Homeostatic serum IgE is secreted by plasma cells in the thymus and enhances mast cell survival.

Increased serum levels of immunoglobulin E (IgE) is a risk factor for various diseases, including allergy and anaphylaxis. However, the source and ontogeny of B cells producing IgE under steady state conditions are not well defined. Here, we show plasma cells that develop in the thymus and potently secrete IgE and other immunoglobulins, including IgM, IgA, and IgG. The development of these IgE-secreting plasma cells are induced by IL-4 produced by invariant Natural Killer T cells, independent of CD1d-mediated interaction. Single-cell transcriptomics suggest the developmental landscape of thymi

Recombinant Arabidopsis HSP70 Sustains Cell Survival and Metastatic Potential of Breast Cancer Cells.

The chaperone HSP70 protein is widely present in many different tumors and its expression correlates with an increased cell survival, low differentiation, and poor therapeutic outcome in human breast cancer. The intracellular protein has prevalently a cytoprotective function, while the extracellular HSP70 mediates immunologic responses. Evolutionarily, HSPs are well conserved from prokaryotes to eukaryotes, and human HSP70 shows a strong similarity to that of plant origin. In the current article, we have tested the potential effect of recombinant HSP70, from Arabidopsis thaliana, on cell survi…

Serotonin activates cell survival and apoptotic death responses in cultured epithelial thyroid cells.

Anatomic and physiological interactions between central serotonergic system and thyroid gland are well established. However, the effects of locally available serotonin on the thyroid functions are poorly known. Here, we first demonstrate the expression of serotonin transporter SERT and 5-HT2A receptor subtype in rat thyroid epithelial cell line FRT both at mRNA and protein levels. In order to investigate the molecular mechanisms of serotonin action, FRT cells were exposed to increasing concentrations of the amine. Low concentrations of serotonin (up to 5 μM) enhanced FRT cell growth, and ERK1/

Endogenous polysialylated neural cell adhesion molecule enhances the survival of retinal ganglion cells.

PURPOSE: During development, all retinal cells express polysialylated neural cell adhesion molecule (PSA-NCAM). PSA is localized only on glia in the adult retina, but as Müller glial processes ensheathe most retinal neurons, PSA remains in the extracellular environment of adult neurons. The authors sought to investigate the influence of endogenous PSA on the survival of neonatal as well as adult normal and injured retinal ganglion cells (RGCs). METHODS: Endogenous retinal PSA was selectively degraded by application of endoneuraminidase. PSA presence and removal were confirmed by immunohistoch

USP7 inhibits FOXO1 activity in chronic lymphocytic leukaemia cells enhancing cell survival.

Signals emanating from the B cell receptor (BCR) are vital for driving cell survival and proliferation in chronic lymphocytic leukaemia (CLL). BCR-mediated signals in CLL cells promote nuclear exclusion and inactivation of the transcription factor forkhead box protein O1 (FOXO1) and transient upregulation of FOXO1 expression. To investigate the mechanisms that regulate FOXO1 inactivation, we focused on the role of deubiquitinase (DUB) family proteins. Stratifying expression of individual DUB proteins, we established that ubiquitin-specific protease 7 (USP7), USP8, USP9x, USP10 and USP14 are si…

A ketogenesis-ferroptosis axis maintains leukemic stem cell survival and leukemia progression.

Hepatic ketogenesis generates ketone bodies as an alternative energy source during carbohydrate restriction or ketogenic diets, yet its role in non-hepatic cell types remains poorly defined. Here, we show that leukemic stem cells (LSCs) in acute myeloid leukemia (AML) exhibit elevated ketogenesis, driven by fatty acid oxidation (FAO), to produce β-hydroxybutyrate (BHB). LSCs express high levels of 3-hydroxy-3-methylglutaryl-coenzyme A (CoA) synthase 2 (HMGCS2), the rate-limiting enzyme in ketogenesis, compared with blast cells and normal hematopoietic stem cells (HSCs). Deletion of Hmgcs2 in A

Salicylic acid fine-tunes cell survival during plant senescence and pathogen infection.

Salicylic acid (SA) is a crucial plant hormone synthesized via two primary pathways: the ISOCHORISMATE SYNTHASE (ICS) pathway and the PHENYLALANINE AMMONIA-LYASE (PAL) pathway. SA biosynthesis and homeostasis are precisely regulated in plants. Furthermore, SA functions in a concentration-dependent manner to enhance resistance to both abiotic stresses and pathogens, as well as to regulate plant senescence. However, the underlying mechanisms of SA action remain to be elucidated. Recent studies have demonstrated that in juvenile plants, age-related cues, autoimmunity, and autophagy contribute to

Human and mouse regenerative macrophages enhance beta cell survival, function, and proliferation.

AIMS: Type 1 diabetes is caused by immune-mediated destruction of beta cells. Interestingly, individuals with long-standing type 1 diabetes have residual beta cells, suggesting regenerative mechanisms may help beta cell survival. Islet-resident macrophages have an important role in diabetes, and can adopt a tissue-regenerating phenotype that may support beta cells. However, the roles of macrophages in beta cell survival, function, and proliferation remain poorly defined. This study aimed to elucidate how macrophage subtypes influence beta cell survival, function, and proliferation. METHODS: M…

Retinal ganglion cell autophagy: a housekeeper essential for neuronal homeostasis and survival.

Retinal ganglion cells (RGCs) are the sole projection neurons of the retina and the only direct link between retinal circuitry and the brain. Maintaining this lifelong connection requires constitutive autophagy to preserve organelle quality control and neuronal homeostasis. Although autophagy has been widely studied following ocular hypertension and optic nerve injury, its physiological role in healthy RGCs has remained unclear. We have recently revealed that basal autophagy is highly active in RGCs and that conditional deletion of Atg5 or Atg7 is sufficient to induce progressive RGC dysfuncti

PHPT1/Ocnus negatively regulates the JAK/STAT pathway to control cell survival.

The JAK/STAT signaling pathway is a highly conserved regulator essential for animal development. Here, we show that the Drosophila phosphohistidine phosphatase 1, PHPT1/Ocnus, is required for cell survival by regulating JAK/STAT signaling. Knockdown of ocnus (ocn) leads to apoptosis of germ cells during the transit-amplifying divisions of spermatogonia in the second instar larvae. In Drosophila S2 cells, ocn overexpression reduces phosphorylated STAT (pSTAT) levels, limits its nuclear accumulation, and alters the expression of STAT target genes. Mechanistically, Ocn directly dephosphorylates S…

Improved Survival of HPV-Related Sinonasal Squamous Cell Carcinoma With Chemoradiation Treatment.

OBJECTIVE: Human papillomavirus-related (HPV+) sinonasal squamous cell carcinoma (SNSCC) is associated with improved overall survival (OS) compared to similarly staged HPV-negative SNSCC. However, data on treatment-specific outcomes remain limited. METHODS: We analyzed the National Cancer Database (NCDB) for all SNSCC cases diagnosed between 2010 and 2017 with available HPV testing data. Prognostically significant variables were identified via multivariable Cox regression. Propensity score-matched cohorts were then created, matching HPV-positive and HPV-negative patients exactly by treatment

Glioblastoma invasion of neural stem cell regions; molecular patterns and survival rates.

Glioblastoma is an aggressive form of brain cancer and poses a challenge in treatment due to its profound heterogeneity and capacity for extensive infiltration into the brain parenchyma. Research has shown glioblastomas near the ependyma have poorer survival rates. Therefore, our aim was to identify distinct molecular features of glioblastoma invading the ependyma of lateral ventricles and neural stem cell region and the survival prognosis of these patients. A retrospective review of 170 patients with a new histologically confirmed diagnosis of glioblastoma between 2018 and 2019. Patients were

Murine T Cell-Dependent Response Affects Leishmania amazonensis Amastigote Survival and Virulence.

Our group has previously described the infection course of Leishmania amazonensis in BALB/c and BALB/cnu/nu mice, which have distinct T-cell dependent responses. In this study, lesion-derived amastigotes exhibited differential protein expression in a host-dependent manner. Herein, we evaluate the impact of the immune system of these two mouse strains on parasite survival and virulence. For this, BALB/c and BALB/cnu/nu mice were infected with promastigotes of the LV79 strain, and after 13-weeks of infection, amastigotes were isolated from the lesion footpads. The amastigotes recovered from BALB

Critical Role of RPS4X in Modulating SCF Complex Formation and Cell Survival.

Ribosomal proteins have long been recognized as vital components of ribosomes that are involved in protein synthesis. However, emerging evidence indicates that some ribosomal proteins exhibit extraribosomal functions. In this study, we investigated the role of the ribosomal protein S4 X-linked (RPS4X) in the regulation of the Skp1-Cullin1-F-box (SCF) ubiquitin ligase complex and apoptosis. We found that RPS4X expression interfered with SCF complex formation by disrupting the interaction between Cullin1 and Skp1. This disruption suppressed ubiquitination of multiple SCF complex substrates, incl

Knockout of SIN3B modulates transcriptional programs and cell survival in cutaneous melanoma.

SIN3 is a critical component of the histone deacetylase complex. Utilizing whole transcriptome data from melanoma patient samples we reveal that elevated levels of SIN3B are associated with poor survival outcomes with in vitro studies showing increased SIN3B expression in BRAF-mutant metastatic melanoma cell lines. The generation of isogenic SIN3B knockout cell lines indicated that SIN3B disruption led to a decrease in pathways associated with tumor invasion, migration, and cell-cell interactions. Moreover, pooled genome-wide CRISPR/Cas9 screens highlighted POLE4 and STK11 as crucial for the f

Creatine kinase B promotes non-small cell lung cancer survival and metastasis.

Creatine kinase (CK) catalyzes the reversible transfer of a phosphoryl group from ATP to creatine. There are four distinct CK genes (CKM [muscle-type CK], CKB [brain-type CK], CKMT1 [ubiquitous-type CK], and CKMT2 [sarcomeric-type CK]) with cell-type selective expression and subcellular localization. In cancer, uncontrolled cell proliferation drives aggressive migration and invasion into nearby tissues and distant organs. While creatine metabolism is known to support cancer cell survival, the specific roles of individual CK isoenzymes remain unclear. Here, we demonstrate that CKB is essential …

Th7R predicts chemo-immunotherapy response and survival in small cell lung cancer.

Th7R is a novel Th1-like CD4+ T cell lineage characterized by TCF7 and IL-7 receptor expression, exhibiting distinct transcriptional and epigenetic profiles compared with classical Th1 cells. The clinical significance of Th7R, including the efficacy of immune checkpoint inhibitors and the prediction of postoperative disease-free survival, has been demonstrated in non-small cell lung cancer (NSCLC) but not in small cell lung cancer (SCLC). In this study, we investigated Th7R expression as a predictive marker in patients with extensive-stage SCLC (ES-SCLC) who received chemotherapy or chemo-immu
