Repeated Cross-Sectional Surveillance and ORF5-Based Molecular Epidemiology of Porcine Reproductive and Respiratory Syndrome Virus in Anhui Province, China, 2019-2024.

Porcine reproductive and respiratory syndrome virus (PRRSV) remains a major threat to swine production, and its circulation after the African swine fever outbreak requires continued surveillance. This study investigated the temporal, regional, and genetic characteristics of PRRSV in Anhui Province from September 2019 to November 2024. Ten rounds of repeated cross-sectional surveillance were conducted at 147 slaughterhouses and 21 rendering plants. Tissue samples were collected by random, cluster, or risk-based sampling, pooled in groups of five, and tested by RT-qPCR. Representative positive s

Functional characterization of recombinant major envelope protein (rB2L) of orf virus.

Orf, or contagious ecthyma, a highly contagious transboundary disease of sheep and goats, is caused by a double-stranded DNA virus (ORFV) belonging to the genus Parapoxvirus of the family Poxviridae. The ORFV genome encodes the major envelope proteins B2L and F1L, which have been found to be highly immunogenic and have multiple functional characteristics. In order to investigate the functional properties of the B2L protein, in this study, the B2L gene of ORFV strain 59/05, encoding recombinant mature B2L (aa 1M-D334), was produced as a fusion protein in Escherichia coli. The functional charact

[Development of a colloidal gold immunochromatographic test strip for detecting gB antibodies of porcine pseudorabies virus].

This study aims to establish a colloidal gold immunochromatographic test strip for the detection of glycoprotein B (gB) antibodies of porcine pseudorabies virus (PRV), thus providing a simple and rapid technique for the detection of PRV gB antibodies. The monoclonal antibodies to gB were prepared by the hybridoma technology. The gB protein was coupled with colloidal gold, and then gB and its monoclonal antibodies were separately sprayed onto a nitrocellulose membrane as the test line and control line. After optimization of conditions, a colloidal gold immunochromatographic test strip for detec

Development of an EvaGreen-based multiplex real-time PCR assay with melting curve analysis for simultaneous detection and differentiation of six viral pathogens of porcine reproductive and respiratory disorder.

Concurrent infection of pigs with two or more pathogens is common in pigs under intensive rearing conditions. Porcine circovirus type 2 (PCV2), porcine parvovirus (PPV), porcine reproductive and respiratory syndrome virus (PRRSV), classical swine fever virus (CSFV), Japanese encephalitis virus (JEV) and pseudorabies virus (PRV) are all associated with reproductive or respiratory disorders or both and can cause significant economic losses in pig production worldwide. An EvaGreen-based multiplex real-time PCR (EG-mPCR) with melting curve analysis was developed in this study for simultaneous dete

No evidence of spread of Linda pestivirus in the wild boar population in Southern Germany.

Lateral-shaking inducing neuro-degenerative agent virus (LindaV) is a novel member of the highly diverse genus Pestivirus within the family Flaviviridae. LindaV was first detected in Austria in 2015 and was associated with congenital tremor in piglets. Since then, the virus or specific antibodies have been found in a few further pig farms in Austria. However, the actual spatial distribution and the existence of reservoir hosts is largely unknown. Since other pestiviruses of pigs such as classical swine fever virus or atypical porcine pestivirus can also infect wild boar, the question arises wh

[Effect of specific vaccinal antigens and preparations on general resistance in growing experimental and domestic animals].

Studied was the effect of some viral and bacterial antigens as well as of a preparation obtained by Filatov's method (modified by the author) on the general resistance in growing laboratory and domestic animals. It was found that in infantile albino mice the best protection against challenge with Escherichia coli and Pasteurella avicida was provided through the treatment with a biostimulator and a killed culture of a strongly proteolytic, unidentified strain of the 'T3' bacterium. The vaccines against Newcastle disease, hog cholera+erysipelas, and anthrax were shown to have a weaker action. It

Development of a Competitive Chemiluminescent Enzyme Immunoassay Based on Recombinant FMDV VP2 Protein for Rapid and Simultaneous Detection of Antibodies Against Foot-and-Mouth Disease Virus Serotypes O and A.

Timely detection of specific antibodies against foot-and-mouth disease virus (FMDV) is critical to effective prevention and control of the disease. In this study, we developed a competitive chemiluminescent enzyme immunoassay (cCLEIA) based on recombinant FMDV VP2 protein (FMDV-VP2 cCLEIA) for simultaneous detection of antibodies against FMDV serotypes O and A in serum samples. Using a cut-off of 35% percentage inhibition (PI), the FMDV-VP2 cCLEIA achieved a relative sensitivity of 101.5% and a relative specificity of 102% compared with commercial FMDV serotypes O and A liquid-phase blocking E

Validation of a Broad-Range Conventional RT-PCR Assay for Pestivirus Surveillance and Molecular Detection of BVDV-2a in Pigs in Costa Rica.

The genus Orthopestivirus, within the family Flaviviridae, includes important pathogens such as classical swine fever virus (CSFV), bovine viral diarrhea virus types 1 and 2 (BVDV-1 and BVDV-2), and border disease virus (BDV). Although Costa Rica is endemic for BVDV, it remains officially free of CSFV and BDV, highlighting the need for reliable molecular tools for pestivirus surveillance and differential diagnosis in swine populations. This study validated a conventional broad-range in-house RT-PCR assay targeting the 5' untranslated region (5'UTR) of pestiviruses, a genomic region that is hig

Development of a loop-mediated isothermal amplification assay for the detection of porcine hokovirus.

Hokoviruses have recently been detected as pathogens belonging to the family Parvoviridae, which comprises porcine hokovirus (PHoV) and bovine hokovirus (BHoV). In this study, we developed a loop-mediated isothermal amplification (LAMP) assay for the rapid, specific and sensitive detection of PHoV. A set of four primers specific for six regions within the PHoV VP1/2 genes was designed using online software. The reaction temperature and time were optimized at 65°C and 60 min, respectively. LAMP products were detected by agarose gel electrophoresis or by visual inspection of a color change cause

Identification and genetic characterization of new bovine viral diarrhea virus genotype 2 strains in pigs isolated in China.

Classical swine fever (CSF)-like symptoms in pigs regarded as free from CSF has been reported previously. From sick pigs with CSF-like symptoms, and who had been inoculated with the hog cholera vaccine, samples were collected and subjected to RT-PCR using specific primers. Twelve bovine viral diarrhea virus 2 (BVDV-2) strains were screened and isolated. Homology comparison showed that the E2 genes of the twelve isolates were highly conserved. The genome of the one of the BVDV-2 isolates (named as SH-28) from the sick pigs, which showed a noncytopathic effect in MDBK cell cultures and strong re

Flaviviridae RdRp exploits NSUN2-driven m5C methylation to establish persistent infection.

Flaviviridae viruses constitute formidable zoonotic agents with substantial global health and economic ramifications, attributable to their adeptness at circumventing host immune surveillance and establishing persistent infections across human and animal populations. Despite their pervasive impact, broadly effective antiviral strategies remain elusive. Emerging studies underscore the pivotal role of RNA modifications, particularly 5-methylcytosine (m5C), in fine-tuning host-pathogen interactions. Expanding upon prior evidence linking NSUN2-mediated m5C deposition to Classical swine fever virus

Seroprevalence of the viral pig diseases among backyard pigs in Chiang Mai, Thailand.

The Participatory One-Health Disease Detection project (PODD) (www.cmonehealth.org) developed a health-based surveillance system with the local government of Chiang Mai community ownership that has been created a mobile application on smartphone for reporting an abnormal event, especially animal health. Previously, the PODD project has obtained a significant number of pig abnormal events. Therefore, there are likely to be some diseases that are currently circulating among backyard pigs. A cross-sectional serological study was undertaken to determine the risk factors for virus infection and pre

Safety and Immunogenicity of Chimeric Pestivirus KD26_E2LOM in Piglets and Calves.

A chimeric pestivirus (KD26_E2LOM) was prepared by inserting the E2 gene of the classical swine fever virus (CSFV) LOM strain into the backbone of the bovine viral diarrhea virus (BVDV) KD26 strain. KD26_E2LOM was obtained by transfecting the cDNA pACKD26_E2LOM into PK-15 cells. KD26_E2LOM chimeric pestivirus proliferated to titers of 106.5 TCID50/mL and 108.0 TCID50/mL at 96 h post-inoculation into PK-15 cells or MDBK cells, respectively. It also reacted with antibodies specific for CSFV E2 and BVDV Erns, but not with an anti-BVDV E2 antibody. Piglets (55-60 days old) inoculated with a high d

Multi-omics analysis reveals mechanisms of Qingying granules in treating porcine warm disease: dosage optimization and systems biology insights.

BACKGROUND: Warm diseases in swine, particularly those associated with highly pathogenic viruses such as African swine fever virus (ASFV) and porcine reproductive and respiratory syndrome virus (PRRSV), inflict devastating economic losses on global pork production. Current control strategies rely heavily on antibiotics and vaccines; however, emerging antimicrobial resistance and vaccine efficacy variability necessitate alternative therapeutic approaches. Traditional Chinese Veterinary Medicine offers potential solutions, yet rigorous evidence-based studies are required to meet modern regulator

Molecular Epidemiology Questions Transmission Pathways Identified During the Year 2000 Outbreak of Classical Swine Fever in the UK.

The last outbreak of classical swine fever (CSF) in the UK occurred in 2000. A total of 16 domestic pig holdings in the East Anglia region were confirmed as infected over a 3-month period. Obtaining viral genome sequences has since become easier and more cost-effective and has accordingly been applied to trace viral transmission events for a variety of viruses. The rate of genetic evolution varies for different viruses and is influenced by different transmission events, which will vary according to the epidemiology of an outbreak. To examine if genetic changes over the course of any future CSF

Two kinds of traditional Chinese medicine prescriptions reduce thymic inflammation levels and improve humoral immunity of finishing pigs.

In animal husbandry, traditional Chinese medicine (TCM) as a reasonable alternative to antibiotics has attracted more and more concerns to reduce microbial resistance. This study was aimed to investigate the effects of dietary supplementation with TCM prescriptions on serum parameters and thymus inflammation responses in finishing pigs. Thirty finishing pigs were randomly divided into three groups, which included the Con group (basal diet), the TCM1 group (basal diet supplemented with Xiao Jian Zhong prescriptions), and the TCM2 group (basal diet supplemented with Jingsananli-sepsis). Bovine viral diarrhea virus E2 targets SLC3A2 to modulate lipid peroxidation for replication advantage.

Pestiviruses, part of the Flaviviridae family, are responsible for major livestock diseases such as bovine viral diarrhea, classical swine fever, and border disease. These infections lead to substantial economic losses worldwide. However, their pathogenesis remains incompletely understood, limiting the development of effective control strategies. In this study, we explored how bovine vial diarrhea virus (BVDV) regulate host lipid peroxidation (LPO) to support viral replication. We initially found that early infection with BVDV enhanced intracellular redox balance by upregulating the cystine/gl
