Quantum dot-Eu3+ conjugate as a luminescence turn-on sensor for ultrasensitive detection of nucleoside triphosphates.

We report a conjugate of thioglycolic acid (TGA) capped CdTe quantum dot and Eu(3+) ion (TGA-CdTe QD-Eu(3+)) that can be used as an ultrasensitive luminescence turn-on sensor for nucleoside triphosphates (NTPs). The TGA-CdTe QD-Eu(3+) conjugate is a weakly luminescent species as a result of the strong quenching effect of Eu(3+) ion on the luminescence of TGA-CdTe QDs. The conjugate's luminescence can be readily restored by its reaction with adenosine triphosphate (ATP) and other NTPs, and thus gives an ultrasensitive detection of NTPs, with a detection limit of 2 nM. The sensing mechanism has

Label-free fluorescent detection of ions, proteins, and small molecules using structure-switching aptamers, SYBR Gold, and exonuclease I.

We have demonstrated a label-free sensing strategy employing structure-switching aptamers (SSAs), SYBR Gold, and exonuclease I to detect a broad range of targets including inorganic ions, proteins, and small molecules. This nearly universal biosensor approach is based on the observation that SSAs at binding state with their targets, which fold into secondary structures such as quadruplex structure or Y shape structure, show more resistance to nuclease digestion than SSAs at unfolded states. The amount of aptamer left after nuclease reaction is proportional to the concentrations of the targets

Layer-by-layer assembled Fe3O4@C@CdTe core/shell microspheres as separable luminescent probe for sensitive sensing of Cu2+ ions.

A novel multifunctional microsphere with a fluorescent CdTe quantum dots (QDs) shell and a magnetic core (Fe(3)O(4)) has been successfully developed and prepared by a combination of the hydrothermal method and layer-by-layer (LBL) self-assembly technique. The resulting fluorescent Fe(3)O(4)@C@CdTe core/shell microspheres are utilized as a chemosensor for ultrasensitive Cu(2+) ion detection. The fluorescence of the obtained chemosensor could be quenched effectively by Cu(2+) ions. The quenching mechanism was studied and the results showed the existence of both static and dynamic quenching proce

Terbium-macrocycle complexes as chemical sensors: detection of an aspirin metabolite in urine using a salicylurate-specific receptor site.

Salicylurate (SU) is the major metabolite in urine of acetylsalicylic acid (aspirin) and can be used as a metric to monitor aspirin pharmacokinetics and as an indicator of appendicitis, anemia, and liver disease. Detection in urine and plasma currently requires solvent extraction or other sample handling prior to analysis. We present a simple method to quantify SU in urine via chelation to a terbium binary complex with the macrocycle 1,4,7,10-tetraazacyclododecane-1,7-bisacetate (DO2A). Binding of SU to form the [Tb(DO2A)(SU)](-) ternary complex triggers intense luminescence under UV excitatio

Determination of Cr(VI) concentration in diluted samples based on the paper test strip method.

At present, there are many methods for Cr(VI) concentration determination in diluted samples, however, these methods have their own inherent drawbacks, such as long response time, complicated pretreatment, expensive equipment etc. In this paper, a paper-based test strip method specific for determination of diluted Cr(VI) samples was developed. In this method, Aliquat 336, as an anion exchanger was loaded into filter paper to form a test strip. This test strip was used to pre-concentrate Cr(VI) when contacting with the diluted Cr(VI)-containing solution. The Cr(VI)-containing test strip was the

Effect of iron and phosphate on bacterial cyanide formation determined by methemoglobin in two-dimensional gradient microcultivations.

Cyanide formation by microorganisms is typically observed during early stationary growth phase and is facilitated by the presence of iron(III) and inorganic phosphate. Extracellular free cyanide in aqueous solutions might readily react with methemoglobin added and can be determined by UV/VIS spectroscopy. As alternative to existing methods, this provided the basis for an analytical method which has not been used previously for the determination of cyanide in bacterial cultivations. We successfully applied the technique to study the combined effect of both iron(III) and phosphate on the cyanide

Application of surfactant enhanced permanganate oxidation and bidegradation of trichloroethylene in groundwater.

The industrial solvent trichloroethylene (TCE) is among the most ubiquitous chlorinated solvents found in groundwater contamination. The main objectives of this study were to evaluate the feasibility of using non-ionic surfactant Simple Green (SG) to enhance the oxidative dechlorination of TCE by potassium permanganate (KMnO4) employing a continuous stir batch reactor system (CSBR) and column experiments. The effect of using surfactant SG to enhance the biodegradation of TCE via aerobic cometabolism was also examined. Results from CSBR experiments revealed that combination of KMnO4 with surfac

Systematic analysis and overall toxicity evaluation of dioxins and hexachlorobenzene in human milk.

A systematic method for analyzing dioxins (PCDDs, PCDFs and dioxin-like PCBs), hexachlorobenzene (HCB), heptachlor epoxide and beta-hexachlorocyclohexane (HCH) in human milk was developed to determine the residual amount of HCB in human milk and to evaluate the overall toxicity of both dioxins and HCB in human milk. The fractionation behavior of HCB on chromatography with silica gel, alumina, and activated carbon/silica gel, and the concentrated sulfuric acid decomposition method, which is widely used as a dioxin cleanup method, were studied in order to make the preprocessing operation for HCB

A study on the protection to relics and the related problems with diffuse reflectance spectroscopy.

The application of diffuse reflectance spectroscopy to relic protection is studied by using a self-made fiber optics reflectance spectrophotometer. The major work done includes: (1) the composition of pigment on colored relics and its changes are identified; (2) the change on metal surface is monitored; (3) the reflectance spectrum characteristics of relic protection materials are studied. The results tell that diffuse reflectance spectroscopy is a new protection technique, characterized by its quickness and non-destructiveness to the relic.

What's new with the NRC certified reference material program.

The environmental CRMs currently available from NRC Ottawa comprise four natural waters for trace elements, four biological tissues for trace elements and methylmercury, three marine sediments for major and trace elements (one sediment has certified values for butyltins) and one fish tissue for organochlorines (PCBs, dioxins). Although significant effort has been expended to maintain this suite of materials, several new initiatives have also recently been completed or are in progress. The certified value for arsenobetaine in Dogfish Muscle CRM DORM-2 has been established. The certification app

Evaluation of Emit II reagents on the Chem 1.

An evaluation study of Syva Emit II reagents using the Chem 1 was performed for the following drugs: barbiturates, benzodiazepines, cannabinoids, benzoylecgonine, opiates, and phencyclidine. The Emit II reagents (100-mL bottles) were reconstituted to 70 mL and evaluated against the Emit d.a.u. reagents. A minimum of 446 samples were run for each drug. For all drugs tested, there were a total of 11 discordant results between the two reagents. The Emit II reagent was found to be correct on 8 of the 11 discordances after retesting by FPIA or GC/MS. The CV of within-run and day-to-day precision of

Experimental parameters affecting hypercoulometric response in gas-phase coulometry.

The response of an electron-capture detector designed for coulometric response was examined as a function of solute, pulse interval, carrier-gas type, solute amount, carrier-gas flow rate, and chromatographic-peak profile. For CCl3F, it was shown that the average numbers of electrons absorbed per molecule ranged from 1.0 to 4.1, varying primarily with pulse interval but also displaying a complex dependence on sample amount. Comparison of these results with theoretical expectations leads to the conclusion that a 1:1 "coulometric response" may not be observed under certain operating conditions.

Spacer-Enhanced Resolution of Thiamine and Pyridoxine in Hydrodynamically Closed Two-Dimensional Capillary Isotachophoresis Coupled With Tandem Mass Spectrometry.

The effect of specific spacers added to the separation environment of capillary isotachophoresis in a hydrodynamically closed two-dimensional device hyphenated with tandem mass spectrometry is reported in this short communication. Combination of two spacers, here, 2-amino-2-hydroxymethyl-propane-1,3-diol and histidine, led to enhanced resolution of two model analytes (thiamine and pyridoxine) in model and real pharmaceutical samples (tablet drug dosage form). The addition of the spacers at relatively high concentration level (i.e., 20 µg/mL) caused no significant interference and suppression o

Determination of drug-serum protein interactions via fluorescence polarization measurements.

New fast methods for the determination of pharmacokinetic behaviour of potential drug candidates are receiving increasing interest. We present a new homogeneous method for the determination of drug binding and drug competition for human serum albumin and alpha(1)-acid glycoprotein that is amenable to high-throughput-screening. It is based on selective fluorescent probes and the measurement of fluorescence polarization. This leads to decreased interference with fluorescent drugs as compared with previously published methods based on similar probes and the measurement of fluorescence intensity.
