Distribution and estimation of anaerobic zoosporic fungi along the digestive tracts of sheep.

The status of anaerobic zoosporic (Chytridiomycota) fungi along the entire digestive tract of sheep was assessed both analytically and microscopically. Digest samples were taken from different segments of the digestive tracts of three newly killed sheep that previously had been used in experimental dietary studies. These digest samples were tested for the presence of rumen fungi by assessing the recovery of live fungi from the samples, direct observation of digested plant remains under the scanning electron microscope (SEM), and using a chitin assay as an estimation of fungal biomass. Live ana

Molecular cloning and identification of cDNA recombinants coding for bovine prochymosin in E. coli.

Poly(A) RNA was isolated from the gastric mucosa of the bovine fourth stomach (the abomasum) using and analysing several calves not older than 12 days. The amount of the preprochymosin mRNA in the mucosa of those animals at best reaches about 5-10% of the poly(A) RNA as estimated by in vitro translation and immunoprecipitation. Starting from that material double-stranded complementary DNA was synthesized, inserted by dG dC tailing into the PstI site of the vector plasmid pBR322 and used for transformation of E. coli. Tetracycline resistant clones containing DNA sequences coding for the full le

Carbohydrase activities in the bovine digestive tract.

1. The carbohydrase activities of homogenates of mucosa from the abomasum, small intestine, caecum and colon, and of the pancreas of cattle were studied. 2. The disaccharidase activities were located mainly in the small intestine and showed a non-uniform pattern of distribution along the small intestine; trehalase activity was highest in the proximal part, lactase and cellobiase activities were highest in the proximal and middle parts and maltase activity was highest in the distal part. 3. The intestinal lactase and cellobiase activities were highest in the young calf and decreased with age, w

Associations between days in the close-up group and milk production, transition cow diseases, reproductive performance, culling, and behavior around calving of Holstein dairy cows.

The objective of experiment I was to evaluate the association between days in the close-up group (DINCU) and milk production, early lactation diseases, reproductive performance, and culling. In experiment II behavioral changes associated with DINCU were evaluated using a neck-mounted sensor (Smarttag neck, Nedap Livestock Management, Groenlo, the Netherlands). Cow-lactations of 28,813 animals from 14,155 individual cows of 2 farms in northern Germany and western Slovakia, calving between January 2015 and December 2020, were included in the study. After exclusion of cows with a gestation length

Routine serum biomarkers and lightgbm modeling for the diagnosis of abomasal ulcers in culled dairy cows.

Abomasal ulcer (AU) is a common condition in dairy cows that significantly affects productivity and longevity. This study evaluated the diagnostic value of serum biomarkers and the fecal occult blood test (FOBT), and tested a Light Gradient Boosting Machine (LightGBM) model to predict ulcer occurrence. Postmortem analyses were performed on 304 culled dairy cows, from which blood and fecal samples were collected. Animals were classified according to the presence (n = 249) or absence (n = 55) of abomasal ulcers. No individual biomarker, including gastrin or pepsinogen, demonstrated consistent di

Anti-inflammatory but not insulin-sensitizing: Effects of docosahexaenoic acid abomasal infusions on adipose tissue in mid-lactation dairy cows.

Supplementing dairy cow diets with n-3 fatty acids has been an alternative nutritional intervention to reduce systemic inflammation and improve reproductive function. Docosahexaenoic acid (C22:6n-3; DHA) is one of the n-3 fatty acids with the strongest anti-inflammatory effects in humans and animals. However, it is unknown if DHA modulates adipose tissue (AT) function and inflammatory responses in dairy cows. Our objective was to evaluate the effects of DHA on AT insulin sensitivity and AT macrophage infiltration in mid-lactation dairy cows. Eight ruminally cannulated multiparous Holstein cows

Time-dependent RNA transcriptional profiling of abomasal mucosa in cattle infected with Ostertagia ostertagi.

In this study, we infected Holstein calves with Ostertagia ostertagi stage 3 larvae (L3) and determined gene expression profiles of abomasal fundic and pyloric mucosa by RNA sequencing (RNA-seq) at 3-5, 7-9, 10, and 21 days post-infection (dpi), which represent late L3 and early L4 (between 3-5 dpi), mid to late L4 (between 7-9 and 10 dpi) and adult stages (21 dpi) of the parasitic stage. Bioinformatics analyses were performed to profile the transcriptomic changes over time as well as between cattle abomasal tissues. The results will help understand the gastric responses of the host, especiall

Prognostic value of preoperative plasma l-lactate concentrations in calves with acute abdominal emergencies.

Acute abdominal emergencies in calves due to abomasal disorders, gastrointestinal ileus, or peritonitis are characterized by a rapid disease progression and usually require immediate surgical intervention. Those conditions are associated with a guarded prognosis, and the aim of the present study was to assess the prognostic relevance of preoperatively measured plasma l-lactate concentrations (l-LAC) in a large study population of calves with a broad spectrum of acute abdominal emergencies. For the purpose of this study, the medical records of 587 calves admitted to a veterinary teaching hospit

Concentration of Potassium in Plasma, Erythrocytes, and Muscle Tissue in Cows with Decreased Feed Intake and Gastrointestinal Ileus.

BACKGROUND: Healthy cows consume large amounts of potassium and a sudden loss in appetite can lead to hypokalemia. The routine method to evaluate potassium homeostasis is the measurement of the extracellular potassium in plasma or serum, but this does not provide information about the intracellular potassium pool. HYPOTHESIS/OBJECTIVES: To evaluate potassium homeostasis by comparing the extracellular and intracellular potassium concentration in cows with reduced feed intake and gastrointestinal ileus. ANIMALS: Twenty cows 1-3 days postpartum (group 1) and 20 cows with gastrointestinal ileus

Is there a cost of parasites to caribou?

Macroparasites potentially play a significant but often ignored role in the ecology and dynamics of wild ruminant populations. In the Arctic, parasites may impact on host populations by exacerbating the effects of seasonal and limited forage availability on the condition, fecundity and survival of individuals. We studied the effects of abomasal nematode parasites and warble flies, Hypoderma tarandi, on condition and pregnancy of caribou Rangifer tarandus in the Dolphin-Union herd, Nunavut, Canada. By the end of winter, female caribou over 2 years old showed a significant decrease in body weigh

Comparison of 0.9, 3.6, and 7.2% NaCl for correction of experimentally induced hypochloremic, hypokalemic metabolic alkalosis in sheep.

Nine adult female sheep were each surgically fitted with an Ivan and Johnston reentrant cannula in the cranial part of the duodenum just distal to the pylorus. By diversion (loss) of abomasal outflow, this model has been shown to consistently induce hypochloremic, hypokalemic metabolic alkalosis, accompanied by hyponatremia and dehydration. Each sheep was subjected to 3 treatment trials, each preceded by a 24-hour prediversion period, and a diversion period during which a syndrome of hypochloremia (68 +/- 2 mEq/L), hypokalemia, hyponatremia, and metabolic alkalosis was induced. Chloride deficiency in Holstein calves from a low chloride diet and removal of abomasal contents.

Chloride deficiency signs were produced in young Holstein calves by a low chloride diet (.063% chlorine) and daily removal of chloride in abomasal contents. General clinical signs included anorexia, weights loss, lethargy, mild polydipsia, and mild polyuria. In latter stages of the deficiency, severe eye defects (scleral injection, sunken eyes, scaliness around eyes) and reduced respiration rate became evident. Feces contained varying amounts of blood and mucus. The chloride imbalance resulted in severe alkalosis and hypochloremia leading to secondary hypokalemia, hyponatremia, and azotemia. A

Glucose metabolism and the somatotropic axis in dairy cows after abomasal infusion of essential fatty acids together with conjugated linoleic acid during late gestation and early lactation.

Sufficient glucose availability is crucial for exploiting the genetic potential of milk production during early lactation, and endocrine changes are mainly related to repartitioning of nutrient supplies toward the mammary gland. Long-chain fatty acids, such as essential fatty acids (EFA) and conjugated linoleic acid (CLA), have the potential to improve negative energy balance and modify endocrine changes. In the present study, the hypothesis that combined CLA and EFA treatment supports glucose metabolism around the time of calving and stimulates insulin action and the somatotropic axis in cows

Embryonal carcinoma in the abdominal cavity of a male calf.

An embryonal carcinoma was diagnosed in the abdominal cavity of a 55-day-old male calf. Macroscopically, a large volume of ascitic fluid was observed along with white to yellowish-white masses of various sizes densely located on the abdominal wall and the surface of abdominal organs. There was an absence of testes in the scrotum. Histologically, the tumor cells were polygonal, and the tumor was mostly composed of sheets of densely packed solid patterns with occasional papillary and tubular structures. Cell nuclei were variable in size, and cellular mitotic rate was high. Immunohistochemically,
