Adverse hypertrophic scars can form after healing of full-thickness skin wounds. Currently, reliable animal and in vitro models to identify and test novel scar reducing therapeutics are scarce. Here we describe the development and validation of a tissue-engineered human hypertrophic scar (HTscar) model based on reconstructed epidermis on a dermal matrix containing adipose derived mesenchymal stem cells (ASC). Although obtained from normal, healthy skin, ASC, in contrast to dermal mesenchymal cells, were found to facilitate HTscar formation. Quantifiable HTscar parameters were identified: contr

Degeneration of brain tissue following stroke leads to functional impairment with limited brain self-repair. New evidence suggests that delivery of circulating CD34(+) human umbilical cord blood cells can produce functional recovery in an animal stroke model with concurrent angiogenesis and neurogenesis leading to some restoration of cortical tissue. While some alternative interpretations of this data are offered herein, the study provides encouraging evidence of functional recovery from stroke in an animal model using stem cell therapy.

Hepatocyte nuclear factor-4α (HNF4α) presents in multiple isoforms generated using alternative promoter (P1 and P2) and splicing. Neither conservation of tissue distribution of HNF4α isoforms, nor presence of alternative promoter usage is known. In this study, to detect the expression of HNF4α in some species of animals, we have applied monoclonal antibodies against P1 (K9218) and P2 (H6939) promoter-driven and P1/P2 promoter-driven H1415 HNF4α for immunohistochemistry and western blot analysis. Antibody K9218 was observed in the hepatocytes, proximal tubules of the kidney, and epithelial cell

The occurrence of BSE cases in Germany after the ban of meat and bone meal for ruminant feed in 1994 requires a detailed investigation of animal derived feedstuffs regarding their specific risks as vectors for the disease. Accepting the theory that BSE is a prion transmitted disease, the theoretical infectious potential was calculated for animal derived feedstuffs. This calculation was based on the assumption, that risk material (brain, spinal cord) of one clinically diseased cattle was rendered in the process as established in Germany (133 degrees C, 3 bar, 20 min) or, alternatively, that one

Drug-induced liver injury (DILI) presents a major challenge not only in new drug development but also in post-marketing withdrawals and the safety of food, cosmetics, and chemicals. Experimental model organisms such as the rodents have been widely used for preclinical toxicological testing. However, the tension exists associated with the ethical and sustainable use of animals in part because animals do not necessarily inform the human-specific ADME (adsorption, dynamics, metabolism and elimination) profiling. To establish alternative models in humans, in vitro hepatic tissue models have been p

Young and aged animals were tested on a spatial alternation task that consisted of two interleaved components: (1) an "outbound" or alternation component (working memory) and (2) an "inbound" component, requiring the animal to remember to return to a central location in space (spatial memory). In the present study, aged rats made more outbound errors throughout testing, resulting in significantly more days to reach learning criterion, as compared to young rats. Furthermore, while all animals were able to learn the hippocampus-dependent inbound component of the task, most aged animals remained

In controlling Rift Valley fever, public health sector optimises health benefits by considering cost-effective control options. We modelled cost-effectiveness of livestock RVF control from a public health perspective in Kenya. Analysis was limited to pastoral and agro-pastoral system high-risk areas, for a 10-year period incorporating two epidemics: 2006/2007 and a hypothetical one in 2014/2015. Four integrated strategies (baseline and alternatives), combined from three vaccination and two surveillance options, were compared. Baseline strategy included annual vaccination of 1.2-11% animals plu

Hormonal disturbances, such as hyperandrogenism, are considered important for developing polycystic ovary syndrome (PCOS) in humans. Accordingly, directly hormone-regulated animal models are widely used for studying PCOS, as they replicate several key PCOS features. However, the pathogenesis and treatment of PCOS are still unclear. In this review, we aimed to investigate animal PCOS models and PCOS-like phenotypes in animal experiments without direct hormonal interventions and determine the underlying mechanisms for a better understanding of PCOS. We summarized animal PCOS models that used ind

The live Brucella melitensis Rev 1 strain is considered the best vaccine available for the prophylaxis of brucellosis in small ruminants. The classically recommended exclusive vaccination of young replacement animals has failed to control brucellosis in some developed countries and is frequently inapplicable in the developing world. Accordingly, whole-flock vaccination is the only feasible alternative to control B. melitensis infection in small ruminants under the extensive management conditions characteristic of these countries. This review describes the practical problems encountered and the

Complementary and alternative medicine is frequently used by inflammatory bowel disease (IBD) patients; most common are massage, acupuncture, and moxibustion therapy. Massage therapy is poorly studied in IBD patients; therefore, its benefits remain unknown. Acupuncture and moxibustion therapy have been shown to improve inflammation and symptoms in animal and human studies. However, current clinical trials of acupuncture and moxibustion are of insufficient quality to recommend them as alternative therapy. Nonetheless, because these therapies seem generally to be safe, they may have a role as co

The use of controlled mating or artificial insemination is impracticable in the case of large herds, mainly because of labour costs and the need to delimit areas during the breeding period. However, the exclusion of information from animals with uncertain paternity reduces genetic progress. The objectives of this study were as follows: (i) propose an iterative empirical Bayesian procedure to implement the hierarchical animal model (ITER); (ii) calculate the posterior probabilities of paternity by the maximum likelihood method following the concepts; (iii) compare an average numerator relations

Glymphatic transport in rodents has primarily been studied using cisterna magna cannulation (CMC), a minimally invasive method for cerebrospinal fluid (CSF) tracers infusion. However, CMC is suboptimal due to the lack of bony structures to stabilize the cannula, leading to potential movement artifacts. Here, we present an alternative approach involving chronic cannulation of the lateral ventricles of mice for CSF tracer delivery. A direct comparison demonstrated that intraventricular cannulation (IVC) reproduces CMC results in vivo, including perivascular labeling of the middle cerebral artery

Ticks are blood-feeding ectoparasites that cause significant economic losses by reducing animal productivity and damaging hides and skins. Ticks control and prevention strategies primarily rely on the regular use of synthetic acaricides. However, the widespread misuse of these commercial acaricides has led to the emergence of acaricide-resistant ticks, compromising their sustainable effectiveness. Therefore, this study aimed to evaluate alternative plant-based acaricides and determine their effective dose levels. Crude ethanol extracts from six plant species, previously reported to possess aca

With the global ban on antibiotic growth promoters, identifying safe, effective alternatives for poultry production is urgent. Probiotics, particularly Bacillus amyloliquefaciens (BA), have shown promise in improving animal health and performance, but research in geese remains limited. This study aimed to evaluate the effects of dietary BA supplementation on growth performance, immune function, antioxidant capacity, intestinal morphology, microbiota, and transcriptome in growing Zi geese. A total of 180 50-day-old male Zi geese were randomly assigned to a control group (basal diet) or an exper

PRACTICAL RELEVANCE: Urinary tract infection (UTI) is an important cause of feline lower urinary tract disease (FLUTD), particularly in female cats older than 10 years of age. In addition to cats with typical clinical signs of FLUTD or upper UTI, many cats have subclinical bacteriuria, but the clinical relevance of this is currently uncertain. UTIs are one of the most important indications for antimicrobial use in veterinary medicine and contribute to the development of antimicrobial resistance. Adherence to treatment guidelines and confinement to a few first-line antimicrobial agents is imper

Sporotrichosis is a subacute or chronic subcutaneous infection, caused by the fungus Sporothrix schenkii complex, occurring in human and animal tissues. Potassium iodide and itraconazole have been used as effective therapy for first-choice treatment, while amphotericin B may be indicated for disseminated infection. However, the adverse effects of potassium iodide and amphotericin B or the long duration of therapy with itraconazole often weigh against their use, leading to the search for alternatives for the treatment of severe infections. Terpinen-4-ol and farnesol are components of essential

The removal and disposal of dead animals and slaughterhouse offalls by rendering plants to produce meat and bone meal (high nutritive value due to the protein and mineral content) is a model for a successful concept of recycling organic matter. Especially since the discussion on BSE and on the role of meat meal in distribution of this disease the products--inspite of their nutritive value--came under criticism. Besides this development more and more owners of companion animals refuse the removal of their animals by rendering plants, increasing their demand of other kinds of disposal (crematori

Adult golden hamsters inoculated subcutaneously with either of two sandfly fever group viruses, Punta Toro and Gabek Forest (Phlebovirus, Bunyaviridae), developed a fulminating fatal illness characterized by hepatic and splenic necrosis and interstitial pneumonitis. Most animals died within three days after infection; this was accompanied by high levels of viremia. Necropsy and histopathologic examination of the infected animals revealed pathologic changes involving multiple organs that resembled those described in Rift Valley fever. These two hamster-phlebovirus systems may serve as alternati

BACKGROUND: Microsurgical training has traditionally relied on live animal models. While these offer high anatomical and functional fidelity, their use raises increasing ethical, logistical, and economic concerns and alternatives are needed.
METHODS: We conducted a prospective observational study to assess the subjective and objective validity of a pump-perfused chicken thigh model for microsurgical training. Forty-five residents and surgeons were categorized by microsurgical experience. Thirteen participants underwent motion analysis and blinded video-based scoring. All participants performe

With increasing restrictions on the use of antibiotics in animal feed, plant-derived feed additives are being used as potential alternatives in poultry production. Macleaya cordata extract (MCE) has antioxidant and immunomodulatory activities, but its effects in geese remain unclear. This study evaluated the effects of dietary MCE on antioxidant capacity, immune function, intestinal morphology, and cecal microbiota in Sichuan White geese. Geese were randomly divided into a control group, a chlortetracycline group, and a MCE group (400 mg/kg) for a 21-day feeding period. Compared with the contr
