Aryl-substituted cyclopropyl acetylenes as sensitive mechanistic probes in the gold-catalyzed hydration of alkynes. Comparison to the Ag(I)-, Hg(II)-, and Fe(III)-catalyzed processes.

The gold-catalyzed hydration of 2-phenyl- or 2,2-diphenylcyclopropyl acetylene, sensitive probes to trace the formation of vinyl carbocations, provides exclusively the corresponding cyclopropyl methyl ketones. On the other hand, in the Ag(I)- or Fe(III)-catalyzed hydration, a profound vinyl carbocationic character appears in the initially formed metal-alkyne complexes, as judged by the partial (Ag(+)) or exclusive (Fe(3+)) formation of allene-type rearrangement products. These findings provide clear evidence for subtle electronic differences in metal-alkyne complexes, including Au(I or III), A

Biocompatible alkyne arms containing Schiff base fluorescence indicator for dual detection of CdII and PbII at physiological pH and its application to live cell imaging.

An alkyne arms containing salen-type Schiff base ligand, 6,6'-((1E,1'E)-(1,2-phenylenebis(azanylylidene))bis(ethan-1-yl-1-ylidene))bis(3-(prop-2-yn-1-yloxy)phenol) (H2L), is reported here as a dual chemosensor for CdII and PbII ions. The ligand H2L was characterized by various spectral methods. The ligand H2L acts as a dual sensor for CdII and PbII in methanol/HEPES buffer (5 mM, pH 7.3; 1 : 9 v/v) at room temperature with well-separated excitation and emission wavelengths. The emission intensity at 553 nm of H2L is shifted and a new band appeared at 578 nm with the increase in the presence of

Cu/Pd-Catalyzed Tandem Carbonylative Synthesis of Ester-Containing 3,4-Dihydroquinolin-2(1H)-one Derivatives from Alkyne-Tethered α-Bromocarbonyls.

A Cu/Pd-catalyzed tandem radical cyclization and carbonylation of alkyne-tethered α-bromocarbonyls with phenols has been developed for the expedited construction of ester-containing 3,4-dihydroquinolin-2(1H)-one scaffolds. By employing benzene-1,3,5-triyl triformate (TFBen) as the CO source, the reaction proceeded smoothly to afford a series of ester-containing 3,4-dihydroquinolin-2(1H)-one derivatives in high yields. Moreover, this method could be utilized in the late-stage modifications of bioactive molecules.

A 66-Nuclear All-Alkynyl Protected Peanut-Shaped Silver(I)/Copper(I) Heterometallic Nanocluster: Intermediate in Copper-Catalyzed Alkyne-Azide Cycloaddition.

Ligand-protected heterometallic nanoclusters in contrast to homo-metal counterparts show more broad applications due to the synergistic effect of hetero-metals but their controllable syntheses remain a challenge. Among heterometallic nanoclusters, monovalent Ag-Cu compounds are rarely explored due to much difference of Ag(I) and Cu(I) such as atom radius, coordination habits, and redox potential. Encouraged by copper-catalyzed alkyne-azide cycloaddition (CuAAC) reaction, comproportionation reaction of Cu(II)X2 and Cu(0) in the presence of (PhC≡CAg)n complex and molybdate generated a core-shell

Your mother was right, washing matters: An alkyne-analog of ibuprofen reveals unwanted reactivity of aromatic compounds with proteins during copper-catalyzed click chemistry.

Bioorthogonal chemistry, in particular the copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC), has enabled the robust identification of covalent protein targets of probes and drugs. Ibuprofen is commonly used pain and fever reducer and is sold as an enantiomeric racemate. Interestingly, the stereoisomers can be enzymatically converted through an ibuprofen-CoA thioester intermediate, which might non-specifically react with protein nucleophiles. Here, we use an alkyne-analog of ibuprofen to make two discoveries. First, we find that ibuprofen likely does not result in notable chemical labelin

Computational Study Revealing the Mechanistic Origin of Distinct Performances of P(O)-H/OH Compounds in Palladium-Catalyzed Hydrophosphorylation of Terminal Alkynes: Switchable Mechanisms and Potential Side Reactions.

Pd-catalyzed hydrophosphorylation of alkynes with P(O)-H compounds provided atom-economical and oxidant-free access to alkenylphosphoryl compounds. Nevertheless, the applicable P(O)-H substrates were limited to those without a hydroxyl group except H2P(O)OH. It is also puzzling that Ph2P(O)OH could co-catalyze the reaction to improve Markovnikov selectivity. Herein, a computational study was conducted to elucidate the mechanistic origin of the phenomena described above. It was found that switchable mechanisms influenced by the acidity of substrates and co-catalysts operate in hydrophosphorylat

NXS/TBHP as a Reagent System for Expedient Generation of Halogen Radicals: Site-Selective Halogenation of Quinoxalin-2(1H)-ones and Conversion of Terminal Alkynes to gem-Dihaloketones.

We report an efficient and practical protocol for the generation of halogen (Br and Cl) radicals using inexpensive and readily available N-bromo/N-chlorosuccinimide (NXS) and t-butylhydroperoxide (TBHP) at rt. The NXS/TBHP reagent systems are demonstrated for site-selective bromination and chlorination of the benzene-core of quinoxalin-2(1H)-ones. It is shown that the reagent system permits regioselective halogenation at the C7 position of the benzene core over the readily-functionalized C3 position of quinoxalin-2(1H)-ones under mild reaction conditions. We further show that terminal alkynes

Cationic and Neutral PdII and PtII Pincer Complexes of Phosphinamino-Triazolyl-Pyridine [PN(H)N]: Pincer Ligand-Stabilized Palladium Nanoparticles and Their Catalytic Annulation of Internal Alkynes to Indenones.

We describe the synthesis of a triazolyl-pyridine-based aminophosphine, N-(diphenylphosphaneyl)-6-(1-phenyl)-1H-(1,2,3-triazol-4-yl)pyridine-2-amine [2,6-{(PPh2)-N(H)(C5H3N)(C2HN3C6H5)}] [1, PN(H)N hereafter], and its palladium and platinum complexes and their catalytic application. The reaction of 1 with [M(COD)Cl2] (M = Pd or Pt) afforded the cationic complex [(MCl){PN(H)N}-κ3-P,N,N]Cl [M = Pd (2) or Pt (3)]. Alternatively, compounds 2 and 3 were also synthesized by treating [2,6-{H2N(C5H3N)(C2HN3C6H5)}] (A) with [M(COD)Cl2] (M = Pd or Pt), followed by the addition of stoichiometric amounts

Solvent-controlled two-step one-pot syntheses of α-X (X = Br or Cl) enamino ketones/esters and 3-(2,5-dioxopyrrolidin-1-yl)acrylate by using terminal carbonyl alkynes.

A new two-step one-pot aminobromination/chlorination of carbonyl alkynes has been achieved via a Michael addition of aliphatic secondary amines and subsequent β-bromination/chlorination of the obtained enamines to afford various α-X (X = Br or Cl) enamino ketones/esters in moderate to good yields. A solvent-controllable protocol has been developed to produce versatile 3-(2,5-dioxopyrrolidin-1-yl)acrylates in moderate yields by using toluene as the solvent and chain alkyl propiolates as alkynyl substrates.

Close the ring to break the cycle: tandem quinolone-alkyne-cyclisation gives access to tricyclic pyrrolo[1,2-a]quinolin-5-ones with potent anti-protozoal activity.

Expanding the chemical space of quinolones led to a tandem quinolone-alkyne-cyclisation reaction allowing chemoselective control of the synthesis of tricyclic pyrrolo[1,2-a]quinolin-5-ones. Importantly, we discovered anti-protozoal activity against Plasmodium and Toxoplasma with specific potency of one of the compounds against the liver stage of the malaria parasite in the nanomolar range.

Decoration of Coiled-Coil Peptides with N-Cysteine Peptide Thioesters As Cyclic Peptide Precursors Using Copper-Catalyzed Azide-Alkyne Cycloaddition (CuAAC) Click Reaction.

The development of a copper-catalyzed azide-alkyne cycloaddition (CuAAC) protocol for the decoration of coiled coils with N-cysteine peptide thioesters as cyclic peptide precursors is presented. The reaction conditions include tert-butanol/PBS as the solvent and CuSO4/THPTA/ascorbate as the catalytic system. During these studies, partial formylation of N-terminal cysteine peptides is observed. Mechanistic analysis leads to identification of the formyl source and, hence, to the development of reaction conditions, under which the undesired side reaction was suppressed.

Development of Noval, Recyclable, and Tunable Aryl Imidazolium Salts with a Dual Brønsted-Lewis Acid Character as Catalysts for the Copper(I)-Catalyzed Azide-Alkyne Cycloaddition Reaction (CuAAC).

The development of novel aryl imidazolium salts (ISs) with dual Brønsted-Lewis acid character represents a significant advancement in the field of green chemistry and catalysis. These imidazolium salts serve as efficient catalysts for the copper-(I)-catalyzed azide-alkyne cycloaddition (CuAAC) reaction, a cornerstone of click chemistry. The unique design of the aryl imidazolium framework, incorporating both Brønsted and Lewis acid sites, enhances the catalytic efficiency and selectivity of the CuAAC reaction. Furthermore, the use of these imidazolium salts as catalysts eliminates the need for

Modular Medical Imaging Agents Based on Azide-Alkyne Huisgen Cycloadditions: Synthesis and Pre-Clinical Evaluation of 18 F-Labeled PSMA-Tracers for Prostate Cancer Imaging.

Since the seminal contribution of Rolf Huisgen to develop the [3+2] cycloaddition of 1,3-dipolar compounds, its azide-alkyne variant has established itself as the key step in numerous organic syntheses and bioorthogonal processes in materials science and chemical biology. In the present study, the copper(I)-catalyzed azide-alkyne cycloaddition was applied for the development of a modular molecular platform for medical imaging of the prostate-specific membrane antigen (PSMA), using positron emission tomography. This process is shown from molecular design, through synthesis automation and in vit

Identification of recombinant AtPYL2, an abscisic acid receptor, in E. coli using a substrate-derived bioactive small molecule, a biotin linker with alkyne and amino groups, and a protein cross-linker.

Target protein identification of bioactive small molecules is one of the most important research in forward chemical genetics. The affinity chromatography technique to use a resin bound with a small molecule is often used for identification of a target protein of a bioactive small molecule. Here we report a new method to isolate a protein targeted with a bioactive small molecule using a biotin linker with alkyne and amino groups, protein cross-linker containing disulfide bond, and a bioactive small molecule with an azido group (azido probe). After an azido probe is associated with a target pro

Potent naphthoquinones against antimony-sensitive and -resistant Leishmania parasites: synthesis of novel α- and nor-α-lapachone-based 1,2,3-triazoles by copper-catalyzed azide-alkyne cycloaddition.

Continuing our screening program for novel anti-parasite compounds, we synthesized seven 1,4-naphthoquinones coupled to 1,2,3-triazoles, five nor-β-lapachone-based 1,2,3-triazoles and ten α-lapachone-based 1,2,3-triazoles. These and other naphthoquinonoid compounds were evaluated for their activity against promastigote forms of antimony-sensitive and -resistant strains of Leishmania infantum (syn. Leishmania chagasi) and Leishmania amazonensis. The toxicity of these compounds to mammalian cells was also examined. The substances were more potent than an antimonial drug, with IC50 values ranging

A thiol-alkyne click chemistry-driven lipoic acid precise delivery platform for targeted therapy of cerebral ischemic stroke via redox homeostasis restoration and neuroinflammatory cascade suppression.

Redox homeostasis imbalance, an early triggering event in cerebral ischemic stroke (CIS), precipitates neuronal death and neuroinflammatory response. The endogenous redox pair α-lipoic acid (LA) and dihydrolipoic acid (DHLA) serve as potent antioxidants and possess enormous potential for treating ischemia/reperfusion-induced organ injury. However, their application is constrained by a short half-life and DHLA's susceptibility to metal ion complexes in the bloodstream. Herein, a borneol (Bo)-mediated brain-targeted, ROS-responsive release of DHLA hyperbranched polymer nanomedicine (Bo-HBP(LA) N

A tri-modal biosensor based on alkyne-functionalized gold nanoparticles and molecular beacon for detection of resistance-related point mutations in epidermal growth factor receptor-tyrosine kinase inhibitors.

BACKGROUND: Acquired resistance to epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors in non-small cell lung cancer (NSCLC) is frequently driven by point mutations such as EGFR T790M. This poses a major clinical challenge that significantly compromises treatment efficacy and patient survival. Conventional detection methods often rely on single-mode signaling and lack built-in validation, which limits their reliability and accessibility, particularly in resource-limited settings. METHODS AND RESULTS: We developed a tri-modal biosensor that integrates alkynyl-modified gold nanop

Photochemical or electrochemical bond breaking - exploring the chemistry of (μ2-alkyne)Co2(CO)6 complexes using time-resolved infrared spectroscopy, spectro-electrochemical and density functional methods.

The photochemistry of (μ2-CRCR')Co2(CO)6 complexes (R = pyrenyl, R' = H; R = pyrenyl, R' = ferrocenyl; R = ferrocenyl, R = H) was investigated by ps-time-resolved infrared spectroscopy at room temperature in dichloromethane solution. The main focus of these studies was to determine the primary photoprocess relevant to the light assisted Pauson-Khand reaction. These studies were supported by spectro-electrochemical investigations and density functional calculations which suggest that the primary process to initiate the Pauson-Khand reaction involves a homolytic cleavage of the Co-Co bond formin
