A chiral stationary phase (CSP) was prepared by coating amylose-tris(3,5-dimethylphenyl carbamate) (ADMPC) onto aminopropylated spherical silica gel. We present a method for the enantioselective analysis of albendazole sulfoxide (ABZSO) in plasma for application in clinical pharmacokinetic studies. ABZSO enantiomers were separated on a 5-micron Chiralcel OB-H column (4.6 x 150 mm) using hexane:ethanol (93:7, v/v) as the mobile phase and fluorescence detection. With the objective of knowing adequately the spectrum of activity of albendazole against intestinal helminthiases, we made observations regarding hymenolepiasis caused by Hymenolepis nana. Two series of investigations were carried out: a) treatment of mice with single doses of either 25 or 50 mg/kg, repeated after ten days, using as controls animals treated with 25 mg/kg of praziquantel or not treated with any antiparasitic drugs; b) treatment of adults and children with 400 mg daily for three consecutive days, repeated after ten days.

One hundred two children (43 males and 59 females) aged 6 to 14 years with positive stool examination by Kato-Katz and/or Harada-Mori culture techniques for N. americanus, were randomly divided into two groups. A clinical trial concerning the efficacy of albendazole has been performed in an area outside the endemic zone. Sixty-eight patients were treated with albendazole in doses varying from 4-30 mg/kg/day for 3-6 days. A high cure rate of 86 per cent was obtained with albendazole 16 mg/kg/day for three days and repeated once after a fortnight. The albendazole was well tolerated and no side effects were imputed to the treatment. Successful treatment was confirmed by seven negative, consecutive coprocultures. The follow up period ranged from 35 to 615 days.

The efficacy of multiple and single dose regimens of albendazole on Trichuris trichiura infection was evaluated by counting the number of worms expelled/day from two pair-matched groups of children, for nine days following therapy. Albendazole was given to some men for the first time. 131 patients (43 children) received different dosages, without clinical intolerance. Biological parameters were normal. Each study was made in two parts: - determination of the good dosage, - administration in a double-blind study (except for Strongyloides Stercoralis). Albendazole has a good efficiency against Enterobius Vermicularis, Trichuris trichiura, Ascaris lumbricoides and Necator americanus. Strongyloides Stercoralis seems to be killed by this drug, but complementary studies will be necessary.
