Genotyping of Acinetobacter baumannii clinical isolates from Polish hospitals uncovers novel sequence types.

Acinetobacter baumannii is an opportunistic pathogen associated with healthcare-acquired infections. This study examined the clonal relatedness and drug resistance of 38 clinical isolates of A. baumannii from three hospitals in Poland and five isolates from pig manure. PFGE analysis identified seven clusters (A-G); of these, clusters A and B comprised the majority of clinical isolates, while A and C were dominated by the pig manure isolates, and cluster C included the reference strain ATCC 17978. MLST analysis of the clinical isolates revealed nine sequence types (STs), including three previou

α-Pinene: Inhibitor of Acinetobacter baumannii biofilms and potential therapeutic agent for pneumonia.

Acinetobacter baumannii is a Gram-negative bacterium whose biofilm formation and mechanisms contribute to its persistent infectivity and drug resistance in clinical settings. Inhibition or disruption of biofilms might hold the key to resolving the issue of drug resistance in A. baumannii. α-Pinene, a bicyclic terpene olefin derived from the essential oils of plants, exhibits multiple biological activities, including antimicrobial, antioxidant, and anti-inflammatory effects. In this investigation, we discovered that α-Pinene had powerful antimicrobial activity against A. baumannii 390015, and i

The Multifaceted Landscape of Healthcare-Associated Infections Caused by Carbapenem-Resistant Acinetobacter baumannii.

Carbapenem-resistant Acinetobacter baumannii (CRAB) is an emerging and important major cause of nosocomial infections, posing a significant challenge to clinicians worldwide. The intrinsic and acquired resistance mechanisms exhibited by CRAB, associated with its ability to persist in healthcare environments, have transformed it into a critical public health concern. The clinical implications of CRAB infections include severe manifestations, like ventilator-associated pneumonia and bloodstream infections. These infections are often associated with increased morbidity and mortality, particularly

Induction of pneumonia in multidrug-resistant Acinetobacter baumannii infected immunocompetent BALB/c mice.

Assessing the efficacy and safety of potential therapeutics for multidrug-resistant (MDR) Acinetobacter baumannii infections necessitates the use of in vivo models, typically involving mice and highly virulent isolates of the bacterium. In this study, we investigated the clinical isolate Ab35 of MDR Acinetobacter baumannii to determine its ability to infect and induce pneumonia in a mouse infection model. Immunocompetent BALB/c mice were infected through the oropharyngeal aspiration route. Enlarged spleen germinal center, reduced lung air space, and infiltration of immune cells within the lung

Minocycline-based therapy for carbapenem-resistant Acinetobacter baumannii pneumonia: A multicenter retrospective study.

BACKGROUND: Minocycline is one of the few antibiotics that remain highly effective against carbapenem-resistant Acinetobacter baumannii (CRAB). However, the efficacy of minocycline for treating CRAB pneumonia has not been clearly demonstrated yet. METHODS: This multicenter retrospective cohort study evaluated adults treated for CRAB pneumonia at three tertiary care hospitals in South Korea from 2017 to 2022. Patients who received antibiotic regimens including minocycline were compared with those who received regimens not containing minocycline. The primary outcome measure was a 14-day clinica

Synergistic mechanism of Phyllanthus emblica extract and tetracycline against multidrug-resistant Acinetobacter baumannii.

The rising incidence of multidrug-resistant (MDR) Acinetobacter baumannii infections underscores the urgent need for novel antimicrobial strategies. The aim of this study was to investigate the synergistic effects between a polyphenol-rich extract from Phyllanthus emblica fruit and tetracycline against MDR A. baumannii strains. The extraction process was optimized using the Box-Behnken design approach to maximize the total phenolic content (TPC) of the P. emblica extract. Key variables, including ethanol concentration, extraction time, and solid-to-liquid ratio (w/v), were determined through s

Multidrug-resistant Acinetobacter baumannii meningitis and cerebellar abscess: case report and therapeutic considerations.

INTRODUCTION: Brain abscess is defined as a suppurative collection resulting from hematogenous dissemination as an extension from otorhinolaryngologic infectious foci, or secondary to cranial trauma and neurosurgical procedures. Its evolution follows four histopathological stages, the most severe complication being intraventricular rupture, which is associated with extremely high mortality. Central nervous system infections caused by multidrug-resistant (MDR/XDR) Acinetobacter baumannii are rare but severe, significantly limiting therapeutic options due to the reduced penetration of the blood-

A novel Saclayvirus Acinetobacter baumannii phage genomic analysis and effectiveness in preventing pneumonia.

Acinetobacter baumannii, which is resistant to multiple drugs, is an opportunistic pathogen responsible for severe nosocomial infections. With no antibiotics available, phages have obtained clinical attention. However, since immunocompromised patients are often susceptible to infection, the appropriate timing of administration is particularly important. During this research, we obtained a lytic phage vB_AbaM_P1 that specifically targets A. baumannii. We then assessed its potential as a prophylactic treatment for lung infections caused by clinical strains. The virus experiences a period of inac

Activity of cefiderocol in combination with tetracycline analogues against carbapenem-resistant Acinetobacter baumannii.

Therapeutic options for carbapenem-resistant Acinetobacter baumannii (CA-AB) are quite limited. Cefiderocol, a novel siderophore cephalosporin, has shown potent in vitro activity against CR-AB, and new tetracycline analogues such as eravacycline and omadacycline have been available in recent years. However, the synergism of cefiderocol with tetracycline analogues against CR-AB has not been well investigated. In this study, we evaluated the in vitro synergistic activity of cefiderocol in combination with tetracycline analogues (minocycline, tigecycline, eravacycline and omadacycline) against 48

Dietary zinc deficiency promotes Acinetobacter baumannii lung infection via IL-13 in mice.

Dietary zinc deficiency is a major risk factor for pneumonia. Acinetobacter baumannii is a leading cause of ventilator-associated pneumonia and a critical public health threat due to increasing rates of multidrug resistance. Patient populations at increased risk for A. baumannii pneumonia are also at increased risk of zinc deficiency. Here we established a mouse model of dietary zinc deficiency and acute A. baumannii pneumonia to test the hypothesis that host zinc deficiency contributes to A. baumannii pathogenesis. We showed that zinc-deficient mice have significantly increased A. baumannii b

Sub-MIC antibiotics increased the fitness cost of CRISPR-Cas in Acinetobacter baumannii.

INTRODUCTION: The escalating prevalence of bacterial resistance, particularly multidrug-resistant bacteria like Acinetobacter baumannii, has become a significant global public health concern. The CRISPR-Cas system, a crucial defense mechanism in bacteria against foreign genetic elements, provides a competitive advantage. Type I-Fb and Type I-Fa are two subtypes of CRISPR-Cas systems that were found in A. baumannii, and the I-Fb CRISPR-Cas system regulates antibiotic resistance in A. baumannii. However, it is noteworthy that a majority of clinical isolates of A. baumannii lack or have incomplet

Replicative Acinetobacter baumannii strains interfere with phagosomal maturation by modulating the vacuolar pH.

Bacterial pneumonia is a common infection of the lower respiratory tract that can afflict patients of all ages. Multidrug-resistant strains of Acinetobacter baumannii are increasingly responsible for causing nosocomial pneumonias, thus posing an urgent threat. Alveolar macrophages play a critical role in overcoming respiratory infections caused by this pathogen. Recently, we and others have shown that new clinical isolates of A. baumannii , but not the common lab strain ATCC 19606 (19606), can persist and replicate in macrophages within spacious vacuoles that we called A cinetobacter C ontaini

The effect of phage-antibiotic combination strategy on multidrug-resistant Acinetobacter baumannii biofilms.

Acinetobacter baumannii (A. baumannii) is considered a critical human pathogen due to multi-drug resistance and increased infections. As a result of the resistance of A. baumannii biofilms to antimicrobial agents, it is necessary to develop new biofilm control strategies. In the present study, we evaluated the efficacy of two previously isolated bacteriophage C2 phage, K3 phage and phage cocktail (C2 + K3 phage) as a therapeutic agent in combination with antibiotic (colistin) against biofilm of multidrug-resistant A. baumannii strains (n = 24). The effects of phage and antibiotics on mature bi
