OBJECTIVE: Red blood cells (RBCs) must be highly deformable to pass through capillaries narrower than their own diameter for delivering oxygen to the tissues. Ion channels are central regulators of RBC deformability and volume. Gain-of-function mutations in Piezo1, as seen in hereditary xerocytosis, cause RBC dehydration and have been associated with an increased risk of thromboembolism. Here we investigated how pharmacological RBC-Piezo1 activation affects RBC membrane potential, RBC ion content, RBC deformability, coagulation dynamics, and clot structure.

BACKGROUND: Umbilical cord blood has been widely used in clinical transplantation. Blood gas analysis of umbilical cord blood is routinely used to evaluate neonatal asphyxia. This study aimed to evaluate an improved umbilical cord blood collection method that does not affect the results of umbilical cord blood gas analysis and hematopoietic stem cell transplantation-related indices.
METHODS: Three hundred pregnant women were recruited between December 2019 and August 2022. In total, 270 umbilical cord blood samples were included and randomly divided into 3 groups. Group A was defined as the g

Preeclampsia is associated with significant morbidity and mortality. Women who experienced preeclampsia require close blood pressure surveillance postpartum. Remote monitoring of blood pressure using a mobile health application may be a viable method of surveillance in this population. The purpose of this project was to assess the feasibility of using the MyWellSpan mobile application to engage postpartum women who experienced preeclampsia in blood pressure self-monitoring. Women who chose to participate were provided an automatic blood pressure cuff and educational materials and were enrolled

Intensive systolic blood pressure (SBP) control improved outcomes in SPRINT (Systolic Blood Pressure Intervention Trial). Our objective was to expand on reported findings by analysis of baseline characteristics, primary outcomes, adverse events, follow-up blood pressure, and medication use differences by baseline SBP (tertile 1 [T1], <132; tertile 2 [T2], 132-145; and tertile 3 [T3], >145 mm Hg). Participants with higher baseline SBP tertile were more often women and older, had higher cardiovascular risk, and lower utilization of antihypertensive medications, statins, and aspirin. Achieved SBP

Allogeneic hematopoietic cell transplantation (HCT) recipients have substantial transfusion requirements. Factors associated with increased transfusions and the extent of blood product use in umbilical cord blood (UCB) recipients are uncertain. We reviewed blood product use in 229 consecutive adult recipients of allogeneic HCT at the University of Minnesota: 147 with leukemia, 82 lymphoma or myeloma; 58% received unrelated UCB and 43% sibling donor peripheral blood stem cell (PBSC) grafts. Although neutrophil recovery was prompt (UCB median 17, range: 2-45 days, and PBSC 14, range: 3-34 days),

Canine xenotransfusion may be a temporary emergency option for type-B cats when compatible feline blood is unavailable, but the in vitro compatibility of type-B feline blood with canine blood has not been fully characterized. This study evaluated and compared the slide-agglutination crossmatch compatibility of type-B feline blood with canine blood with different dog erythrocyte antigen (DEA) 1 phenotypes and with type-A feline blood. Blood samples were obtained from 70 type-B cats, 17 type-A cats, 26 DEA 1-positive dogs, and 18 DEA 1-negative dogs. Among 300 major crossmatches, incompatibility

Blood transfusions come with risks and high costs, and should be utilized only when clinically indicated. Decisions to transfuse are however not always well informed, and lack of clinician knowledge and education on good clinical transfusion practices contribute to the inappropriate use of blood. Low and middle-income countries in particular take much strain in their efforts to address blood safety challenges, demand-supply imbalances, high blood costs as well as high disease burdens, all of which impact blood usage and blood collections. Patient blood management (PBM), which is a patient-focu

BACKGROUND: Many blood centers now accept donors with diabetes, provided their condition is "well-controlled". However, glycemic control is not routinely assessed at donation, and the impact of donor diabetes on blood product quality remains unclear.
STUDY DESIGN AND METHODS: To screen glycemic control, glycated hemoglobin A1c (HbA1c) was measured in whole blood from 256 unique donors with either type 1 diabetes (T1D), type 2 diabetes (T2D), or without known diabetes. In parallel, routine quality control (QC) data were linked with donor metadata to assess the impact of donor diabetes on red c

The search for an efficacious blood substitute has remained a persistent challenge for more than a century. Despite extensive efforts to develop synthetic oxygen carriers, no broadly approved alternative to human blood has yet been realized. In this review, we first provide an overview of the historical development and repeated failures of artificial blood substitutes, while highlighting the continuing clinical demand for transfusion alternatives. We then discuss recent setbacks in blood substitute development, particularly adverse events observed in human trials. Current research efforts with

BACKGROUND: The most frequently ordered laboratory test worldwide is the complete blood count (CBC). As clinical chemists are increasingly assigned to assist or direct laboratories outside of the traditional clinical chemistry sections, such as the automated hematology section, expertise must be established. This review article is a dedication to that ongoing effort.
CONTENT: In this primer, the white blood cell (WBC) test components of the CBC are introduced, followed by a discussion of the laboratory evaluation of leukopenia and leukocytosis.
SUMMARY: The laboratorian's approach to consult

BACKGROUND: Because knowledge of blood donor motivation is crucial in guiding recruitment and retention efforts, the present study aimed at developing and validating a new scale as a multidimensional measure of blood donation motivation from the perspective of the self- determination theory.
METHODS: This study was conducted in three phases from September 2022 to May 2023. The first phase involved developing a draft scale based on a literature review. In the second phase, face and content validity were performed. The third phase used a cross-sectional design to assess the construct validity a

BACKGROUND: Masked hypertension is more common in individuals with type 2 diabetes than in individuals with normoglycemia. We aimed to explore if there is a discrepancy between office blood pressure (office BP) and home blood pressure monitoring (HBPM) in relation to HbA1c as well as glycemic status in 5,029 middle-aged individuals.
METHODS: HBPM was measured in a subsample of 5,029 participants in The Swedish CardioPulmonary BioImage Study (SCAPIS), a population-based cohort of 50-64 years old participants. Both office BP and HBPM were obtained after 5 minutes' rest using the semiautomatic O

BACKGROUND: Air leak is the most common complication after lung resection and leads to increased length of hospital (LOH) stay or patient discharge with a chest tube. Management by autologous blood patch pleurodesis (ABPP) is controversial because few studies exist, and the technique has yet to be standardized.
METHODS: We retrospectively reviewed patients undergoing ABPP for prolonged air leak (PAL) following lobectomy in three centers, between January 2014 and December 2019. They were divided into two groups: Group A, 120 mL of blood infused; Group B, 60 mL. Propensity score-matched (PSM) a

The blood-brain barrier (BBB), blood-cerebrospinal fluid (CSF) barrier (BCSFB), and CSF-brain barriers (CSFBB) are highly regulated barriers in the central nervous system comprising complex multicellular structures that separate nerves and glia from blood and CSF, respectively. Barrier damage has been implicated in the pathophysiology of diverse hypoxia-related neurological conditions, including stroke, multiple sclerosis, hydrocephalus, and high-altitude cerebral edema. Much is known about the damage to the BBB in response to hypoxia, but much less is known about the BCSFB and CSFBB. Yet, it
