Protection of cartilage in chemical synovectomy by perfusion.

The authors examined the effect of osmium tetroxide (OsO4) on the synovial membrane and cartilage of the rat knee joints by light-, polarization-, and electron microscope. They found that the synovial membrane undergoes severe damage after a 10 min exposure to OsO4. Removing the OsO4 from the joint cavity by perfusion after the 10 min exposure, there was no concomitant damage to the cartilage. Based on experimental results, the authors recommend in human therapy the exposure of the joints to OsO4 for no longer than 10 min and complete removal by exhaustive perfusion.

[Alteration of cartilage by microbial agents and granulocytes].

Severe polyarthritis was induced in 42 SPF piglets by subcutaneous and intraarticular infection in one joint of the bacterium Erysipelothrix rhusiopathiae (Serotype B, strain T 28), which in its chronic stage morphologically resembles human c.P. The light and electron microscopic examination of the articular cartilage and synovial membrane reveals a parallel evolution of hyaline cartilage degeneration, and activation and proliferation of synovial lining cells. The initial cartilage alteration with demasking of collagen fibrils and focal degeneration of chondrocytes in the erysipelas model is c…

Rotation of the alar cartilage in collapsed ala.

There are various causes for collapsed ala nasi. In elder people especially, the lateral crus of the alar cartilage is frequently deformed in shape and position. Model experiments show that by rotation of the lateral crus outwards and upwards, the vestibulum is expanded and the tip of the nose raised. On the basis of these experiments, in 19 patients with collapsed ala nasi, the lateral crus of the alar cartilage was rotated dorsally. The longterm results showed a satisfactory functional and aesthetic outcome.

The problem of the collapsing upper lateral cartilage.

The collapsing upper lateral cartilage is an airway problem that admits to no ready solution, such as the deviated nasal septum or deflected columella. It is my understanding from experienced nasal surgeons that an attack directly on the upper lateral cartilage has not provided a solution, but in general makes the problem worse. A look at the nasal septal base by simultaneous viewing of both nasal chambers may reveal a dimenionsal of 1 cm or more. In that event, a fairly radical narrowing of the septal base in the plane of the pyriform crest by a subperiosteal and subperichondrial approach wit…

The role of hypertrophic cartilage in endochondral ossification.

Normal stages of histogenesis of long bones show that the hypertrophy of cartilage cells is the pre-requisite for the perichondrium to take up osteoblastic activity, (Fell 1925, Lutfi 1971). Cooper (1965) found the cartilage cells from epihysis of the long bones of chick failed to induce chondrogenesis in somites in mice and chick whereas flat cells and early Peripheral cells could do same. Fell and Landauer (1935) noticed that in avian phocomelia the hypertrophied cartilage cells fail to hypertrophy leading subsequently to deformities of long bones. Presently an attempt is made to analyse thi…

Enzyme histochemistry of bone and cartilage cells.

Initial studies indicated that bone and cartilage, when treated with a hypertonic glutaraldehyde fixative for a short period, retained significant enzyme activity for histochemistry and also maintained excellent fine structure. We used 6% glutaraldehyde in 0.1 M cacodylate buffer, pH = 7.2, 4 degrees C to fix small pieces of bone or cartilage for three hours while the tissues were being constantly agitated. These samples were demineralized in 10% ethylene diamine tetraacetic acid, buffered to pH = 7.2 with 0.1 M Tris HC1, at 4 degrees C. The demineralized tissue was frozen and cryostat section…

Ultrastructure of articular cartilage of achondroplastic mice.

Articular chondrocytes of achondroplastic mice (cn/cn) resemble ultrastructurally those of their non-achondroplastic siblings (Cn/Cn or Cn/cn), except for premature deposition of glycogen and a tendency to undergo regression. The latter may be slight or extreme. The ultimate cause of the vulnerability of the chondrocytes and the cause of the heterogeneity of the reaction could not be determined with the method employed. Nevertheless, increased vulnerability accounts for cell death under conditions which are usually not injurious, and which may be either physiologic or altered by endogenous or

Ultrastructure of articular cartilage in pyogenic arthritis.

The articular cartilage in pyogenic arthritis, induced in rabbit by intra-articular injection of Staphylococcus aureus, was studied with the electron microscope. The cartilage showed erosion of the surface and degeneration and necrosis of the chondrocytes starting from 24 hours onward, following the intra-articular injection of bacteria. The surface erosion and chrondrocyte necrosis progressed rapidly. The changes are believed to be due mainly to liberation of lysosomal enzymes from the synovial cells and polymorphonuclear leukocytes.

The influence of trauma on the growing septal cartilage.

In 34 children biopsies of the nasal septum which had formerly been traumatized by force or surgery, are histologically examined. The long-term reactions of the injured septal cartilage can be described by loss, incomplete and complete regeneration of cartilage. In most cases the regeneratioen of the cartilage is undirected, thus causing deformation of the nasal septum.

Construction of cartilaginous organoids based on cartilage extracellular matrix microcarriers to promote articular cartilage regeneration through immune regulation.

OBJECTIVE: To investigate the feasibility of constructing cartilaginous organoids (CORGs) using cartilage extracellular matrix microcarriers (CEMMs), evaluate their ectopic chondrogenic potential, and analyze their impact on in situ repair and regeneration of knee cartilage in SD rats. METHODS: Cartilage extracellular matrix microcarriers (CEMMs) were created through a combination of decellularization, wet milling, and layered sieving methods. The evaluation of their biological function was conducted through live/dead staining, CCK-8 assay, scratch assay, and Transwell assay in a laboratory s…

Combined MPFL Reconstruction with Tibial Tubercle Osteotomy and Repair of Patellar Cartilage Defect with Particulated Juvenile Articular Cartilage.

UNLABELLED: Medial patellofemoral ligament (MPFL) reconstruction with tibial tubercle osteotomy (TTO) and particulated juvenile articular cartilage (PJAC) grafting can be performed in combination for the treatment of recurrent patellar instability with associated patellar cartilaginous defects. DESCRIPTION: Preoperative planning is an essential component for this procedure. Measurement of the tibial tubercle to trochlear groove (TT-TG) distance and the Caton-Deschamps index (CDI) allows for determination of the degree of medial and anterior translation and helps to identify whether distalizat…

Repair of articular cartilage defect using adipose-derived stem cell-loaded scaffold derived from native cartilage extracellular matrix.

The purpose of this study was to investigate the feasibility of adipose-derived stem cells (ADSCs) as the seed cells of cartilage tissue engineering. ADSCs were isolated from adipose tissue that was harvested under sterile conditions from the inguen fold of porcines and cultured in vitro. Acellular cartilage extracellular matrix (ACECM) scaffolds of pigs were then constructed. Moreover, inflammatory cells, as well as cellular and humoral immune responses, were detected using hematoxylin and eosin staining staining, immunohistochemical staining, and western blot analysis. The results showed tha…
