The search for an efficacious blood substitute has remained a persistent challenge for more than a century. Despite extensive efforts to develop synthetic oxygen carriers, no broadly approved alternative to human blood has yet been realized. In this review, we first provide an overview of the historical development and repeated failures of artificial blood substitutes, while highlighting the continuing clinical demand for transfusion alternatives. We then discuss recent setbacks in blood substitute development, particularly adverse events observed in human trials. Current research efforts with

BACKGROUND: We have demonstrated previously that valproic acid (VPA), a histone deacetylase inhibitor, can improve survival in lethal models … ed significant anemia (Hb<6 g/dL) and lactic acidosis (lactate 3-5 mmol/L). Acidosis improved after blood transfusion and worsened in the other two groups. VPA treatment increased phospho-Akt (activated), phospho-GSK-3beta (Glycogen synthase kinase 3beta), beta-catenin and Bcl-2 (B-cell leukemia/lymphoma 2) protein levels compared to control group (P = .01, .01, .0 …

The therapy by blood transfusion, blood plasma and infusion solutions, duration and dosage are individual for each patient and dependent on the previous state of health of a patient, size of injure, degree of bleeding, expressed shock and disturbed general state. For taking care of the polytraumatized persons and especially for taking care of the parenhematozic organs and gastrointestinal bleedings greater quantities of blood are required. We assessed the seriousness of shock and hypovolemia on the basis of the clinical picture, decreased arterial pressure, haemogram, hematocrytes and diuresis…

Red blood cell (RBC) transfusion is essential for treating severe anemia, acute blood loss, and RBC production disorders by restoring oxygen-carrying capacity. Ensuring the safety and efficacy of transfused RBCs involves collection, processing, and storage. Despite optimal conditions, stored RBCs experience the "storage lesion," involving biochemical and morphological changes that affect their quality. Post-transfusion recovery (PTR), indicating the survival and functionality of transfused RBCs, is influenced by the storage lesion and varies among donors due to genetic and environmental factor

OBJECTIVES: Prehospital blood transfusion (PHBT) improves outcomes among patients with traumatic hemorrhagic shock, yet the epidemiology and geographic distribution of patients likely to require PHBT remain poorly characterized. We sought to develop predictive models for PHBT among motor vehicle crash (MVC) patients using National Emergency Medical Services Information System (NEMSIS) data and to estimate the national distribution of patients with a high predicted probability of receiving PHBT.
METHODS: We conducted a retrospective cohort study using the 2024 and 2025 NEMSIS data. Eligible en…

OBJECTIVES: The safety and efficacy of peripartum transfusion support depends on a comprehensive risk analysis, the provision of substantiated criteria for ordering blood products, and surveillance of outcomes over time. We aimed to determine bleeding frequencies and volumes of blood loss, associated patient risk factors, and the effectiveness of our transfusion support scheme.
METHODS: Birth records of 12,255 pregnant women who gave birth at our hospital were screened for haemorrhage volumes, mode of delivery, bleeding aetiology, accompanying risk factors as well as outcome. Transfusion rela

Alloantibodies, which can arise naturally in early life or after exposure to allogeneic red blood cells (RBCs), can pose a significant risk to blood transfusion. When patients with alloantibodies receive incompatible RBCs, they may experience potentially fatal hemolytic transfusion reactions (HTRs). Transfusion services work to prevent such reactions by identifying compatible blood; however, complications can still occur, especially in patients with complex alloantibody profiles. Given the consequences of these reactions clinically, various preclinical models have been developed to study incom…

OBJECTIVE: Major trauma is associated with blood loss and hypothermia. It is common to replace lost fluid with red cells stored at 2-6°C, and/or colloid/crystalloid fluid stored at ambient temperature, thus increasing hypothermia risk. At trauma and medical retrieval sites, mains electricity powered fluid warmers cannot be generally used. Latent heat provides an alternate practical method of portable temperature-controlled intravenous fluid warming. This work investigates the safety and efficacy of a fluid warmer powered by latent heat.
METHODS: Twenty-five haematology patients received red c

OBJECTIVES: To study the rational use of the blood components in pediatric and neonatal wards.
METHODS: It was a retrospective study conducted by department of pediatrics of a tertiary care centre in western part of India. The patients were included from the pediatric ward, pediatric surgery ward, neonatal intensive care unit (NICU), pediatric intensive care unit (PICU) over a period of three months. All the patients below 12 y of age receiving blood components, admitted in general pediatric wards, pediatric intensive care unit, neonatal intensive care unit and pediatric surgery ward were inc
