OBJECTIVE: The objective of this study was to compare reproductive performance parameters for two protocols for estrus synchronization in dairy heifers in Germany.
MATERIAL AND METHODS: In the CIDR group (n=93) all heifers received a controlled intravaginal progesterone releasing insert (Eazi-Breed™ CIDR®; Pfizer Pharma GmbH; containing 1,38g of progesterone) on day 0. On day 7 these cows were given prostaglandin F2α (PGF2α) analogue cloprostenol (Estrumate®, Intervet Deutschland GmbH, 0.5mg per animal i.m.), and the CIDR® insert was removed. Over the last 2 years, data on cervical mucus, thawed semen and their interaction in vitro have been collected at the time of insemination in all patients referred to our clinic for artificial insemination by donor (AID). Six women with normal menstrual cycles were treated over six cycles with Fysioquens, a normophasic contraceptive consisting of 7 tablets of ethinyloestradiol (EE) 0-050 mg and 15 tablets containing 1-0 mg lynestrenol + 0-050 mg EE. Secretory leukocyte protease inhibitor (SLPI) is a potent inhibitor of human leukocyte elastase. SLPI transcripts in the cervical tissue were detected during the menstrual cycle by reverse transcription-polymerase chain reaction (RT-PCR). Western blot analysis revealed that the intensity of SLPI protein in cervical tissue in the ovulatory phase was stronger than in other phases. Immunohistochemistry using an anti-SLPI polyclonal antibody revealed positive staining in the epithelial cells of the endocervix. Western blot analysis also revealed that SLPI protein was present in the cervical mucus.

Fungi have already been described as etiological agents of reproductive diseases such as endometritis and infertility in cows. However, few studies have been developed to elucidate the entire cervicovaginal fungal communities in cows.
