Environmental infestation and rickettsial infection in ticks in an area endemic for Brazilian spotted fever.

Brazilian spotted fever (BSF), caused by Rickettsia rickettsii, is endemic in the municipality of Americana, southeastern Brazil, where the disease is transmitted by the tick Amblyomma cajennense. This study evaluated the tick fauna and rickettsial infection in free-living ticks that were captured monthly using dry ice traps in areas endemic for BSF in Americana, from July 2009 to June 2010. Two tick species were captured: A. cajennense (6,122 larvae; 4,265 nymphs; 2,355 adults) and Amblyomma dubitatum (7,814 larvae; 3,364 nymphs; 1,193 adults). The immature stages of A. cajennense and A. dubi…

Mapping Potential Risks for the Transmission of Spotted Fever Rickettsiosis after Environmental Changes in an Atlantic Forest Region in Brazil.

Brazilian Spotted Fever (BSF), caused by Rickettsia rickettsii, is a fatal tick-borne disease endemic to southeastern Brazil. This study eva … ticks identified Rickettsia bellii and a Rickettsia tamurae/Rickettsia monacensis-like genotype in Amblyomma dubitatum and Amblyomma sculptum, respectively; however, R. rickettsii DNA was not detected in any vector. These findings suggest a dichotomy in transmission cycles: a peri-urban cycle with high canine exposure driven by A. sculptum, and a riparian …

Multiplex TaqMan® Quantitative PCR Assays for Host-Tick-Pathogen Studies Using the Guinea Pig-Tick-Rickettsia System.

Spotted Fever Rickettsiosis (SFR) is caused by spotted fever group Rickettsia spp. (SFGR), and is associated with symptoms common to other illnesses, making it challenging to diagnose before detecting SFGR-specific antibodies. The guinea pig is a valuable biomedical model for studying Spotted Fever Rickettsiosis (SFR); its immune system is more like the human immune system than that of the murine model, and guinea pigs develop characteristic clinical signs. Thus, we have a compelling interest in developing, expanding, and optimizing tools for use in our guinea pig-Amblyomma-Rickettsia system f…

Parasites of domestic and wild animals in South Africa. XXX. Ectoparasites of kudus in the eastern Transvaal Lowveld and the eastern Cape Province.

Sets of four kudus were shot and examined for arthropod parasites at approximately monthly intervals from April 1981 to March 1983 in the southern part of the Kruger National Park, eastern Transvaal Lowveld. These animals harboured 10 ixodid tick species of which Boophilus decoloratus followed by Amblyomma hebraeum were the most abundant. The seasonal abundances of these ticks and of Amblyomma marmoreum, Rhipicephalus appendiculatus, Rhipicephalus evertsi evertsi and Rhipicephalus zambeziensis were determined. The kudus were also infested with 3 lice and 1 louse fly species, as well as the nym…

Beyond the Bite: Shedding Light on Alpha-Gal Syndrome.

Alpha-gal syndrome (AGS) is an emerging tick-borne allergic condition associated with bites from the lone star tick (Amblyomma americanum). Unlike typical tick-borne diseases caused by bacteria or viruses, in AGS there is a delayed allergic reaction to galactose-α-1,3-galactose, a sugar referred to as alpha-gal, which is present in tick saliva; most mammalian meat, such as beef, pork, and lamb; animal-derived products, such as gelatin and dairy (including cow's milk); and certain medications containing animal products. AGS was first identified in the early part of this century, with a notable …

Molecular detection of Ehrlichia ruminantium in ticks from ruminants during the 2021 Rift Valley fever outbreak in Mananjary, Madagascar.

Ehrlichia ruminantium, the causative agent of heartwater, is a tick-borne pathogen affecting livestock in Africa and the Caribbean. This disease is transmitted primarily by Amblyomma variegatum ticks and poses a significant threat to animal health. In Madagascar, the prevalence of E. ruminantium remains poorly documented. During a Rift Valley fever (RVF) outbreak in Mananjary, Madagascar (April-May 2021), we conducted a field study to assess the circulation of vector-borne pathogens in ticks collected from ruminants. Ticks were morphologically identified, and DNA was extracted for quantitative…

Immunoproteomic profiling of Rickettsia parkeri and Rickettsia amblyommii.

Rickettsia parkeri is an Amblyomma-associated, spotted fever group Rickettsia species that causes an eschar-associated, febrile illness in multiple countries throughout the Western Hemisphere. Many other rickettsial species of known or uncertain pathogenicity have been detected in Amblyomma spp. ticks in the Americas, including Rickettsia amblyommii, "Candidatus Rickettsia andeanae" and Rickettsia rickettsii. In this study, we utilized an immunoproteomic approach to compare antigenic profiles of low-passage isolates of R. parkeri and R. amblyommii with serum specimens from patients with PCR- a…

Ticks and Tick-Borne Pathogens Associated with Dromedary Camels (Camelus dromedarius) in Northern Kenya.

Ticks and tick-borne pathogens (TBPs) are major constraints to camel health and production, yet epidemiological data on their diversity and impact on dromedary camels remain limited. We surveyed the diversity of ticks and TBPs associated with camels and co-grazing sheep at 12 sites in Marsabit County, northern Kenya. We screened blood and ticks (858 pools) from 296 camels and 77 sheep for bacterial and protozoan TBPs by high-resolution melting analysis and sequencing of PCR products. Hyalomma (75.7%), Amblyomma (17.6%) and Rhipicephalus (6.7%) spp. ticks were morphologically identified and con…

Novel quaranjavirus and other viral sequences identified from ticks parasitizing hunted wildlife in Trinidad and Tobago.

Hunters are at a higher risk for exposure to zoonotic pathogens due to their close interactions with wildlife and arthropod vectors. In this study, high throughput sequencing was used to explore the viromes of two tick species, Amblyomma dissimile and Haemaphysalis juxtakochi, removed from hunted wildlife in Trinidad and Tobago. We identified sequences from 3 new viral species, from the viral families Orthomyxoviridae, Chuviridae and Tetraviridae in A. dissimile.

Tick-crocodilian interactions: a review, with the first record of tick (Acari: Ixodidae) infestation in the saltwater crocodile (Crocodylus porosus), and a concise host-parasite index.

Interactions between ticks and crocodilians (crocodiles, alligators, caiman, and gharials) are poorly studied but may have significant bearing on the ecology and health of these reptiles. The first record of tick infestation of the saltwater crocodile (Crocodylus porosus) is reported along with the first case of infestation by Amblyomma cordiferum on Cuvier's dwarf caiman (Paleosuchus palpebrosus). A review is also provided of tick-crocodilian interactions with a concise host-parasite index.

[STARI--a new tick borne spirochetosis].

Southern Tick Associated Rash Illness (STARI), also known as Masters disease is caused by Borrelia lonestari spirochetes and it is transmitted by Amblyomma americanum tick. Early symptoms of STARI are similar to early Lyme borreliosis (skin rash resembling Erythema migrans). In diagnostic of STARI traditional methods are not effective. The decision of antibiotic treatment (similar to Lyme borreliosis) still remain based on clinical picture.

Scarcity of Hepatozoon americanum in Gulf Coast tick vectors and potential for cultivating the protozoan.

American canine hepatozoonosis (ACH) is a debilitating tick-borne disease characterized by pyrexia, body wasting, myopathy, mucopurulent ocular discharge, and periosteal proliferation. The causative agent, Hepatozoon americanum, is an apicomplexan that utilizes the Gulf Coast tick, Amblyomma maculatum, as its definitive host and vector. Unlike most tick-borne disease agents, H. americanum is not transmitted via a tick bite, but is transmitted when canids ingest a tick vector that contains sporulated oocysts within the tick hemocoel or paratenic hosts with cystozoites. Our understanding of H. a…
