Association between ambient air pollution exposure and pubertal development: An ecological study.

BACKGROUND: The incidence rate of precocious puberty (PP) is rising, and ambient air pollutants may influence pubertal timing. This study explores the association between air pollution and PP and normal variants of puberty (NVP) in a Spanish pediatric population. METHODS: We conducted a retrospective ecological study in Asturias, Spain. Children ≤9 years diagnosed with PP or NVP between 2021 and 2024 were included. Data on pollutants (PM10, PM2.5, SO2, NO2, CO, O3, NO, NOx) from the 2 years prior to diagnosis were obtained from the nearest monitoring stations. The region was divided into 12 z

An update in the MRI features, classification, and associated brain abnormalities in hypothalamic hamartomas.

OBJECTIVES: Hypothalamic hamartomas (HHs) are rare, non-neoplastic malformations associated with gelastic seizures and, less frequently, precocius puberty. Recent classifications have linked HH subtypes with clinical features, and emerging evidence suggests that HH may co-occur with other brain abnormalities. This study investigates the imaging characteristics of HH, the prevalence of concurrent structural abnormalities, and metabolic alterations using magnetic resonance spectroscopy (MRS). METHODS: We retrospectively analyzed MRI and MRS findings in 18 patients with HH (mean age, 9.24 ± 8 ye

Pubertal delay in patients with paternally derived 15q11.2-q13 duplication syndrome and MKRN3 overexpression.

CONTEXT: Duplications of the 15q11.2-q13 region are primarily associated with neurodevelopmental disorders, while their endocrine implications remain poorly defined. The imprinted gene MKRN3, located within this locus, inhibits pubertal onset, and loss-of-function variants cause central precocious puberty. However, the clinical impact of increased MKRN3 dosage remains unclear. This study investigates whether paternally derived 15q11.2-q13 duplications lead to MKRN3 overexpression and are associated with delayed pubertal maturation. MATERIALS AND METHODS: Four patients with confirmed paternal

Internal Exposure to BTEX in Tropical Children: Does Exposure Speed Up Pubertal Development?

Benzene, toluene, ethylbenzene, and xylene (BTEX) are a common class of volatile organic compounds linked to adverse health outcomes. Although BTEX have been shown to have endocrine disrupting properties, their potential impacts on pubertal development in children remain unclear. In this study, for the first time, we investigated the possible association of BTEX exposure with precocious puberty (PP) and early puberty (EP) in children. We conducted a case-control study that included 246 children diagnosed with PP or EP and a controls category matched for sex and age. Urinary concentrations of s

Serological biomarker models composed of luteinizing hormone, kisspeptin, vitamin D and estradiol, and their clinical test value in girls.

OBJECTIVE: Central precocious puberty (CPP) may lead to premature pubertal onset. While the GnRH stimulation test remains the gold standard, its invasive nature and prolonged procedure limit clinical utility, particularly in pediatric populations. Emerging biomarkers, including luteinizing hormone, estradiol, kisspeptin, and vitamin D, have shown promise in distinguishing CPP from normal puberty. This study systematically evaluated the diagnostic utility of these biomarkers both individually and in novel combinatorial models, with the aim of establishing a non-invasive and more accessible diag

Pubertal, Auxological, and Adult Height Outcomes in Children Treated for Adrenocortical Tumors: Half a Century Experience.

INTRODUCTION: Most children with adrenocortical tumors (ACTs) present with accelerated growth and skeletal maturation at diagnosis, which potentially compromises their adult heights (AHs). Knowledge about growth and pubertal patterns after ACT resection is scarce. This study presents the pubertal and auxological development of patients treated for ACT and followed up at a single pediatric endocrinology service in Brazil. METHODS: Retrospective cohort study including 63 patients (47 girls) followed up between 1966 and 2021. Pubertal and auxological data from ACT diagnosis to AH were analyzed.

Severe Acquired Hypothyroidism and Van Wyk-Grumbach Syndrome in Two Children.

The primary manifestations of chronic hypothyroidism in children include growth arrest, delayed skeletal maturity, and delayed puberty. In 1960, Van Wyk and Grumbach reported three girls with hypothyroidism and a combination of incomplete isosexual precocious puberty (early breast development, menstruation, and absence of pubic hair), galactorrhea, delayed bone age, and pituitary enlargement. All abnormalities regressed after appropriate thyroid hormone replacement therapy. Over the years, an increasing number of reported cases has allowed for a more precise understanding of the clinical, bioc

Long-term effects of prenatal undernutrition on female rat hypothalamic KNDy neurons.

The nutritional environment during development periods induces metabolic programming, leading to metabolic disorders and detrimental influences on human reproductive health. This study aimed to determine the long-term adverse effect of intrauterine malnutrition on the reproductive center kisspeptin-neurokinin B-dynorphin A (KNDy) neurons in the hypothalamic arcuate nucleus (ARC) of female offspring. Twelve pregnant rats were divided into ad-lib-fed (control, n = 6) and 50% undernutrition (UN, n = 6) groups. The UN group was restricted to 50% daily food intake of the control dams from gestation

Comparison of the Clinical and Anthropometric Features of Treated and Untreated Girls with Borderline Early Puberty.

STUDY OBJECTIVE: Risks associated with precocious puberty might be observed in the rapidly progressive form of borderline early puberty (BEP). Differentiating the rate of progression is important for deciding treatment with gonadotropin-releasing hormone analogue (GnRHa). The aim was to examine the treatment characteristics and effect of treatment on predicted adult height (PAH). DESIGN: Retrospective observational study. SETTING: Single-center, a pediatric endocrinology unit. PARTICIPANTS: A total of 135 girls, pubertal findings starting between 7-10 years of age. INTERVENTIONS: Data were

Endocrine Long-Term Follow-Up of Children with Neurofibromatosis Type 1 and Optic Pathway Glioma
.

BACKGROUND/AIMS: Children with optic pathway glioma (OPG) face sequelae related to tumour location and treatment modalities. We aimed to assess the prevalence of hypothalamic-pituitary dysfunctions in children with neurofibromatosis type 1 (NF1) and OPG who did not receive radiotherapy or surgical resection. The causative role of tumour location on endocrinopathy development is investigated. METHODS: A retrospective follow-up study of 40 children with NF1 and OPG evaluated between August 1996 and May 2015 was undertaken. Patients who underwent radiotherapy or surgical resection were excluded

The longitudinal study of adrenal maturation during gonadal suppression: evidence that adrenarche is a gradual process.

The physical changes that herald the onset of puberty result from the combination of adrenarche and gonadarche. To examine adrenal maturation and associated changes in growth without the confounding effects of changes in the gonadal steroid milieu, we performed a longitudinal study in 14 young girls with idiopathic central precocious puberty during long-term pituitary-gonadal suppression. Beginning at the mean age of 2.9 yr, dehydroepiandrosterone sulfate levels, linear growth, skeletal maturation, body mass index, and secondary sexual development were evaluated at 3- to 6-month intervals for

ADHTransNet-based radiomics on multimodal pituitary MRI for non-invasive hormone prediction in children.

BACKGROUND AND OBJECTIVE: Growth hormone deficiency (GHD) and idiopathic central precocious puberty (ICPP) are typically diagnosed through invasive stimulation tests that require multiple blood samples collected over time. To reduce the need for such procedures, the study aims to establish an adjunctive tool by devising a fully automated pipeline for adenohypophysis segmentation and radiomics-based prediction of growth hormone (arg-pGH and ins-pGH in GHD) and gonadotropin (pLH and pLH/FSH in ICPP) levels in children. METHODS: A total of 274 subjects with 548 scans (T1-weighted and T2-weighted

Clinical spectrum and uncommon features of McCune-Albright syndrome in children: a cohort study from a National Referral Center.

INTRODUCTION: McCune-Albright syndrome (MAS) is a rare disease caused by somatic gain-of-function variants in the GNAS gene that lead to constitutive activation of the G protein alpha subunit (Gsα). Pathologic consequences can involve several tissues. Fibrous dysplasia (FD), café-au-lait skin macules and hyperfunctioning endocrinopathies are classic manifestations. However, the phenotypic spectrum of MAS is considerably wider and more complex. METHODS: We performed a pediatric retrospective study from our National Referral Center between 2007 and 2021 to describe the clinical spectrum of MAS

Adult height of children with congenital adrenal hyperplasia due to 21-hydroxylase deficiency.

BACKGROUND/PURPOSES: Congenital adrenal hyperplasia attributable to 21-hydroxylase deficiency (21-OHD) is a disorder of adrenal steroidogenesis. Achievement of optimal growth by such patients is challenging. We evaluated the adult height of Taiwanese children with 21-OHD and the effect of a gonadotropin-releasing hormone analogue (GnRHa) in patients with central precocious puberty (CPP) complicating 21-OHD. METHODS: Among 116 patients with 21-OHD in Taiwan, 90 who had attained adult height were subjected to an analysis of height outcomes. Nine with progressive CPP were treated with GnRHa and

Pelvic and breast ultrasound abnormalities and associated metabolic disturbances in girls with premature pubarche due to adrenarche.

OBJECTIVE: Premature adrenarche (PA) has been suggested as a risk factor for future health problems, such as metabolic syndrome and early menarche. However, not all girls with PA have these features and it is not certain who will develop them. We propose that these abnormalities might be identified earlier, even before they are visible. DESIGN: Case-control study. SETTING: Tertiary care hospital. PARTICIPANTS: Forty-eight girls with premature pubarche due to PA and age (mean age 7.6 ± 1.0 years), weight, body mass index (BMI), birth weight and gestational age-matched 49 girls with no palpab

The diagnosis of androgenetic alopecia in children: Considerations of pathophysiological plausibility.

Androgenetic alopecia (AGA), one of the most common causes of hair loss in men and women, is an infrequent cause of alopecia in children. In AGA, patients generally start noticing hair thinning after the onset of puberty due to progressive miniaturisation of the hair follicle which leads to vellus transformation of terminal hair. However, the occurrence of prepubertal AGA has rarely been reported in the literature. The pathophysiology of AGA is tightly linked to androgen hormones; prepubertal children do not usually produce significant amounts of adrenal or gonadal androgens. When it does occu

Early Experiences of Threat, but Not Deprivation, Are Associated With Accelerated Biological Aging in Children and Adolescents.

BACKGROUND: Recent conceptual models argue that early life adversity (ELA) accelerates development, which may contribute to poor mental and physical health outcomes. Evidence for accelerated development in youths comes from studies of telomere shortening or advanced pubertal development following circumscribed ELA experiences and neuroimaging studies of circuits involved in emotional processing. It is unclear whether all ELA is associated with accelerated development across global metrics of biological aging or whether this pattern emerges following specific adversity types. METHODS: In 247 c
