Assisted reproductive technology represents a possible risk factor for development of epimutation-mediated imprinting disorders for mothers aged ≥ 30 years.

BACKGROUNDS: The proportion of assisted reproductive technology (ART)-conceived livebirths of patients with imprinting disorders (IDs) is higher than that of the general population. Whether this is due to ART or confounding effects of advanced parental age was not investigated. We examined the association of ART and parental ages at childbirth for the development of eight epimutation-mediated imprinting disorders (epi-IDs). RESULTS: We enrolled 136 patients with epi-IDs and obtained general population ART data from the Japanese robust nationwide registry. We compared the proportion of ART-con

The DNA methylome of human sperm is distinct from blood with little evidence for tissue-consistent obesity associations.

Epidemiological research suggests that paternal obesity may increase the risk of fathering small for gestational age offspring. Studies in non-human mammals indicate that such associations could be mediated by DNA methylation changes in spermatozoa that influence offspring development in utero. Human obesity is associated with differential DNA methylation in peripheral blood. It is unclear, however, whether this differential DNA methylation is reflected in spermatozoa. We profiled genome-wide DNA methylation using the Illumina MethylationEPIC array in a cross-sectional study of matched human b

DNA Methylation and Regulatory Elements during Chicken Germline Stem Cell Differentiation.

The production of germ cells in vitro would open important new avenues for stem biology and human medicine, but the mechanisms of germ cell differentiation are not well understood. The chicken, as a great model for embryology and development, was used in this study to help us explore its regulatory mechanisms. In this study, we reported a comprehensive genome-wide DNA methylation landscape in chicken germ cells, and transcriptomic dynamics was also presented. By uncovering DNA methylation patterns on individual genes, some genes accurately modulated by DNA methylation were found to be associat

BAFopathies' DNA methylation epi-signatures demonstrate diagnostic utility and functional continuum of Coffin-Siris and Nicolaides-Baraitser syndromes.

Coffin-Siris and Nicolaides-Baraitser syndromes (CSS and NCBRS) are Mendelian disorders caused by mutations in subunits of the BAF chromatin remodeling complex. We report overlapping peripheral blood DNA methylation epi-signatures in individuals with various subtypes of CSS (ARID1B, SMARCB1, and SMARCA4) and NCBRS (SMARCA2). We demonstrate that the degree of similarity in the epi-signatures of some CSS subtypes and NCBRS can be greater than that within CSS, indicating a link in the functional basis of the two syndromes. We show that chromosome 6q25 microdeletion syndrome, harboring ARID1B dele

Cross-tissue integration of genetic and epigenetic data offers insight into autism spectrum disorder.

Integration of emerging epigenetic information with autism spectrum disorder (ASD) genetic results may elucidate functional insights not possible via either type of information in isolation. Here we use the genotype and DNA methylation (DNAm) data from cord blood and peripheral blood to identify SNPs associated with DNA methylation (meQTL lists). Additionally, we use publicly available fetal brain and lung meQTL lists to assess enrichment of ASD GWAS results for tissue-specific meQTLs. ASD-associated SNPs are enriched for fetal brain (OR = 3.55; P < 0.001) and peripheral blood meQTLs (OR = 1.5

Long-lived epigenetic interactions between perinatal PBDE exposure and Mecp2308 mutation.

The widespread use of persistent organic polybrominated diphenyl ethers (PBDEs) as commercial flame retardants has raised concern about potential long-lived effects on human health. Epigenetic mechanisms, such as DNA methylation, are responsive to environmental influences and have long-lasting consequences. Autism spectrum disorders (ASDs) have complex neurodevelopmental origins whereby both genetic and environmental factors are implicated. Rett syndrome is an X-linked ASD caused by mutations in the epigenetic factor methyl-CpG binding protein 2 (MECP2). In this study, an Mecp2 truncation muta

Drugs and addiction: an introduction to epigenetics.

Addiction is a debilitating psychiatric disorder, with a complex aetiology involving the interaction of inherited predispositions and environmental factors. Emerging evidence suggests that epigenetic alterations to the genome, including DNA methylation and histone modifications, are important mechanisms underlying addiction and the neurobiological response to addictive substances. In this review, we introduce the reader to epigenetic mechanisms and describe a potential role for dynamic epigenetic changes in mediating addictive behaviours via long-lasting changes in gene expression. We summariz

Systemic elevation of gut derived p-cresol during one year in space: implications for brain, heart, bone, liver and microbiome in astronauts.

INTRODUCTION: We observed the elevation of p-cresol in the untargeted metabolome of the multiomics portion of the NASA Twins Study of One Year in Space. p-Cresol is a uremic and neurotoxin that is produced via the metabolism of dietary tyrosine by gut bacteria. Intestinally produced p-cresol is absorbed into systemic circulation where it exerts its adverse metabolic effects and drives phenotypic changes relevant to astronauts, including the brain, heart, bone, liver, kidney, and colon. METHODS: A pair of monozygotic twins were studied for 25 months, during which one subject spent 340 days abo

Associations between per- and polyfluoroalkyl substances (PFAS), DNA methylation and gene expression from background exposed Norwegian women (2003-2006).

Per- and polyfluoroalkyl substances (PFAS) are a large group of widespread synthetic substances associated with several adverse health effects. A potential mechanism underlying these effects is epigenetic modifications, such as DNA methylation. However, evidence on associations between PFAS exposure and DNA methylation among adults is limited, and few studies have investigated the potential influence of PFAS-related methylation changes on gene expression. In this study, we investigated associations between PFAS exposure and DNA methylation in blood. Furthermore, the associations between these

EZH2 Inhibition Remodels Cell States and Enhances EGFRi Sensitization in Bladder Cancer.

BACKGROUND: Bladder cancer (BCa) displays interpatient heterogeneity and epigenetic plasticity that may limit treatment efficacy. OBJECTIVE: To investigate whether inhibition of Enhancer of zeste homolog 2 (EZH2) remodels tumor cell states and creates therapeutic vulnerabilities in BCa. DESIGN, SETTING, AND PARTICIPANTS: We integrated single-nucleus ribonucleic acid sequencing (snRNA-seq) of formalin-fixed paraffin-embedded (FFPE) primary tumors with single-cell ribonucleic acid sequencing (scRNA-seq) of matched patient-derived organoids (PDOs) from four BCas spanning stages Ta-T3. Placental Methylation Profile in Women Exposed to Fluoride and Its Association with Kidney Volume at Birth.

KEY POINTS: Placental DNA methylation patterns were associated with neonatal kidney volume in a population with high fluoride exposure. Newborns with high fluoride exposure and low kidney volume showed methylation changes in genes involved in kidney development pathways. These placental epigenetic patterns revealed early markers of altered kidney development, potentially linked to reduced nephron endowment. BACKGROUND: Mexico has a high burden of CKD. The state of Aguascalientes is among the regions with the highest reported prevalence of ESKD worldwide. In this setting, CKD of unknown etiolo

Delineating novel diagnostic biomarkers and therapeutic targets for oral submucosal fibrosis: an integrative multi-omics and machine learning approach.

BACKGROUND: Oral submucosal fibrosis (OSF) is a chronic and progressive disorder, caused by chewing areca nuts, affecting the oral cavity and oropharynx. OSF is characterized by severe symptoms like severe burning sensation, restricted mouth opening, etc. Given the multifactorial and poorly understood molecular basis of the disease, there is a need for novel biomarkers and therapeutic targets. METHOD: We downloaded 3 RNA-seq, two microarray, one epigenomic, and one single-cell RNA-seq datasets from the gene expression omnibus database. Differentially expressed genes (DEGs) were characterized

The adenovirus oncoprotein E1B-55K reshapes epigenetic histone modifications in primary human cells.

Oncogenic viruses can induce epigenetic changes in host cells, which may contribute to cell transformation and tumorigenesis by altering gene expression patterns. Human adenoviruses, while primarily known for their lytic infections, have also been implicated in oncogenic transformation in experimental settings, but the mechanisms underlying adenovirus oncoprotein-induced epigenetic dysregulation remain poorly defined. Building on our recent findings that the adenoviral oncoprotein E1B-55K coordinates cellular transformation through interactions with DNA-bound host transcription factors, we nex

Breast Milk Bioactive Components and Early-Life Epigenetic Programming: Implications for Nutrigenomics and Population Health.

Multifactorial diseases cannot be fully explained by genetic variation alone, highlighting the importance of epigenetic mechanisms that integrate early-life environmental exposures with long-term health outcomes. Within the Developmental Origins of Health and Disease (DOHaD) framework, human breast milk has emerged as a critical regulator of early-life epigenetic programming. Breast milk-derived bioactive components, including non-coding RNAs, extracellular vesicles, immune factors, hormones, and human milk oligosaccharides, actively modulate DNA methylation, chromatin structure, and gene expr

Changes in methylation associated with development of metabolic syndrome in testicular cancer patients treated with cisplatin chemotherapy.

Patients with metastatic testicular cancer (TC) treated with cisplatin-based chemotherapy (CBCT) are prone to develop metabolic syndrome (MetS). In an epigenome wide association study in patients with TC we have shown that DNA methylation was associated to CBCT. The aim of the study was to investigate whether there was a change in DNA methylation after treatment with CBCT, and if methylation status is related to the presence or development of MetS. In a prospective cohort of 67 TC patients who received CBCT, we assessed whole blood global methylation long interspersed nuclear element-1 (LINE-1

Associations of Biological Aging With Perceived Stress in Patients With Head and Neck Cancer Undergoing Intensity Modulated Radiation Therapy: A Longitudinal Study.

PURPOSE: Accelerated biological aging increases morbidity and mortality in patients with head and neck cancer (HNC) undergoing intensity modulated radiation therapy (IMRT). Stress from HNC and IMRT may exacerbate this process, yet the link between biological aging and perceived stress during treatment remains unclear. The purpose of this study was to examine associations between biological aging and perceived stress in HNC patients receiving IMRT across 4 time points: pre-IMRT, the end of IMRT, and 3 and 12 months post-IMRT. METHODS AND MATERIALS: This prospective longitudinal study assessed

Genome-wide DNA methylation risk scores for schizophrenia derived from blood and brain tissues further explain the genetic risk in patients stratified by polygenic risk scores for schizophrenia and bipolar disorder.

BACKGROUND: Genetic and environmental factors contribute to the pathogenesis of schizophrenia (SZ) and bipolar disorder (BD). Among genetic risk groups stratified by combinations of Polygenic Risk Score (PRS) deciles for SZ, BD and SZ versus BD, genetic SZ risk groups had high SZ risk and prominent cognitive impairments. Furthermore, epigenetic alterations are implicated in these disorders. However, it was unclear whether DNA Methylation Risk Scores (MRSs) for SZ risk derived from blood and brain tissues were associated with SZ risk, particularly the PRS-stratified genetic SZ risk group.
