This case report presents a 42-year-old woman with metastatic breast cancer refractory to multiple conventional treatments, including surgery, chemotherapy, radiotherapy, hormonal therapy, and bilateral oophorectomy. As a salvage, she received 177 Lu-FAPI-2286 radioligand therapy. After the first cycle, an unexpected serum β-hCG elevation was observed-unusual given her history of total hysterectomy and bilateral salpingo-oophorectomy. Three hypotheses were proposed: assay interference (heterophile antibodies), emergence of second neoplasm secreting β-hCG, and tumor dedifferentiation with ectop

We report a unique case of a 22-year-old male with a progressively enlarging mass on the right thumb. Initial biopsies suggested a low-grade osteogenic tumor. Following partial amputation, gross examination revealed a well-circumscribed 3.7cm mass involving the metacarpal bone with soft tissue invasion. Histologically, the tumor displayed features of low-grade central osteosarcoma with areas transitioning to a round blue cell morphology resembling Ewing sarcoma. Comprehensive molecular testing-including next-generation sequencing (NGS) and cytogenomic microarray analysis (CMA)-identified ampli

Despite the increasing success of interventional coronary reperfusion strategies, mortality related to acute myocardial infarction (MI) is still substantial. MI is defined as sudden death of myocardial tissue caused by an ischemic episode. Ischaemia leads to adverse remodelling in the affected myocardium, inducing metabolic and ionic perturbations at a single cell level, ultimately leading to cell death. The adult mammalian heart has limited regenerative capacity to replace lost cells. Identifying and enhancing physiological cardioprotective processes may be a promising therapy for patients wi

N-glycosylation is a dynamic post-translational modification that critically regulates cancer cell differentiation through modulating receptor signaling, cell adhesion, and plasticity. Aberrant N-glycosylation promotes dedifferentiation, drives EMT, and confers therapy resistance across malignancies. This review summarizes the role of N-glycosylation in determining lineage commitment and altering responses to differentiation therapies. Targeting the N-glycosylation apparatus can reprogram tumor cells toward differentiated phenotypes, potentiating the effects of agents such as ATRA and NaBu. Ev

Mitf encodes a basic helix-loop-helix-leucine-zipper (bHLHzip) protein that is known to function in the development of melanocytes, pigmented epithelial cells (PECs), osteoclasts, and mast cells. In this paper, we report on the isolation, expression, and overexpression of the chicken Mitf and discuss the role of its protein product in the differentiation and transdifferentiation of PECs. Northern blotting showed that chicken Mitf is predominantly expressed in embryonic retinal pigmented epithelium (PE), but is expressed at low levels in other tissues. A 5' RACE analysis revealed differences in

Physical damage to the peripheral nerves triggers Schwann cell injury response in the distal nerves in an event termed Wallerian degeneration: the Schwann cells degrade their myelin sheaths and dedifferentiate, reverting to a phenotype that supports axon regeneration and nerve repair. The molecular mechanisms regulating Schwann cell plasticity in the PNS remain to be elucidated. Using both in vivo and in vitro models for peripheral nerve injury, here we show that inhibition of p38 mitogen-activated protein kinase (MAPK) activity in mice blocks Schwann cell demyelination and dedifferentiation f

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive sarcomas arising from Schwann cells and characterized by marked cellular an … romatous neoplasms of uncertain biologic potential, suggest that MPNST progression involves Schwann cell dedifferentiation into a more primitive, stem-like state. In this review, we summarize key discoveries from single-cell characterization studies, and discuss how these findings illuminate MPNST tumorigenesis, cellular plasticity, and potential therapeutic vulnerabili …

Adrenal cortical steroidogenesis and proliferation are thought to be modulated by gap junction-mediated direct cell-cell communication of regulatory molecules between cells. Such communication is regulated by the number of gap junction channels between contacting cells, the rate at which information flows between these channels, and the rate of channel turnover. Knowledge of the factors regulating gap junction-mediated communication and the turnover process are critical to an understanding of adrenal cortical cell functions, including development, hormonal response to adrenocorticotropin, and

BACKGROUND & AIMS: Although the liver exhibits remarkable regenerative capacity, expanding hepatocytes in large quantities in vitro without compromising their function remains challenging, presumably due to the absence of key environmental signals. Identifying factors that enable long-term in vitro expansion of hepatocytes is essential for advancing liver research.
METHODS: Mouse models of hepatectomy and hepatocyte transplantation were used to identify cytokines involved in liver regeneration. Mouse and human hepatocytes were cultured and passaged in IL-22 medium to assess their expansion ab

Gastrointestinal stromal tumours (GISTs) are the most common mesenchymal neoplasms of the gastrointestinal tract. Dedifferentiation in these tumours occurs rarely, and when it does occurs most commonly after prolonged treatment with imatinib. We report the case of a 64-year-old man who presented with a mass of 8×7×3 cm dimensions involving the duodenum and head of the pancreas. On histopathology, areas of anaplastic tumour cells were negative for DOG-1, c-kit, CD-34, desmin and panCK along with a molecular level study showing wild-type KIT and PDGFRA (platelet-derived growth factor receptor al

To study disease development, an inventory of an organ's cell types and understanding of physiologic function is paramount. Here, we performed single-cell RNA-sequencing to examine heterogeneity of murine pancreatic duct cells, pancreatobiliary cells, and intrapancreatic bile duct cells. We describe an epithelial-mesenchymal transitory axis in our three pancreatic duct subpopulations and identify osteopontin as a regulator of this fate decision as well as human duct cell dedifferentiation. Our results further identify functional heterogeneity within pancreatic duct subpopulations by elucidatin…

UNLABELLED: Anaplastic thyroid cancer (ATC) is the most aggressive type of thyroid cancer with survival time of only 7 to 10 months. Previous work revealed reduced proportions and cytotoxicity of NK cells in the ATC tumor microenvironment (TME). In this study, we investigated the role of super-enhancers (SE), clusters of adjacent enhancers that drive high expression of genes, in reshaping the TME in ATC. Comprehensive profiling of the SE landscapes in ATC revealed the activation of oncogenic SEs as a mechanism underlying the dedifferentiation and anaplastic transformation of thyroid cancer. An

Well-differentiated liposarcoma (WDL) can show a morphologic spectrum, including lipoma-like, sclerosing and inflammatory subtypes. It does not metastasize but can dedifferentiate, acquiring metastatic potential. Hibernomas are benign neoplasms that show variable differentiation toward brown fat, and can sometimes occur in the abdomen or retroperitoneum. We illustrate a case of retroperitoneal WDL that showed extensive hibernoma-like morphology, with sheets of multivacuolated adipocytes of varying sizes, with abundant cytoplasm and numerous lipid vacuoles or granular eosinophilic cytoplasm. Pancreatic α cell senses amino acid availability to adjust secretion function and proliferation, yet the underlying molecular mechanisms remain unclear. Here, α cell-specific deletion of CBP/p300 in mice leads to hypoglucagonemia and hyperaminoacidemia, along with decreased functional pancreatic α cell mass due to impaired cell proliferation, dedifferentiation, and cell loss. The knockout of CBP/p300 blocks glucagon receptor antibody-stimulated α cell proliferation and mTORC1 signaling in mice. In CBP/p300-deficient α cells, single-cell RNA sequencing reveals upregulated autophagy-related gene

BACKGROUND: Metastasis to the peritoneum is a rare but aggressive manifestation of squamous cell carcinoma of the head and neck (SCCHN). The specific drivers of metastasis in SCCHN remain unclear. We describe a case of recurrent, metastatic SCCHN and explore the potential role of acquired genomic alterations on the disease course.
METHODS: Mutational profiling of serial tumor specimens was conducted. Identified mutations were compared to TCGA SCCHN data.
RESULTS: Tumor specimens showed the progressive accumulation of mutations in TP53, NOTCH1, APC, and SMAD4.
CONCLUSION: This case provides

Organ development depends on multiple signaling pathways working in concert to specify cell fates. Improper activity or inactivity of specific signaling pathways such as EGF-Ras-MAPK can lead to dedifferentiation and cancer. In C. elegans , gain of function mutations in Ras/ let-60 lead to ectopic development of multiple ventral vulva-like lesions resembling tumors. However, this phenotype depends on normal insulin-like growth factor (IGF) signaling. Here, we probe how factors downstream of the IGF receptor daf-2 modify Ras signaling. These investigations led us to identify regulators of cell

Forty cases of squamous cell carcinoma and twenty cases of adenocarcinoma of the cervix were subjected to DNA ploidy analysis to find out the frequency of aneuploid DNA pattern in different types of invasive carcinoma of the uterine cervix and their possible correlation with the clinical stage and age of the patients. From the DNA analysis an increasing trend of DNA aneuploidy was observed from well differentiated (63.6%) to moderately differentiated (70.8%) to poorly differentiated squamous cell carcinoma (83.3%). However, 85% adenocarcinomas of endocervix showed aneuploid DNA pattern. It ind

Few studies have analyzed the prevalence of TERT amplification in thyroid cancer, showing discrepancies in various topics. The impact on tumor recurrence and patient survival in papillary thyroid carcinoma (PTC) remains unknown. Thirteen cancer cell lines and 215 tumor samples from 91 patients, who underwent surgery for PTC (41), poorly differentiated thyroid carcinoma (PDC = 15), or anaplastic thyroid carcinoma (ATC = 35), were analyzed. Clonality, spread with tumor dedifferentiation or metastatic PTC cells, and coexistence with TERTp, BRAF, RAS, and PIK3CA mutations were also investigated. T
