streptococcus pneumoniae, inconsistent with th
