Holoprosencephaly as a possible embryonic alcohol effect.

Three mothers of infants with holoprosencephaly consumed alcohol heavily in pregnancy. We postulate that early alcohol exposure is a possible cause of their malformation. The 3 mothers consumed alcohol only in the first trimester but the first mother continued to take chlordiazepoxide and imipramine throughout the pregnancy. Her infant had an alobar holoprosencephaly associated with a median cleft lip, ocular hypotelorism, and a flat nose. The other infants had semilobar holoprosencephaly and hydrocephalus. These latter 2 infants did not show the characteristic facies of the fetal alcohol synd…

Psychotropic Drugs With Long Half-Lives: Implications for Drug Discontinuation, Occasional Missed Doses, Dosing Interval, and Pregnancy Planning.

The half-life of a drug is the time taken for the blood level of the drug to fall by half, provided that no more doses of the drug are administered in the intervening period. Many psychotropic drugs and their active metabolites, if any, have very long half-lives that extend for 2 days or longer. Examples are chlordiazepoxide, diazepam, fluoxetine, vortioxetine, aripiprazole, brexpiprazole, cariprazine, penfluridol, donepezil, and memantine. Other drugs with long half-lives that psychiatrists may prescribe include levothyroxine and zonisamide. Psychotropic drugs with long half-lives take long t…

Pharmacotherapy in alcoholism.

This review categorizes five main uses of pharmacologic agents in the treatment of alcoholism: reversing the active pharmacologic effects of … e and dependence with alcohol, benzodiazepines--especially intermediate acting preparations such as chlordiazepoxide and diazepam--are the mainstay in treating alcohol withdrawal, including convulsions and delirium tremens. Studies suggest that serotonin uptake inhibitors such as zimelidine, citalopram, viqualine, and fluoxetine may reduce alcohol consumption and …

Differential pharmacological reactivity of aversion induced by stimulation of periaqueductal gray or mesencephalic locomotor region.

Rats were trained to switch-off aversive electrical brain stimulations applied to the periaqueductal gray (PAG) or mesencephalic locomotor region (MLR) by pressing a bar (switch-off behavior). We investigated the effects of IP injections of the benzodiazepine (BZ) receptor inverse agonist FG 7142 (2.5, 5, 10 mg/kg) or BZ receptor agonist chlordiazepoxide (CDP: 5 mg/kg) on the switch-off latency, i.e., the time elapsed between the onset of the stimulation and its offset by a press of the bar. It was found that FG 7142 decreased, whereas CDP increased the mean switch-off latency for electrical s…

Benzodiazepines: reconsidered.

The benzodiazepines were first introduced in 1960. Chlordiazepoxide (Librium) was the first of the class of drugs called benzodiazepines, in a deliberate attempt to synthesize a tranquilizer without the sedative properties and abuse, addiction, tolerance, and dependence potential of the barbiturates, and other sedative/hypnotic drugs. The popularity of the benzodiazepines rose steadily to a peak period in the mid 1970s when diazepam (Valium) was the most commonly prescribed drug of any kind, including antihypertensive, analgesic and other psychotropic medications. The current evaluations of be…

Buspirone, gepirone and ipsapirone disrupt both active and passive avoidance responding in rats.

The anxiolytic drug buspirone and its structural analogues gepirone and ipsapirone produce behavioural effects which differ from those seen … er injection of these drugs. A similar effect on the passive avoidance response was not produced by chlordiazepoxide; diazepam, CL 218, 872, haloperidol, imipramine or d-amphetamine. These disruptions of the performance of both active and passive avoidance indicate a unique pharmacological profile for buspirone and similar drugs.

Quantitative electrocortical changes in the rat induced by phencyclidine and other stimulants.

An automated technique for the continuous analysis of different frequency bands of the electrocorticogram (ECoG) of the rat has been used to … se the changes in the ECoG. Chlorpromazine caused a slight shift in the dose-response curves as did chlordiazepoxide when used with phencyclidine. The GABA agonists, THIP and muscimol, had no effect on the stimulation of the ECoG. In contrast another presumed GABA agonist, baclofen, proved to be the most effective agent for blocking the stimulation induced by phen …

Psychotropic drug metabolism in fetal alcohol syndrome.

Simultaneous intake of ethanol with chlorpromazine (thorazine), an antipsychotic drug, leads to about 60% decrease in the chlorpromazine rem … aternal liver. Simultaneous acute administration of ethanol (2g/kg) with psychotropic drugs such as chlordiazepoxide (librium), diazepam (valium), chlorpromazine (thorazine) or meprobamate (equanil) to pregnant or non-pregnant rats led to a decrease in the blood alcohol clearance rates. In another group of nonpsychotropic drugs tested, tolbutamide (orinase) produc …

Anticonvulsant activity of delta9-tetrahydrocannabinol compared with three other drugs.

Delta9-tetrahydrocannabinol (THC) was compared with diphenylhydantoin (DPH), phenobarbital (PB) and chlordiazepoxide (CDP) using several standard laboratory procedures to determine anticonvulsant activity in mice, i.e., the maximal electroshock test (MES), and seizures induced by pentylenetetrazol, strychnine and nicotine. In the MES test, THC was the least potent and DPH the most potent blocker of hind limb tonic extensor convulsions whereas THC was the most potent and DPH the least potent in increasing the latency to this response and in preventing mortality. Seizures and mortality induced b…

The clinical pharmacology of oxazepam.

The benzodiazepines have been available now for about 15 years but new aspects of their actions are being elucidated all the time. Oxazepam has a short half-life and inactive metabolites and therefore differs from most other benzodiazepines. Some aspects of its clinical pharmacology are reviewed in outline. Firstly, the effects of the drug on psychological functioning is discussed. Next, the controversy regarding benzodiazepines and aggression is examined. Oxazepam appears exceptional here in that it is not associated with paradoxical release of aggression in patients. Finally, the ethical imp

Rapid tolerance and cross-tolerance as predictors of chronic tolerance and cross-tolerance.

Hypothermia and motor impairment (tilt-plane) tests were used to assess the phenomenon of rapid tolerance to ethanol and cross-tolerance to … both tests, to the various alcohols (n-propanol, n-butanol, and t-butanol) and the benzodiazepines (chlordiazepoxide, diazepam, oxazepam, and flurazepam) tested. Ethanol pretreatment also conferred clear rapid cross-tolerance to barbital and phenobarbital, but did not result in rapid cross-tolerance to pentobarbital, secobarbital, amobarbital, or thiopental. The r …

[Analysis of benzodiazepine derivative mixture by gas-liquid chromatography].

The analysis of mixture of benzodiazepine derivates (chlordiazepoxide, flunitrazepam, medazepam, nitrazepam, oxazepam and tetrazepam) by gas--liquid chromatography (GLC) in purpose to separate and identify these psychotropic drugs in mixture is presented in this article. The experiment was carried out in vitro, accommodating this method for identification and separation of drugs, isolated from biological objects (blood and urine). Referring to data of annual reports of chemical investigations (1) above-mentioned psychotropic drugs are very frequent among drug intoxication. In most cases they a…
