Acyl-CoA dehydrogenase activity in the riboflavin-deficient rat. Effects of starvation.

Riboflavin deficiency in weanling rats causes a metabolic disorder characterized by failure to oxidize fatty acids. The disorder is similar to that seen in several human diseases, some of which are responsive to pharmacological doses of riboflavin. Previous analysis of the riboflavin-deficient rat has shown that the failure of fatty acid oxidation is due to a decrease in the activity of the acyl-CoA dehydrogenases of beta-oxidation. The activity of these flavoenzymes in liver rapidly decreases when a riboflavin-deficient diet is initiated. The objectives of these experiments were to analyse th

Acylation-based post-modification synthesis of an efficient fluorophilic magnetic microporous organic network for improved extraction of fluorine-containing drugs.

A novel pentafluorophenyl-functionalized fluorophilic magnetic microporous organic network (MMON-PFB) was synthesized via an acylation-based post-modification strategy for highly improved magnetic solid-phase extraction (MSPE) of trace fluorine-containing drugs (FCDs) in animal-derived foods and water samples. After modifying MMON-OH with PFB, MMON-PFB simultaneously possesses F-F, hydrogen bonding, π-π staking, and hydrophobic interaction sites, and the MSPE recovery largely increased from 45.2% to 99.4%, enabling efficient and selective enrichment of FCDs. A sensitive MMON-PFB-MSPE-HPLC-UV m…

Mechanistic and Kinetic Insights into the Acylation Reaction of Hepatitis C Virus NS3/NS4A Serine Protease with NS4B/5A Substrate.

Reaction mechanisms and rate constants of the acylation reaction of the hepatitis C virus (HCV) NS3/NS4A serine protease with the NS4B/5A natural substrate were studied using SCC-DFTB/MM (self-consistent charge density functional tight binding/molecular mechanics) and EA-VTST/MT (ensemble-averaged variational transition state theory/multidimensional tunneling) methods, considering the isotope effect (H/D). This reaction is crucial in the HCV life cycle. The reaction follows an essentially concerted mechanism. Although two elementary steps are involved, no intermediate step has been found betwe…

Nuanced differences in adenylate cyclase toxin production, acylation, and secretion may contribute to the evolution of virulence in Bordetella species.

Bordetella pertussis, which causes the acute human disease whooping cough, evolved from Bordetella bronchiseptica, which causes long-term, chronic infections in a broad range of mammals. Both B. pertussis and B. bronchiseptica produce adenylate cyclase toxin (ACT), a bifunctional protein with adenylate cyclase-mediated cell intoxication and pore-forming activities. CyaC-mediated acylation of ACT is important for cell intoxication and required for pore-forming activity in vitro, but its role in vivo was unknown. Our comparative analysis showed that although ACT secreted by B. pertussis is fully…

Copper(II)-catalyzed C3-acylation of quinoxalin-2(1H)-ones with methyl ketones via the formal decarbonylation of 1,2-diketones.

The inexpensive copper(II) bromide-catalyzed cross-dehydrogenative coupling/Kornblum oxidation/formal decarbonylation of N-substituted/unsubstituted quinoxalinones in DMSO is reported. It has offered a new technique for the cleavage of C(CO)-C(CO) bonds under base- and ligand-free conditions, allowing for the direct synthesis of a diverse range of C3-acylated quinoxalinones. This persulfate-peroxide-free catalytic method provides good yields, a broad substrate scope, and good tolerance to various functionalities.

Comprehensive S-acylation profiling of the porcine epididymis and exosomes reveals a role in cargo sorting and long-distance trafficking.

The epididymis orchestrates sperm maturation through microenvironmental regulation and epididymosome-mediated cargo delivery. Despite emerging evidence implicating protein S-acylation in vesicular trafficking, its compartment-specific dynamics and functional implications in epididymal physiology remain poorly characterized. Here, we employed acyl-biotin exchange-based 4D proteomics to decode the S-acylation proteomic of porcine caput/cauda epididymidis and their exosomes. Comparative analysis identified 2780 and 2084 S-acylated proteins in caput and cauda tissues, respectively, with 317 upregu…

We further show that the monomeric preparation displayed hemolytic and cytotoxic activities suggesting that the monomer is the genuine, physiologically active form of the toxin. Photo-Catalyzed Acyl Azolium Promoted Selective α-C(sp3 )-H Acylation of Acetone via HAT: Access to Thermodynamically Less Favoured (Z)-α,β-Unsaturated Ketones.

Mono α-acylation of acetone has been achieved for the first time by reacting with bench-stable acyl azolium salts under violet-LED light at room temperature. The intermolecular hydrogen atom transfer (HAT) from acetone to triplet state of azolium salts under violet LED irradiation resulted in thermodynamically less favourable (Z)-α,β-unsaturated ketones with up to 99 : 1 selectivity via C-C bond formation. This compelling protocol access the desired α-C(sp3 )-H acylation product under metal-, ligand- and oxidant-free conditions on a wide range of substrates.

Studies of intramolecular rearrangements of acyl-linked glucuronides using salicylic acid, flufenamic acid, and (S)- and (R)-benoxaprofen and confirmation of isomerization in acyl-linked delta 9-11-carboxytetrahydrocannabinol glucuronide.

NMR and HPLC have been used to investigate the rearrangements of 1-O-acylglucuronides in vitro and the occurrence of rearranged isomers in urine. Glucuronides of flufenamic acid, (S)- and (R)-benoxaprofen, salicylic acid, and delta 9-11-carboxytetrahydrocannabinol were synthesized, by use of immobilized enzymes, or purified from urine. Ester-linked isomers of these gluruconides were characterized, and isomers derived from flufenamic acid, (S)-benoxaprofen, and salicylic acid were purified for further study by NMR and HPLC. The positions of the new ester linkages could be identified by two-dime

Design, synthesis, and antimicrobial evaluation of heteroaromatic or acyl-substituted benzenesulfonamide-linked aminoquinolinequinones.

A series of aminated quinolonequinones (AQQ1-13) was synthesized from quinolinequinone and substituted aromatic amines containing a sulfonamide group with conformationally restricted heterocyclic structures or an acyl group. The obtained AQQs were characterized by various spectroscopic techniques including FTIR, NMR, and MS analyses. Structural modifications of the sulfonamide group in these compounds were selected by introducing various heterocyclic substituents, such as isoxazole, thiadiazole, pyridine, pyrimidine, pyrazine, and pyridazine, or by attaching acyl groups (acetyl or benzamide) w

Zn Complexes with the 4-Acyl-Pyrazol-5-One Basis and Their Thin Films.

Three zinc(II) complexes based on 1-phenyl-3-methyl-4-acyl-5-pyrazolone were synthesized and characterized by elemental analysis, by X-ray crystallography and spectroscopy (UV-vis, fluorescence and IR). Crystallographic studies reveal that the complexes have a mononuclear structure in the solid state; however, intermolecular interactions combine the complexes into a 1D polymer chain. The complexes exhibit weak luminescence in the polycrystalline state and moderate emission in solutions and thin amorphous films. Thin films of the studied compounds were deposited on a glass substrate using therm

Pseudomonas aeruginosa acyl-CoA dehydrogenases and structure-guided inversion of their substrate specificity.

Fatty acids are a primary source of carbon for Pseudomonas aeruginosa (PA) in the airways of people with cystic fibrosis (CF). Here, we use tandem mass-tag proteomics to analyse the protein expression profile of a CF clinical isolate grown on different fatty acids. Two fatty acyl-CoA dehydrogenases (designated FadE1 and FadE2) are strongly induced during growth on fatty acids. FadE1 displays a strong preference for long-chain acyl-CoAs, whereas FadE2 exclusively utilizes medium-chain acyl-CoAs. Structural analysis of the enzymes enables us to identify residues comprising the substrate selectiv

Peptide Backbone Editing via Post-Translational O to C Acyl Shift.

Despite tremendous efforts to engineer translational machinery, replacing the encoded peptide backbone with new-to-nature structures remains a significant challenge. C, H, O, and N are the elements of life, yet ribosomes are capable of forming only C-N bonds as amides, C-O bonds as esters, and C-S bonds as thioesters. There is no current strategy to site-selectively form C-C bonds as ketones embedded in the backbones of ribosomal products. As an alternative to direct ribosomal C-C bond formation, here we report that peptides containing a dehydrolactic acid motif rapidly isomerize to generate b

Ligand-Controlled Ni-Catalyzed Regiodivergent Hydroacylation of Unactivated Alkenes with Acyl Chlorides.

Herein, we report a ligand-controlled, amide-assisted, nickel-catalyzed regiodivergent hydroacylation of unactivated alkenes with readily available acyl chlorides. This strategy enables the efficient and selective synthesis of amino ketones with diverse regioselectivities, including α- and β-migratory insertion, as well as Markovnikov-type addition, all governed by ligand tuning. The reaction proceeds under mild conditions, exhibits broad functional group tolerance, and offers a straightforward and cost-effective approach to structurally diverse branched ketones.

Expanding chemical space of N-acyl sulfonamides for carbonic anhydrase inhibitor discovery.

N-Acyl sulfonamides are widely recognized carboxylic acid isosteres that can fill the gap related to acidic compounds in chemical catalogs and bioactivity studies. This study presents the generation of synthetically tractable N-acyl sulfonamide chemical space based on the two-step parallel synthetic methodologies yielding structurally diverse compounds with high efficiency. Amide formation and SNAr combined with primary sulfonamide acylation proved to be highly effective in generating structurally diverse N-acyl sulfonamides with typically good synthesis success rates (up to 83 %) and average …

Effect of Fatty Acyl Composition for Lysophosphatidylinositol on Neuroinflammatory Responses in Primary Neuronal Cultures.

Lysophosphatidylinositol (LPI) is an endogenous signaling molecule for the GPR55 receptor. Previous studies have shown that arachidonoyl-lysophosphatidylinositol (LPI-20:4) produced an increase in the inflammatory mediators NLPR3 (inflammasome-3 marker) and IL-1b in neurons from both rat dorsal root ganglion (DRG) and hippocampal cultures. Because LPI is comprised of a family of lipid structures that vary in fatty acyl composition, the current work examined neuroinflammatory responses to various LPI structures in DRG and hippocampal cultures as assessed by high-content fluorescent imaging. DDHD2 possesses both lipase and transacylase capacities that remodel triglyceride acyl chains.

Hereditary spastic paraplegia subtype SPG54 is a genetic neurological disorder caused by mutations in the DDHD2 gene. Excessive lipid drople … acyl chain remodeling. Adipose triglyceride lipase, but not hormone sensitive lipase, also has transacylation activity and can remodel TAG acyl chains, but to a lesser extent than DDHD2. Taken together, this provides evidence that DDHD2 is a neutral lipid lipase and transacylase whose broad specificity enables TAG acyl-chain remodeling.

Chemoselective Approach to Versatile Acyl Fluorides by Photoinduced Activation of p-Methoxybenzyl Esters.

A new strategy for the metal-free photoinduced activation of p-methoxybenzyl esters is developed using Selectfluor and benzil for the generation of acyl fluoride intermediates that enable various transformations. The highlight of this activation method is its high chemoselectivity in the presence of other functionalities, such as esters, amides, and ketones. A synthetic application for the preparation of peptide mimetics that possess two different amide units is also described.
