Sixty-seven human tumor cell lines and 15 lines derived from normal tissue were examined for the production of the oncodevelopmental markers carcinoembryonic antigen, alpha and beta subunits of chorionic gonadotropin, placental and nonplacental forms of alkaline phosphatase, gamma-glutamyltransferase, cystyl aminopeptidase, and calcitonin. Both intracellular and extracellular levels of these markers were determined at three phases during the growth of each culture. Sixty-eight percent of the cell lines produced elevated levels (greater than or equal to 90th percentile) of at least one marker.

Cytotrophoblasts (from term placentae) and cells from the choriocarcinoma cell line JAr were cultivated either separately or in co-culture for 72 h. RNA was isolated from the cell cultures and Northern blots were developed using equal amounts of RNA. The RNA was hybridized with cDNA probes for CG alpha, CG beta and hPL. Corresponding m-RNAs were detected in the three RNAs except for hPL m-RNA which was absent from JAr cells RNA. Ten patients with ectopic pregnancy, probably non complicated, diagnosed by transvaginal sonography and serial determinations of subunit B of human chorionic gonadotrophin, were treated with a single injection of methotrexate intraovular (5 cases), intramuscular (3 cases) or with expectant management. We obtained clinical resolution and undetectable hormone levels in seven patients and three underwent additional surgery.

Three patients had an undetected pregnancy when they commenced a GnRH-a on the 21st day of their cycle before ovulation stimulation for IVF. Two of the three pregnancies were later diagnosed as ectopic gestations. Based on this and previous case reports in the literature, it is suggested that luteal phase administration of GnRH-a may predispose to EPs, particularly in patients with damaged tubes, by reducing tubal motility secondary to a transient increase in circulating P concentrations.

A case of ectopic pregnancy with a gestational sac in the right tube of 25 mm diameter and beating fetal heart was treated conservatively by transvaginal, ultrasonically guided injection of potassium chloride and methotrexate. Serum levels of beta-human chorionic gonadotrophin returned to normal in 42 days and the tubal mass collapsed gradually and resolved in 7 weeks.

As polyembryomas are special forms of human teratomas in which numerous structures mimicking the early stages of the human embryo are conspicuous, immunoperoxidase staining of these embryoid bodies (EB) has been used to reveal the initial appearance and localization of beta HCG and AFP, respectively, in amnion and in primary yolk-sac. EB are involved in the building of various questionable patterns of teratomatous germ-cell tumours. Special emphasis has been placed on tracking yolk-sac tumour patterns through dislocating EB, using a specific anti-human AFP serum.

An enzyme immunoassay (EIA) for hCG-beta CTP was newly developed. Anti hCG-beta CTP antibody was prepared from antiserum to the synthetic peptide corresponding to the carboxyl terminal region by immunization of rabbits with hCG-beta subunit. Specimens to be tested were reacted with the antibody labelled with peroxidase and determined with a spectrophotometer after the addition of o-phenylenediamine. The atypical course after laparoscopic conservative treatment of tubal pregnancy is described. Routine measurements of the serum HCG levels postoperatively showed, after a short period of decreasing HCG levels, a new increase of HCG values. Under the suspicion of a disseminated intraperitoneal trophoblastic tissue the patient was treated with 20 mg methotrexate q.i.d. per os for 5 days, as a result of which HCG levels returned to normal within a short time.

A 56-year-old woman showed renal obstruction on the right side. Further examinations revealed a tumor of the renal pelvis. Nephroureterectomy on the right side was done. Within our surgical collection clinically inactive pituitary adenomas represent 30.7% of all pituitary tumours.
