T-cell colony formation in patients with T-cell malignancies: growth factor requirements for in-vitro proliferation of peripheral blood T-cell colony-forming cells.

Peripheral blood T colony-forming cells (T-CFC) from patients with T-cell malignancies are capable of proliferation in methylcellulose, in the absence of added growth factors or mitogenic stimulation. We show here that based on the spontaneous plating efficiencies of their T-CFC, two groups of patients can be established: Group A (10 patients) with a high colony number (more than 100 colonies/10(5) seeded cells), and group B (12 patients) with less than 100 colonies/10(5) cells. The addition of interleukin 2-(IL2) containing PHA-leukocyte conditioned medium enhanced colony growth from group B

Meta-analysis of randomized controlled trials of prophylactic granulocyte colony-stimulating factor and granulocyte-macrophage colony-stimulating factor after autologous and allogeneic stem cell transplantation.

PURPOSE: The primary objective of our meta-analysis was to determine whether prophylactic hematopoietic colony-stimulating factors (CSFs) after hematopoietic autologous and allogeneic stem-cell transplantation (SCT) reduced documented infections. Our secondary objectives were to determine whether prophylactic CSFs affected other outcomes including parenteral antibiotic therapy duration, infection-related mortality, graft-versus-host disease (GVHD), or treatment-related mortality. METHODS: We included studies if there was random assignment between CSFs and placebo/no therapy and CSFs were give

MALDI Tandem Mass Spectrometry for Colony-Based Dereplication of Natural Products.

Microbial libraries remain an important resource for natural product discovery; however, constructing taxonomically and chemically diverse collections remains a challenge. Advances in dereplication strategies, including molecular networking, have reduced the rediscovery of known bioactive molecules and facilitated the identification of novel chemical scaffolds, but these approaches are typically applied after library construction. Furthermore, many dereplication workflows require fermentation and extraction, increasing the time needed to assess a microbe's metabolite profile. Here, we integrat

Mechanical interactions govern self-organized ordering in bacterial colonies on surfaces.

Bacterial colonies growing on surfaces are shaped by mechanical stresses transmitted through the community, governed by the balance between cell growth and steric and cell-substrate interactions. Using overdamped dynamics simulations of nonmotile, stress-responsive bacteria, we systematically investigate how these mechanical factors determine colony morphology and internal organization. Growth-induced extensile stresses compete with steric constraints, giving rise to the spontaneous formation of microdomains composed of highly aligned cells. We characterize this self-organization through the d

Quantitative evaluation of early fungal morphogenesis identifies markers of colony establishment.

Quantitative methods to identify when a fungal colony becomes an established, radially expanding structure remain limited, particularly during early growth on solid media. Here, we present an image-based framework to evaluate early colony morphology in the filamentous fungus Trichoderma atroviride for different inoculum sizes. Two-dimensional colonies initiated either from a single conidium or from thousands of spatially confined conidia were imaged at defined time points following germination and analyzed using digital image processing to extract morphology metrics based on skeleton and tip s

An Efficient Colony PCR Method for Construct Verification in the Basidiomycete Ustilago maydis.

Rapid advances in AI-driven annotation and automation are increasing the demand for fast and reliable screening protocols for reverse genetic approaches. Traditional methods for genomic DNA extraction from the basidiomycete Ustilago maydis are often time-consuming and require expensive reagents and hazardous chemicals, making them unsuitable for large-scale mutant screening. Existing colony PCR protocols using crude lysates without DNA precipitation are fast but often fail in terms of reproducibility because cellular components can interfere with polymerase activity or DNA stability. These lim

Impact of pesticides on honeybees (Apis mellifera) physiology, foraging and colony: An overview.

Honeybees (Apis mellifera) play an indispensable role in global agricultural production, pollinating crops that support approximately one-third of the world's food supply and bolstering biodiversity through these essential ecosystem services. Yet, their survival is severely threatened by multiple environmental stressors, including biological (e.g. pathogens, parasites) and chemical (e.g. plant protection products/agents) factors, particularly pesticides. Pesticides, a broad spectrum of agrochemicals such as insecticides, herbicides, and fungicides are widely deployed to protect crops from inse

Stimulus Optimization Using the Artificial Bee Colony Algorithm for Visual Neuroprostheses.

OBJECTIVE: Optimization of electrical stimulation patterns can enhance the performance of visual neuroprosthetics by replicating the selective and temporally precise neural responses characteristic of natural vision. METHODS: An in silico approach employing the Artificial Bee Colony (ABC) algorithm was developed to optimize electrical stimulus waveforms that evoke physiologically realistic responses in retinal ganglion cells (RGCs). The optimization was guided by a computational model of ON and OFF RGCs, with the goal of matching recorded neural responses from the rat dorsolateral geniculate

Expression and Role of Colony Stimulating Factor 1 Receptor During Odontogenesis.

In osteopetrotic mice with homozygous inactivating mutations in the colony stimulating factor 1 (Csf1op/op) or its receptor (Csf1r-/-) gene, teeth fail to erupt due to severe reduction in osteoclastogenesis. Dental abnormalities have been described in the unerupted teeth of these models, but it remains unclear whether these defects arise from direct roles of CSF1R in odontogenesis or indirectly from impaired bone remodeling associated with failed eruption. Here, we examined the spatiotemporal expression of CSF1R during tooth development and inhibited CSF1R pharmacologically in utero using PLX5

Colony Stimulating Factor-1 Receptor-Related Disorder Treated With Ilunazebart.

Colony stimulating factor-1 receptor-related disorder (CSF1R-RD) is a rare, autosomal dominant neurodegenerative disease caused by loss-of-function variants in the CSF1R gene, leading to microglial dysfunction and progressive white matter degeneration. Therapeutic strategies targeting microglial pathways, including activation of triggering receptor expressed on myeloid cells 2 (TREM2), have been proposed to compensate for impaired CSF1R signaling. Iluzanebart (VGL101), a monoclonal antibody TREM2 agonist, has shown promise in preclinical models. We report a longitudinal clinical course, neuroi

Selection bias in microbial mutation accumulation studies and the impact of colony growth.

In microbial mutation-accumulation (MA) studies, it is widely thought that natural selection is silenced by repeated single-cell bottlenecks. In support of this claim, 40 published tests for selection bias failed to detect a deficit of non-synonymous relative to synonymous mutations in wild-type microbes. Here we show that this most likely reflects selective reporting and lack of power. Our meta-analysis of 10,856 mutations from wild-type microbial MA reveals a clear signal of selection: non-synonymous mutations are observed 7.7\% less often than synonymous mutations. However, our inference of

Intestinal entamoebiasis in a captive colony of Anderson's crocodile newts Echinotriton andersoni.

Entamoeba protozoa are known to cause enteric and sometimes systemic disease in both domestic and non-domestic animals. In amphibians, Entamoeba spp. are primarily documented as incidental gastrointestinal inhabitants, with rare reports of clinically significant entamoebiasis. In the last decade, mortality outbreaks caused by a recently described protozoan, Entamoeba sp. CT1, have been documented in free-ranging anurans in Australia and managed anurans in the USA associated with gastroenteritis and substantial morbidity and mortality. In 2023, a small population of Anderson's crocodile newts E

Recovering the pre-colonial population structure of Khoe-San descendant populations.

San populations from Botswana and Namibia retain exceptional linguistic, cultural and genetic diversity, but few Khoisan-speaking groups remain south of the Kalahari Desert. However, historically, far southern Africa was home to many San and Khoekhoe groups. Popular opinion often implies that such populations do not contribute to the ancestry of contemporary South Africans. Here, we characterize the genetic ancestry of self-identified South African Coloured groups and reconstruct pre- and colonial population structure from 620 newly sampled individuals. These groups retain majority Khoe-San ge

Genomic insights into ancestry and infectious disease in 17th-century colonial Brazil.

During urban redevelopment in the historic district of a Brazilian metropolis, archaeologists uncovered a previously undocumented 17th-century cemetery, containing the closely spaced remains of over two dozen young adult males of military age. Historical records suggest the site once housed a makeshift hospital, raising questions about the origins and causes of death of those interred, particularly given the absence of skeletal evidence for violent or fatal trauma. The current study integrates bioarchaeological, historical, and genomic data to investigate the ancestry and putative disease-rela

A Dark Horse: Colonial System of Integration in Ctenostome Bryozoans (Gymnolaemata: Ctenostomata).

The colonial system of integration (CSI) provides intracolonial nutrient supply in many gymnolaemate bryozoans. In Ctenostomata, its presence is known for species with stolonal colonies, for example, vesicularioideans, but its structure is almost unexplored. The CSI is thought to be absent in alcyonidioideans and other ctenostomes. Here, we present the first detailed description of the CSI ultrastructure in both autozooids and kenozooids of two vesicularioideans, Buskia nitens and Amathia gracilis, and two alcyonidioideans, Alcyonidium hirsutum and Flustrellidra hispida. We revealed difference

Colony-Stimulating Factor 3 Receptor Mutations and Variants in Hematological Malignancies.

Colony-stimulating factor 3 (CSF3), additionally called granulocyte colony-stimulating factor (G-CSF), is the major cytokine regulating neutrophil production and also impacting their function. The actions of this cytokine are mediated through its unique receptor, the colony-stimulating factor 3 receptor (CSF3R). Several classes of pathogenic mutations in the CSF3R gene have been identified that have distinct biological properties and clinical impacts. This review provides an overview of CSF3R, the various pathogenic CSF3R mutations/variants and their biological effects. It also details the dis

Rabies Outbreak in an Urban, Unmanaged Cat Colony - Maryland, August 2024.

Exposure to a rabid animal is a life-threatening emergency because infection is almost always fatal if rabies postexposure prophylaxis (PEP) is not initiated promptly. In August 2024, the Maryland Department of Health was notified that an aggressive feral cat had been captured and euthanized and had subsequently received a positive test result for rabies. The cat was part of a group of approximately 20 local feral cats and kittens that was receiving no, or little, human care (i.e., an unmanaged cat colony). Cecil County (Maryland) Health Department and the Maryland Department of Health coordin

Test of the negative feedback hypothesis of colony size sensing in social insects.

Social insects can sense colony size-even without visual information in a dark environment. How they achieve this is yet largely unknown. We empirically tested a hypothesis on the proximate mechanism using ant colonies. In Diacamma colonies, the monogynous queen is known to increase the effort devoted to queen pheromone transmission behaviour (patrolling) as the colony grows, as if she perceives colony size. The negative feedback hypothesis assumes that, through repeated physical contact with workers, the queen monitors the physiological state (fertility) of workers and increases her patrollin

Longitudinal Survey of Coronavirus Circulation and Diversity in Insectivorous Bat Colonies in Zimbabwe.

BACKGROUND: Studies have linked bats to outbreaks of viral diseases in human populations such as SARS-CoV-1 and MERS-CoV and the ongoing SARS-CoV-2 pandemic. METHODS: We carried out a longitudinal survey from August 2020 to July 2021 at two sites in Zimbabwe with bat-human interactions: Magweto cave and Chirundu farm. A total of 1732 and 1866 individual bat fecal samples were collected, respectively. Coronaviruses and bat species were amplified using PCR systems. RESULTS: Analysis of the coronavirus sequences revealed a high genetic diversity, and we identified different sub-viral groups in
