PCR-based karyotyping of Anopheles gambiae inversion 2Rj identifies the BAMAKO chromosomal form.

BACKGROUND: The malaria vector Anopheles gambiae is polymorphic for chromosomal inversions on the right arm of chromosome 2 that segregate nonrandomly between assortatively mating populations in West Africa. One such inversion, 2Rj, is associated with the BAMAKO chromosomal form endemic to southern Mali and northern Guinea Conakry near the Niger River. Although it exploits a unique ecology and both molecular and chromosomal data suggest reduced gene flow between BAMAKO and other A. gambiae populations, no molecular markers exist to identify this form. METHODS: To facilitate study of the BAMAK…

Repellency effect of forty-one essential oils against Aedes, Anopheles, and Culex mosquitoes.

Since ancient times, plant products were used in various aspects. However, their use against pests decreased when chemical products became developed. Recently, concerns increased with respect to public health and environmental security requiring detection of natural products that may be used against insect pests. In this study, 41 plant extracts and 11 oil mixtures were evaluated against the yellow fever mosquito, Aedes aegypti (Linnaeus), the malaria vector, Anopheles stephensi (Liston), and the filariasis and encephalitis vector, Culex quinquefasciatus (Say) (Diptera: Culicidae) using the sk…

Stream-bank shade and larval distribution of the Philippine malaria vector Anopheles flavirostris.

The principal malaria vector in the Philippines, Anopheles flavirostris (Ludlow) (Diptera: Culicidae), is regarded as 'shade-loving' for its breeding sites, i.e. larval habitats. This long-standing belief, based on circumstantial observations rather than ecological analysis, has guided larval control methods such as 'stream-clearing' or the removal of riparian vegetation, to reduce the local abundance of An. flavirostris. We measured the distribution and abundance of An. flavirostris larvae in relation to canopy vegetation cover along a stream in Quezon Province, the Philippines. Estimates of …

Anopheles gambiae laminin interacts with the P25 surface protein of Plasmodium berghei ookinetes.

Laminin is a major constituent of the basal lamina surrounding the midgut of the malaria vectors that has been implicated in the development of the Plasmodium oocyst. In this report we describe the cloning of the Anopheles gambiae gene encoding the laminin gamma 1 polypeptide and follow its expression during mosquito development. To further investigate the putative role of laminin in the transmission of the malaria parasite we studied the potential binding of the P25 surface protein of Plasmodium berghei using a yeast two-hybrid system. Heterodimer formation was observed and does not require a…

Heat shock response during development of the malaria vector Anopheles stephensi (Culicidae: Diptera).

The pattern of synthesis of heat shock proteins (HSP) and thermotolerance to elevated temperatures during the development of the malaria vector Anopheles stephensi normally reared at 28 +/- 2 degrees C was studied using SDS-PAGE. In total twelve heat shock proteins (i.e. 31, 33, 38, 43, 44, 51, 57, 62, 69, 71, 113 and 121 kD were induced by heat shock during various stages of development. Eight polypeptides (HSP during one or other of the instars) appeared during normal development of the adult, which showed very little response towards heat shock. Only two polypeptides (57 and 69 kD) were ind…

Metabolic resistance of Anopheles funestus to pyrethroids and organochlorines in Toumbokro, rural Côte d'Ivoire.

Insecticide resistance in malaria vectors threatens control efforts. Anopheles funestus Giles s.l. increasingly contributes to malaria transmission in Côte d'Ivoire especially in central and western regions, yet resistance data remain poorly known. This study assessed the susceptibility of An. funestus s.s. from Toumbokro to major insecticides and identified underlying metabolic resistance mechanisms. Blood-fed An. funestus s.l. females were collected indoors with an electric aspirator and induced to oviposit. F1 progeny from PCR-confirmed An. funestus s.s. were exposed to permethrin (0.75%), …

Effects of a caffeine-based diet on insecticide resistance and longevity in infected Anopheles coluzzii.

BACKGROUND: Alkaloids such as caffeine can be toxic for insects. However, although mosquitoes feed on many plants with nectar-containing alkaloids, the impact of these alkaloids on the vectorial capacity of mosquitoes is not known, in particular regarding increased resistance to insecticides. METHODS: The effects of caffeine on Anopheles coluzzii female mosquitoes in terms of resistance to the insecticide deltamethrin, the rate at which mosquitoes are knocked-down during insecticide exposure, mortality within 48 h of insecticide exposure and longevity following exposure were studied. We also …

Evidence incriminating Anopheles maculatus as a potential vector of Plasmodium knowlesi and Plasmodium inui.

In Indonesia, 29 anopheline species are vectors of human malarias. Many of these morphologically indistinguishable vectors are in species complexes requiring molecular analysis to identify. Data on vectors in North Sumatra remain limited. This study investigated mosquitoes of the Maculatus Subgroup in Ujung Bandar Village, Langkat Regency, to identify them and their potential roles in malaria transmission. Mosquitoes collected via human landing catches (July 2022 - June 2023) were analysed using ITS2 PCR assays and a phylogenetic analysis. Screening for Plasmodium DNA was performed on mosquito

Exploring the gut micriobiome in Anopheles mosquitoes: A promising new approach to malaria control.

Malaria continues to pose significant public health issues globally. This disease is primarily spread through the bites of female Anopheles mosquitoes that are infected with the Plasmodium parasites. While conventional vector control strategies such as insecticide-treated bed nets (ITNs) and indoor residual spraying (IRS) have significantly reduced malaria burden, their long-term effectiveness is increasingly compromised by widespread insecticide resistance. Recent research has proposed manipulation of the Anopheles gut microbiome as a complementary malaria control strategy; however, the stren…

Seasonal variation in aquatic habitat availability and use by the malaria vector, Anopheles funestus.

BACKGROUND: Larval source management (LSM) can be highly effective for controlling malaria vectors such as Anopheles funestus s.s., which typically exploit large and permanent aquatic habitats. While these habitats can persist year-round in endemic regions of Africa, their availability and use shift between wet and dry seasons. Understanding these seasonal changes is essential for identifying the habitats that sustain vector populations and for determining when and where LSM would be most effective. METHODS: We investigated the availability and use of An. funestus larval habitats across wet a…

Decoding cis-regulatory elements in the germline of the human malaria vector Anopheles gambiae.

Cis-regulatory elements (CREs) drive tissue- and cell-specific gene expression and are essential for safe, sustainable genetic control strategies in pest and vector insects, including the engineering of gene drives in the primary human-malaria vector Anopheles gambiae. Yet CREs remain poorly defined in mosquitoes due to limited computational tools and practical methods for identification and validation. We present a systematic in silico approach for CRE discovery, correlating targeted DNA-motif searches with gene expression, followed by frequency and distribution analysis within putative promo…

MODELING THE POTENTIAL CLIMATIC SUITABILITY OF ANOPHELES PSEUDOPUNCTIPENNIS, THE HISTORICAL VECTOR OF MALARIA IN ARGENTINA.

Anopheles pseudopunctipennis is one of the most important malaria vectors in the Americas and historically contributed to transmission in northwestern Argentina. Although Argentina was certified malaria-free in 2019, surveillance of vector populations remains relevant to prevent reintroduction. We used ecological niche models to identify climatically suitable areas for An. pseudopunctipennis in Argentina, using occurrence records and bioclimatic variables from WorldClim 2.1. Records were compiled from the literature, biodiversity databases, and personal collections, and were filtered to remove…

Effects of temperature and nutrition stress on Anopheles stephensi, an Indian urban malaria vector.

BACKGROUND: Changing climate and complexity in ecological landscape can potentially expand the geographic distribution of mosquitoes (Diptera: Culicidae) to adapt and transmit various vector-borne diseases, including malaria. Anopheles stephensi is a potential urban malaria vector in the Indian subcontinent. Temperature and nutrients are the important environmental stressors, which influence the life cycle and vectorial competence of mosquitoes. METHODS: Three experimental setups were designed in the laboratory by exposing An. stephensi to low-temperature (LT) at 4 °C for eggs and 18 ± 1 °C f…
