Reversal of T cell exhaustion enables cancer vaccine efficacy in BRCA1-deficient ovarian cancer.

BRCA mutations in ovarian cancer (OC) are associated with increased tumor-infiltrating lymphocytes and inflammatory features, yet immunotherapy for OC has shown limited efficacy. Using the ID8 mouse model, we discovered that Brca1 deficiency-but not Brca2-impairs the effectiveness of a dendritic cell-based cancer vaccine (OCDC). OCDC vaccine reduced tumor growth and improved survival in mice bearing Trp53 -/- and Trp53 -/- Brca2 -/- tumors but was ineffective against Trp53 -/- Brca1 -/- tumors. Transcriptomic analysis revealed metabolic and immunologic reprogramming in OCDC-responsive tumors,

Combined prostate cancer vaccine plus immune checkpoint inhibition synergizes to eliminate prostate cancer.

Immunotherapy has improved outcomes in many cancers, yet the clinical benefits remain limited in prostate cancer. We evaluated whether an adenovirus-based bivalent prostate cancer vaccine (Ad-PS2) targeting two tumor antigens could be strengthened by combination with immune checkpoint blockade. Using immunocompetent mouse models, we found that Ad-PS2 combined with low-dose anti-CTLA4 generated robust anti-tumor immunity capable of eliminating established tumors, exceeding the effects of either treatment alone. Tumor-free mice resisted subsequent tumor rechallenge, indicating durable immune pro

Cancer Prevention in Women with Sickle Cell Disease: A Scoping Review of HPV Vaccination and Cancer Screening.

BACKGROUND: Advances in sickle cell disease (SCD) care have substantially improved survival. This resulted in a growing population of adult women for whom cancer prevention is increasingly relevant. Female-specific cancer prevention strategies, including human papillomavirus (HPV) vaccination, cervical cancer screening, and breast cancer screening, are well-established in the general population. However, their implementation among women with SCD remains poorly characterized. METHODS: We conducted a scoping review of PubMed and Embase from inception through January 2026 to identify studies, ev

WT1 Trio Peptide-Based Cancer Vaccine for Rare Cancers Expressing Shared Target WT1.

No standard treatment has been established for most rare cancers. Here, we report a clinical trial of a biweekly WT1 tri-peptide-based vaccine for recurrent or advanced rare cancers. Due to the insufficient number of patients available for a traditional clinical trial, the trial was designed for rare cancers expressing shared target molecule WT1. The recruitment criteria included WT1-expressing tumors as well as HLA-A*24:02 or 02:01. The primary endpoints were immunoglobulin G (IgG) antibody (Ab) production against the WT1-235 cytotoxic T lymphocyte (CTL) epitope and delayed-type hypersensitiv

STING ligand c-di-GMP improves cancer vaccination against metastatic breast cancer.

Cancer vaccination may be our best and most benign option for preventing or treating metastatic cancer. However, breakthroughs are hampered by immune suppression in the tumor microenvironment. In this study, we analyzed whether cyclic diguanylate (c-di-GMP), a ligand for stimulator of interferon genes (STING), could overcome immune suppression and improve vaccination against metastatic breast cancer. Mice with metastatic breast cancer (4T1 model) were therapeutically immunized with an attenuated Listeria monocytogenes (LM)-based vaccine, expressing tumor-associated antigen Mage-b (LM-Mb), foll

Development of the rationale of a personalized cancer vaccine based on the in situ vaccine effect of radiotherapy: a mechanistic study of the POLARSTAR trial.

PURPOSE: Radiotherapy induces multiple forms of tumor cell death, including immunogenic cell death (ICD) like GSDME-mediated pyroptosis and MLKL-mediated necroptosis; and ICD has been increasingly accepted as a crucial element leading to enhanced anti-tumor adaptive immunity. We aim to clarify whether a vaccine-like effect is intrinsic for radiation-induced tumor cell death, and to explore potential applications. METHODS: A randomized controlled trial was performed to validate the synergism between clinical radiotherapy and PD1 blockade in patients with resectable rectal cancer. Colorectal ca

Immunobiology of Newcastle Disease Virus and Its Use for Prophylactic Vaccination in Poultry and as Adjuvant for Therapeutic Vaccination in Cancer Patients.

Newcastle disease (ND) is one of the most important diseases of poultry worldwide. In the last decades, molecular research has gained a lot of new information about its causative agent, newcastledisease virus (NDV). In poultry industry, certain strains of NDV have been used for preventive vaccination for more than 60 years. NDV has also been applied to cancer patients with beneficial effects for about 50 years, but this is less well known. The molecular basis for these differential effects of NDV in birds and man have been elucidated in the last decades and are explained in this review. The an

Attitudes of Patients with Cancer towards Vaccinations-Results of Online Survey with Special Focus on the Vaccination against COVID-19.

Recently developed COVID-19 vaccines significantly reduce the risk of severe coronavirus disease, which is essential in the particularly vulnerable cancer patient population. There is a growing anti-vaccine concern that may affect the success of the fight against the SARS-CoV2 pandemic. To evaluate opinions and attitudes toward vaccination, we conducted an anonymous online survey among Polish patients diagnosed with cancer. We analyzed how socio-demographic factors, type of cancer, comorbidities, previous influenza vaccinations, and information sources affect the general willingness and opinio

The last ten years with inosine pranobex - from an "old" therapeutic agent to vaccine research, including anti-cancer vaccines.

Inosine pranobex (IP), also known as inosine pranobex dimepranol, is an immunomodulatory drug with a history spanning more than fifty years. It was first introduced in the 1970s and has since been licensed in more than 50 countries around the world. It was originally considered a potential drug for AIDS, which raised high hopes at the time of the discovery of the HIV virus. However, after initial interest, its use in this area declined, and for a long time, IP was no longer discussed significantly in professional literature or clinical practice. Renewed interest came only in the last decade, w

Usefulness of Docking and Molecular Dynamics in Selecting Tumor Neoantigens to Design Personalized Cancer Vaccines: A Proof of Concept.

Personalized cancer vaccines based on neoantigens are a new and promising treatment for cancer; however, there are still multiple unresolved challenges to using this type of immunotherapy. Among these, the effective identification of immunogenic neoantigens stands out, since the in silico tools used generate a significant portion of false positives. Inclusion of molecular simulation techniques can refine the results these tools produce. In this work, we explored docking and molecular dynamics to study the association between the stability of peptide-HLA complexes and their immunogenicity, usin…

The effect of adjuvanting cancer vaccines with herpes simplex virus glycoprotein D on melanoma-driven CD8+ T cell exhaustion.

Two vaccines expressing CD4(+) and CD8(+) T cell epitopes of melanoma-associated Ags (MAAs) by a chimpanzee-derived replication-defective AdC68 vector were compared in a mouse model of melanoma. In one vaccine, termed AdC68-gDMelapoly, the epitopes were expressed as a fusion protein within HSV-1 glycoprotein D (gD), which blocks immunoinhibitory signaling through the herpes virus entry mediator pathway. The other vaccine, termed AdC68-Melapoly, expressed only the MAA epitopes. AdC68-gDMelapoly induced more potent MAA-specific CD8(+) T cell responses especially to the subdominant MAA epitopes.

Clinical efficacy and challenges of cancer-testis antigen vaccines in advanced solid tumors: A systematic review and meta-analysis.

Cancer-testis antigens (CTA) vaccines are being actively explored for advanced solid tumors, yet their clinical value remains uncertain. We conducted a systematic review and meta-analysis to December 21, 2024, synthesizing single-arm outcomes (objective response rate [ORR], disease control rate [DCR]) and randomized evidence for overall survival (OS) and disease-free survival (DFS) using random-effects models. Across 73 single-arm trials (n = 1435), the pooled ORR was 13.0 % (95 % CI 11.0-16.0; I²=32.7 %) and the DCR was 39.0 % (95 % CI 34.0-44.0; I²=6.5 %); higher ORRs were seen in melanoma (

Supramolecular Lipid Nanoparticles Based on Host-Guest Recognition: A New Generation Delivery System of mRNA Vaccines For Cancer Immunotherapy.

Dendritic cell (DC) maturation is a crucial process for antigen presentation and the initiation of T cell-mediated immune responses. Toll-like receptors play pivotal roles in stimulating DC maturation and promoting antigen presentation. Here, a novel message RNA (mRNA) cancer vaccine is reported that boosts antitumor efficacy by codelivering an mRNA encoding tumor antigen and a TLR7/8 agonist (R848) to DC using supramolecular lipid nanoparticles (SMLNP) as a delivery platform, in which a new ionizable lipid (N2-3L) remarkably enhances the translation efficiency of mRNA and a β-cyclodextrin (β-

Expanding the Potential of Neoantigen Vaccines: Harnessing Bacille Calmette-Guérin Cell-Wall-Based Nanoscale Adjuvants for Enhanced Cancer Immunotherapy.

Personalized antitumor immunotherapy utilizing neoantigen vaccines holds great promise. However, the limited immunogenicity of existing recognized neoantigens and the inadequate stimulation of antitumor immune responses by conventional adjuvants pose significant challenges. To address these limitations, we developed a nanovaccine that combines a BCG bacterial cell wall skeleton (BCG-CWS) based nanoscale adjuvant (BCNA) with peptide neoantigens (M27 and M30). This integrated approach provides an efficient translational strategy for cancer immunotherapy. The BCNA nanovaccine, formulated with PLG
