The clinical significance of copy number variants (CNVs) in congenital heart disease (CHD) continues to be a challenge. Although CNVs including genes can confer disease risk, relationships between gene dosage and phenotype are still being defined. Our goal was to perform a quantitative analysis of CNVs involving 100 well-defined CHD risk genes identified through previously published human association studies in subjects with anatomically defined cardiac malformations. A novel analytical approach permitting CNV gene frequency "spectra" to be computed over prespecified regions to determine pheno

Brugada syndrome is characterized clinically by the onset of syncopes or sudden death related to ventricular tachyarrhythmias in patients with a structurally normal heart. Its electrocardiographic features include right bundle branch bloc and ST-segment elevations in the precordial leads V1-V3. The estimated prevalence is 1 per 1000 in Asian countries and probably lower elsewhere: Asia is probably a birthplace of the syndrome. Its transmission is autosomal dominant with variable penetrance. Mutations have been identified in a gene coding for the alpha subunit of the sodium channel (SCN5A) in o

A nonlinear mixed-effect modeling (NONMEM) program was used to evaluate the effects of cytochrome P450 (CYP) 2C9 and CYP2C19 polymorphisms on the phenobarbital (PB) population clearance for Japanese epileptics. The pharmacokinetics of the 260 PB concentrations at a steady-state obtained from 79 patients was described with a one-compartment open pharmacokinetic model with first-order elimination. The covariates screened included the total body weight (BW), age, gender, PB daily dose, CYP2C9 and CYP2C19 genotypes, the coadministered antiepileptic drugs (AEDs), and complications. The final model

BACKGROUND: Duodenal and pancreatic neuroendocrine tumors are the leading cause of mortality in multiple endocrine neoplasia type 1, due to distant metastasis. Although surgery is the cornerstone of management, patients may require reintervention for disease progression or hormone-related morbidity. The clinical and genetic determinants of reintervention and long-term oncologic outcomes after index duodenal and pancreatic neuroendocrine tumor surgery in multiple endocrine neoplasia type 1 remain unclear.
METHODS: We retrospectively analyzed patients with genetically confirmed multiple endocri

BACKGROUND: Diabetic nephropathy (DN), a severe complication of diabetes, is influenced by genetic, immune, and gut microbial factors but lacks targeted therapeutic strategies. Druggable genes (DGGs) present a promising avenue, yet their causal prioritization in DN and their connections to gut microbiota-metabolite mechanisms remain underexplored.
METHODS: This study utilized differential expression analysis to identify differentially expressed genes (DEGs), Mendelian randomization (MR) to establish causal associations with DN, and machine learning algorithms to pinpoint key genes. Blood samp

INTRODUCTION: In women with a regular cycle, reliable last menstrual period (LMP), and accurate crown-rump length (CRL) estimation, discrepancies in estimated date of delivery (EDD) may indicate impaired fetal growth and increased risk of morbidity. This study examines whether discrepancies between EDD based on LMP (EDDLMP) and CRL (EDDCRL) are associated with chromosomal aberrations, adverse pregnancy outcomes, and obstetric complications.
MATERIAL AND METHODS: A Danish nationwide register-based cohort study including all singleton pregnancies with both CRL and LMP registered between 2008 an

Metabolic renal cell carcinomas (RCC) deficient in succinate dehydrogenase (SDH) or fumarate hydratase (FH) are rare but clinically significant entities formalized in the last WHO classification. Their recognition typically starts from morphology and is corroborated by targeted immunohistochemistry (IHC) and, where appropriate, molecular and germline testing. In routine practice, however, implementation may be uneven. Hence, we conducted a nationwide, web-based survey (made by 25 items) among members of the Italian Study Group of Uropathology (GIUP) to map real-world awareness, diagnostic path

In this study, optimization and biological activities of biologically active components obtained from Prunus armeniaca L. (apricot) fruit were investigated. Within the scope of the study, optimum extraction conditions were determined by using different extraction parameters (temperature, time and ethanol/water ratio) and the most suitable extracts were produced in terms of biological activity. Response Surface Method (RSM) and Artificial Neural Networks-GENetic Algorithms (ANN-GA) techniques were applied for extraction optimization. Antioxidant, antiproliferative and anticholinesterase activit…

The Philadelphia positive acute lymphoblastic leukemia (ALL BCR::ABL1 positive) is characterized by the presence of the t(9;22)(q34;q11) resulting in the formation of the BCR::ABL1 fusion, constant high cellular BCR-ABL1 tyrosine kinase activity, abnormal lymphoid cell proliferation and genetic instability of leukemic cells. Recently, great progress has been made due to the incorporation of tyrosine kinase inhibitors (TKI's) into the treatment algorithms. Despite this, a significant number of patients experienced therapy resistance mainly due to disease evolution and/or acquisition of TKI resi

BACKGROUND: The ataxia-telangiectasia mutated (ATM) kinase phosphorylates and activates several downstream targets that are essential for DNA damage repair, cell cycle inhibition and apoptosis. Germline biallelic inactivation of the ATM gene causes ataxia-telangiectasia (A-T), and heterozygous pathogenic variant (PV) carriers are at increased risk of cancer, notably breast cancer. This study aimed to investigate whether DNA methylation profiling can be useful as a biomarker to identify tumors arising in ATM PV carriers, which may help for the management and optimal tailoring of therapies of th

BACKGROUND: Ascending thoracic aortic dilation is a complex trait that involves modifiable and non-modifiable risk factors and can lead to thoracic aortic aneurysm and dissection. Clinical risk factors have been shown to predict ascending thoracic aortic diameter. Polygenic scores (PGS) are increasingly used to assess clinical risk for multifactorial diseases. The degree to which a PGS can improve aortic diameter prediction is not known. In this study we tested the extent to which the addition of a PGS to clinical prediction algorithms improves the prediction of aortic diameter.

BACKGROUND: Acute myocardial infarction (AMI) has two clinical characteristics: high missed diagnosis and dysfunction of leukocytes. Transcriptional RNA on leukocytes is closely related to the course evolution of AMI patients. We hypothesized that transcriptional RNA in leukocytes might provide potential diagnostic value for AMI. Integration machine learning (IML) was first used to explore AMI discrimination genes. The following clinical study was performed to validate the results.
METHODS: A total of four AMI microarrays (derived from the Gene Expression Omnibus) were included in bioanalysis

BACKGROUND: Congenital heart defects (CHDs) affect approximately half of individuals with Down syndrome (DS) but the molecular reasons for incomplete penetrance are unknown. Previous studies have largely focused on identifying genetic risk factors associated with CHDs in individuals with DS, but comprehensive studies of the contribution of epigenetic marks are lacking. We aimed to identify and characterize DNA methylation differences from newborn dried blood spots (NDBS) of DS individuals with major CHDs compared to DS individuals without CHDs.

Background: Growing evidence suggests the links between moyamoya disease (MMD) and autoimmune diseases. However, the molecular mechanism from genetic perspective remains unclear. This study aims to clarify the potential roles of autoimmune-related genes (ARGs) in the pathogenesis of MMD. Methods: Two transcription profiles (GSE157628 and GSE141025) of MMD were downloaded from GEO databases. ARGs were obtained from the Gene and Autoimmune Disease Association Database (GAAD) and DisGeNET databases. Differentially expressed ARGs (DEARGs) were identified using "limma" R packages.
