Cellular senescence as a response to multiwalled carbon nanotube (MWCNT) exposure in human mesothelial cells.

Cellular senescence is a stable cell cycle arrest induced by diverse triggers, including replicative exhaustion, DNA damaging agents, oncogene activation, oxidative stress, and chromatin disruption. With important roles in aging and tumor suppression, cellular senescence has been implicated also in tumor promotion. Here we show that certain multiwalled carbon nanotubes (MWCNTs), as fiber-like nanomaterials, can trigger cellular senescence in primary human mesothelial cells. Using in vitro approaches, we found manifestation of several markers of cellular senescence, especially after exposure to…

Effect of cellular and ECM aging on human iPSC-derived cardiomyocyte performance, maturity and senescence.

Cardiovascular diseases are the leading cause of death worldwide and their occurrence is highly associated with age. However, lack of knowledge in cardiac tissue aging is a major roadblock in devising novel therapies. Here, we studied the effects of cell and cardiac extracellular matrix (ECM) aging on the induced pluripotent stem cell (iPSC)-derived cardiomyocyte cell state, function, as well as response to myocardial infarction (MI)-mimicking stress conditions in vitro. Within 3-weeks, young ECM promoted proliferation and drug responsiveness in young cells, and induced cell cycle re-entry, an

Premature lung aging and cellular senescence in the pathogenesis of idiopathic pulmonary fibrosis and COPD/emphysema.

Different anatomic and physiological changes occur in the lung of aging people that can affect pulmonary functions, and different pulmonary … ndition (inflammaging). According to recently proposed pathogenic models in COPD and IPF, premature cellular senescence likely affects distinct progenitors cells (mesenchymal stem cells in COPD, alveolar epithelial precursors in IPF), leading to stem cell exhaustion. In this review, the large amount of data supporting this pathogenic view are discussed, with emphasis …

Cellular senescence and regulated cell death in cancer: mechanisms, cross-regulatory networks and therapeutic implications.

Cellular senescence and regulated cell death represent two fundamental but intricately connected biological programs that govern tumor cell fate. Cellular senescence can suppress tumor growth by inducing stable cell-cycle arrest and promoting immune clearance, but persistent senescent cells may also drive tumor progression through the senescence-associated secretory phenotype (SASP). Concurrently, multiple modalities of regulated cell death-including apoptosis, ferroptosis, cuproptosis, necroptosis, pyroptosis, parthanatos, and autophagy-dependent cell death-have emerged as critical determinan…

Ion-coupled transfersome complexes for enhanced transdermal NAD+ repletion and mitigation of cellular senescence signatures.

Nicotinamide adenine dinucleotide (NAD+) is a pivotal coenzyme whose decline drives mitochondrial dysfunction and senescence. However, NAD+ delivery to skin is limited by poor stability and tissue accessibility. Here, we develop an ion-coupled NAD+ transfersome (ICoN) that stabilizes NAD+ and enables efficient, non-invasive transdermal delivery. ICoN restores intracellular NAD+ levels and reverses transcriptomic programs associated with DNA damage, oxidative stress, and senescence in vitro. Functionally, ICoN promotes mitochondrial functional integrity and sustains proliferative and migratory

LaminA/C-dependent cellular senescence signaling promotes skeletal muscle atrophy and abnormalities in Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disease affecting the central nervous system with effects on the skeletal muscle that entail … ciated pathways led to reduced levels of the nuclear membrane protein LaminA/C, causing accelerated cellular senescence in the A53T muscle. We identify a molecular pathway of senescence-associated secretory phenotype activating FoxO signaling, resulting in skeletal muscle loss in the A53T mice. Thus, increased oxidative stress and accumulated cellular senescence coul …

ABCC6 in Cellular Metabolic Homeostasis: Biochemical Links to Extracellular Nucleotide Signaling, Mitochondrial Stress, and Senescence.

ABCC6 (ATP-binding cassette subfamily C member 6) was initially identified as the disease gene for pseudoxanthoma elasticum, a disorder marked by ectopic mineralization. Beyond this classical role, emerging evidence supports ABCC6 as a liver-enriched transporter with broader metabolic relevance. ABCC6 promotes hepatocyte ATP (adenosine triphosphate)/ADP (adenosine diphosphate) release and supports ENPP1 (ectonucleotide pyrophosphatase/phosphodiesterase 1)-dependent PPi (inorganic pyrophosphate) generation, thereby maintaining circulating anti-mineralization buffering; downstream AMP (adenosine

Multi-omics and experimental validation studies reveal key biomarkers of cellular senescence in diabetic retinopathy.

OBJECTIVE: Diabetic retinopathy (DR) is a prevalent microvascular complication of diabetes, contributing to vision impairment and related retinal diseases. Growing evidence indicates that cellular senescence (CS) under high-glucose conditions plays a role in the pathogenesis of DR. This study aims to identify key biomarkers of CS in DR by integrating transcriptomics, single-cell sequencing data, and experimental validation, thereby offering insights for understanding the disease mechanism and developing novel therapeutic strategies. METHODS: DR-related datasets and CS-related genes (CSRGs) wer…

Mechanistic links between obstructive sleep apnea, cellular senescence and aging: The role of cardiometabolic dysfunction.

Obstructive sleep apnea (OSA) is a common condition characterized by repeated airway obstruction during sleep, triggering physiological changes that resemble key features of cellular senescence-oxidative stress, mitochondrial dysfunction, and systemic inflammation. While senescence plays roles in tissue repair, its accumulation also drives chronic inflammation and immune dysregulation. The connection between OSA, senescence, and aging remains complex and underexplored. Mechanisms like the senescence-associated secretory phenotype (SASP), impaired immune surveillance, and reduced regenerative c…

Targeting cellular senescence in dystrophin-/-/utrophin-/-double knockout mice improves musculoskeletal health and increases lifespan.

Duchenne muscular dystrophy (DMD) is a severe genetic muscle disease caused by mutations of the dystrophin gene. Previous studies have detected senescent cells in the skeletal muscle of human DMD, dystrophin-deficient mice (Mdx), and rats. This study aimed to use a more severe dystrophin-/-/utrophin-/- (dKO-Hom) mouse model to identify which cells become senescent and if targeting cellular senescence can improve bone quality and muscle pathology in dKO-Hom mice. Immunohistochemistry of P21 and GLB1 revealed significantly more senescent cells in the skeletal muscle tissues of 4-week-old Mdx and…

Progressive Blood-Brain Barrier Disruption in Sleep-Restricted Young Mice: Cellular Senescence and Neuroinflammation Crosstalk.

Sleep loss promotes a chronic low-grade inflammatory status with increased levels of inflammatory cytokines. Sleep loss also induces low-grade neuroinflammation characterized by glial reactivity and blood-brain barrier (BBB) dysfunction, as evidenced by BBB hyperpermeability and tight junction disassembly. Additionally, it raises molecules related to the senescence-associated secretory phenotype (SASP) in aged subjects, suggesting an increase in senescent cells. Here, we assessed the impact of sleep restriction on cellular senescence, neuroinflammation, and BBB function in the cerebral cortex …

Bisphenol A induces cellular senescence in SW1353 chondrocytes via miRNA-449a-mediated downregulation of SIRT1.

Bisphenol A (BPA) is reportedly linked to multiple human diseases. Recent studies found that it disturbs cartilage development and homeostasis. However, the exact molecular mechanisms are mostly unknown. This study aimed to elucidate how BPA impacts chondrocyte senescence and the regulatory molecular mechanism involved. After exposing SW1353 cells to 2.5, 5 or 10 μM BPA for 72 h, we found that 5 and 10 μM BPA notably increased the percentage of senescence-associated β-galactosidase-positive cells by 34.7 % and 45.3 %, respectively. The expression of senescence-associated proteins p53, p21 and

o-Vanillin enhances bone health and fracture healing by inhibiting cellular senescence in aging mice.

Aging is associated with skeletal fragility driven by impaired bone remodeling, increased oxidative stress, and the accumulation of senescent cells. To determine whether ortho-vanillin (o-Vanillin) can alleviate age-related deficits in bone, we examined femoral bone microarchitecture, biomechanical properties, bone turnover, bone marrow adiposity, oxidative stress, DNA damage, osteocyte senescence, and femoral fracture healing in C57BL/6 J mice. DEXA was additionally used to assess bone mineral density and content at the femur, tibia, spine, and whole body. Aged mice displayed substantial dete

Cellular senescence-Related genes define the immune microenvironment and molecular characteristics in severe asthma patients.

Recent studies have shown that cellular senescence is involved in the pathogenesis of severe asthma (SA). The objective of this study was to investigate the role of cellular senescence-related genes (CSGs) in the pathogenesis of SA. Here, 54 differentially expressed CSGs were identified in SA patients compared to healthy control individuals. Among the 54 differentially expressed CSGs, 3 CSGs (ETS2, ETS1 and AURKA) were screened using the LASSO regression analysis and logistic regression analysis to establish the CSG-based prediction model to predict severe asthma. Moreover, we found that the p…

Persistent elevation of plasma markers of cellular senescence after hip fracture: a pilot longitudinal study.

INTRODUCTION: Hip fractures may result from and contribute to accelerated biological aging. We aimed to evaluate the impact of hip fracture and its surgery on the senescence-associated secretory phenotype (SASP) index, a composite of peripheral protein markers where higher scores are thought to indicate greater levels of cellular senescence and accelerated aging. METHODS: We examined the SASP index in plasma over 12 weeks post-surgery and its prediction of long-term post-surgical functional outcomes. We included 60 older adults: 20 recruited immediately after hip fracture surgery, and 40 comp…

Cellular senescence promotes meibomian gland dysfunction in a chronic graft-versus-host disease mouse model.

PURPOSE: Aging is a well-established risk factor for meibomian gland dysfunction (MGD). We previously reported an accelerated cellular senescence phenomenon in the lacrimal glands of a murine model of chronic graft-versus-host disease (cGVHD). Herein, we aimed to elucidate the relationship between cellular senescence and MGD in cGVHD mice, utilizing the senolytic agent ABT-263. METHODS: A cGVHD mouse model was established through allogeneic bone marrow transplantation (BMT) from B10.D2 to BALB/c mice. Subsequently, cGVHD mice were treated with either ABT-263 or vehicle. The eyelids of recipie…

Genes associated with cellular senescence favor melanoma prognosis by stimulating immune responses in tumor microenvironment.

PURPOSE: Skin cutaneous melanoma (SKCM), a malignant tumor from melanocytes, is the fifth most prevalent tumor. Immune checkpoint inhibitor (ICI) immunotherapy improves prognosis of SKCM, but immune response varies for different populations. Cellular senescence in the tumor microenvironment (TME) promotes antitumor immunity, mediated by dendritic cells (DC) and CD8+ T cells. Therefore, we sought to explore the role of cellular senescence in the TME of SKCM through bioinformatics and machine learning. METHODS: First, we obtained 93 cellular senescence-prognosis genes (CSPGs) by univariate surv…

CLIC1 Inhibition Protects Against Cellular Senescence and Endothelial Dysfunction Via the Nrf2/HO-1 Pathway.

Chloride intracellular channel 1 (CLIC1) is a sensor of oxidative stress in endothelial cells (EC). However, the mechanism by which CLIC1 mediate the regulation of endothelial dysfunction has not been established. In this study, overexpressed CLIC1 impaired the ability of the vascular cells to resist oxidative damage and promoted cellular senescence. Besides, suppressed CLIC1 protected against cellular senescence and dysfunction in Human Umbilical Vein Endothelial Cells (HUVECs) through the Nrf2/HO-1 pathway. We also found that ROS-activated CLIC1-induced oxidative stress in HUVECs. Nrf2 nucle…
