A comparison of susceptibility results of the Bacteroides fragilis group and other anaerobes by traditional MIC results and statistical methods.

Antimicrobial susceptibility is compared by MIC values reflecting activity by weight and/or percentage of strains resistant. This study establishes these parameters in vitro in the traditional fashion and extends analysis statistically for various groups of anaerobic bacteria. Mode MIC, MIC50, MIC90 values and percentage of strains resistant indicated comparable activity for most of the beta-lactams. Geometric and arithmetic MICs suggested ceftizoxime to be the most active beta-lactam. Statistical analysis of all log2 values showed that metronidazole was the most active followed by clindamycin

Comparison of the activities of penicillin G and new beta-lactam antibiotics against clinical isolates of Bacteroides species.

MICs were determined for 218 clinical isolates of Bacteroides by a broth microdilution method. Imipenem was the most active antibiotic tested. Azlocillin, mezlocillin, and cefoxitin had comparable activities, with resistance among members of the B. fragilis group and B. capillosus. Ceftizoxime was the most active cephalosporin tested. Members of the B. fragilis group showed high levels of resistance to cefotetan and ceftazidime. Resistance to penicillin G varied from 0 to 14%.

Bacteroides uniformis ATCC 8492 Utilizes Lotus Seed Resistant Starch via a PUL18-Driven Exterior-to-Interior Degradation Strategy.

Bacteroides uniformis (Bu) is a promising probiotic, yet its mechanism for utilizing lotus seed resistant starch (LRS) remains unclear. Here, growth analysis, starch structural characterization, transcriptomics, and molecular docking were integrated to elucidate the LRS utilization strategy of Bu. Bu efficiently utilized LRS and released abundant extracellular maltooligosaccharides during fermentation. Structural analyses revealed an exterior-to-interior degradation pattern characterized by the preferential removal of peripheral short chains and progressive surface erosion of starch granules. …

The gut commensal Bacteroides thetaiotaomicron harbors prevalent class A and D β-lactamases with cross-taxa dissemination potential.

The abundant gut commensal Bacteroides thetaiotaomicron is repeatedly challenged by β-lactam exposure in the human intestine, yet its β-lactamase repertoire and dissemination potential remain incompletely characterized. Here, we screened 626 publicly available B. thetaiotaomicron genomes and identified 1059 putative β-lactamase homologs, all belonging to class A or class D families. Four highly prevalent representatives-BTA-1 and CfxA-3 (class A) and OXA-347 and OXA-1327 (class D)-were prioritized for experimental validation. Heterologous expression in Escherichia coli revealed heterogeneous r…

Bacteroides cellulosilyticus-derived 2-hydroxyphenylacetic acid rectifies hepatic lipid homeostasis in MASLD by targeting the PPARγ-CD36 axis.

The gut microbiota plays an important role in the occurrence and development of metabolic dysfunction-associated steatotic liver disease (MASLD), but the specific molecular mechanisms involved have not been fully elucidated. In this study, human cohort studies were performed to identify that the relative abundance of Bacteroides cellulosilyticus (B. cellulosilyticus) was significantly decreased in patients with MASLD. Through the integration of metagenomic and metabolomic analyses, it was confirmed that B. cellulosilyticus and its metabolite 2-hydroxyphenylacetic acid (2HPAA) are key factors r…

Novel Approach for Evaluation of Bacteroides fragilis Protective Role against Bartonella henselae Liver Damage in Immunocompromised Murine Model.

Bartonella henselae is a gram-negative facultative intracellular bacterium and is the causative agent of cat-scratch disease. Our previous data have established that Bacteroides fragilis colonization is able to prevent B. henselae damages through the polysaccharide A (PSA) in an experimental murine model. In order to determine whether the PSA is essential for the protection against pathogenic effects of B. henselae in immunocompromised hosts, SCID mice were co-infected with B. fragilis wild type or its mutant B. fragilis ΔPSA and the effects of infection on murine tissues have been observed by…

Antagonism among Bacteroides fragilis group strains isolated from middle ear exudates from patients with chronic suppurative otitis media.

The production and sensitivity of a bacteriocin-like substance in Bacteroides fragilis group strains isolated from middle ear exudates in children with suppurative otitis media were studied through antagonism assayed by the well method. The results of the crossed reactions showed that 10 strains (66.6%) were bacteriocinogenic, 9 were sensitive to at least 1 bacteriocin (60%), and none showed inhibitory activity against homologous strains. Different patterns of susceptibility to bacteriocin-like substances were observed among strains isolated from the same patient as well as different strains i…

Bacteroides lipopolysaccharides (LPS) induce anaphylactoid and lethal reactions in LPS-responsive and -nonresponsive mice primed with muramyl dipeptide.

Lipopolysaccharides (LPS) (100 micrograms/mouse) from Bacteroides gingivalis elicited anaphylactoid reactions in N-acetylmuramyl-L-alanyl-D-isoglutamine (MDP) (100 micrograms)-primed C3H/HeN mice 0-48 h after MDP injection. The reaction resulted in death within 1 h when the LPS was injected 2-10 h after the MDP injection. LPS prepared from other Bacteroides and Salmonella species but not from others, including Escherichia coli, could induce the reaction. LPS-nonresponsive C3H/HeJ and C57BL/6 and C57BL/10 strains were also responsive to the reaction, while AKR, BALB/c, DBA/2, and ICR mice were …

A phospholipase B effector exploits membrane lipid divergence to drive inter-Bacteroides competition and enables universal genome editing.

The Type VI secretion system (T6SS) mediates interbacterial competition, yet the mechanisms underlying its effector diversity and cell-targeting specificity remain poorly understood. Here, we identify Btle, a phospholipase B (PLB) effector defining a unique Tle subfamily (Tle7), encoded in approximately 18.0% of Bacteroides fragilis strains. Lipidomic profiling reveals that Btle preferentially targets Bacteroides glycerophospholipids, disrupting membrane integrity and driving strict inter-Bacteroides competition, with self-intoxication prevented by the cognate immunity protein Btli1. Guided by…

Early-life colonization by enterotoxigenic Bacteroides fragilis remodels gut epithelial stem cell states to drive colorectal cancer susceptibility.

Early-life microbial exposures can shape lifelong disease risk, yet how developmental timing influences the host-microbe interaction remains unclear. During neonatal colonization by enterotoxigenic Bacteroides fragilis (ETBF), the metalloprotease toxin BFT enables lamina propria niche entry and remodeling of colonic epithelial cell states. Early-life ETBF exposure expands murine Lgr5 + stem cells, enhances Atoh1 + secretory differentiation, and induces distal colon-specific Wnt hyperactivation. These epithelial alterations coincide with increased formation of precancerous lesions in Apc Min/+ …

A human-derived Bacteroides strain attenuates depressive-like behavior in a rat model of social defeat-induced stress.

BACKGROUND: The gut microbiome has been linked to mood disorders via communication along the gut-brain axis. We previously found levels of fecal Bacteroides were inversely associated with brain signatures of depression in the prefrontal cortex of human subjects. Bacteroides are important human commensals, playing a keystone role in regulating the immune system and producing bioactive metabolites, like the neurotransmitter gamma-aminobutyric acid and B vitamins. METHODS: To better understand the link between Bacteroides and depression - and test a candidate novel next-generation probiotic - we…

Intestinal Bacteroides modulates inflammation, systemic cytokines, and microbial ecology via propionate in a mouse model of cystic fibrosis.

Persons with cystic fibrosis (CF), starting in early life, show intestinal microbiome dysbiosis characterized in part by a decreased relative abundance of the genus Bacteroides. Bacteroides is a major producer of the intestinal short chain fatty acid propionate. We demonstrate here that cystic fibrosis transmembrane conductance regulator-defective (CFTR-/-) Caco-2 intestinal epithelial cells are responsive to the anti-inflammatory effects of propionate. Furthermore, Bacteroides isolates inhibit the IL-1β-induced inflammatory response of CFTR-/- Caco-2 intestinal epithelial cells and do so in a…

Bacteroides fragilis Supplementation Deteriorated Metabolic Dysfunction, Inflammation, and Aorta Atherosclerosis by Inducing Gut Microbiota Dysbiosis in Animal Model.

BACKGROUND: The gut microbial ecosystem is an important factor that regulates host health and the onset of chronic diseases, such as inflammatory bowel diseases, obesity, hyperlipidemia, and diabetes mellitus, which are important risk factors for atherosclerosis. However, the links among diet, microbiota composition, and atherosclerotic progression are unclear. METHODS AND RESULTS: Four-week-old mice (-/- mice, C57Bl/6) were randomly divided into two groups, namely, supplementation with culture medium (control, CTR) and Bacteroides fragilis (BFS), and were fed a high-fat diet. The gut microbi…

Comparative in-vitro activity of cefoperazone/sulbactam and 11 other agents against multidrug resistant Bacteroides fragilis group species.

The activity of cefoperazone/sulbactam was compared with eleven other antimicrobial agents against 91 strains of the Bacteroides fragilis group known to be resistant to clindamycin and/or beta-lactam agents. Sulbactam alone possessed moderate activity against most strains and improved the activity of cefoperazone and ampicillin. Bacteroides distasonis strains were more resistant than other species. There was no correlation between resistance of an isolate to either clindamycin or cefoxitin with resistance to cefoperazone/sulbactam.
