Sera were collected from 3753 cows in Benhazi cow project (Libya). The rose bengal, complement fixation and serum agglutination serological tests were employed to test these sera for evidence of brucellosis. The overall reactivity was detected in all (0.3%) cows. The prevalence of the highest reactors (1.0%) was in Abo Atney Dairy Station. In other dairy stations investigated the incidence of brucellosis ranged from zero to 0.3%.

The use of a radioimmunoassay and an enzyme immunoassay for early diagnosis of Q fever is described, both of which are based on the IgM antibody-capture principle. A commercially available phase II antigen and a labeled, purified anti-Q-IgG of human origin were employed. With these tests Q fever antibodies were detected earlier in the course of infection than with the complement fixation test.

A Q fever epidemic focus with 46 cases was detected in a rural locality among the workmen of livestock farms. In 33 cases, the subclinical and inapparent form of the disease was detected by serologic tests, performed in the focus. Complement fixation antibody titers persisting in very high titers, a retrospective diagnosis of infection may be established even 3-4 months after the onset of the disease.

August Paul von Wassermann (1866-1925), German bacteriologist, together with Albert Neisser (1855-1916), German dermatologist and venereolog … ch cases were reported in 1909. With the discovery of new and more specific tests for syphilis, the complement fixation tests of Wassermann type gradually went into oblivion. A new era in venereology was started with the discovery of T pallidum by Fritz Schaudinn (1871-1906) and Erich Hoffmann (1868-1954) in 1905, and the development of serology of syphilis by Wassermann, …

In the spring of 1993, an outbreak of respiratory infection affected 113 persons (103 males) in Jedl'ové Kostol'any, a village located in a hilly area of western Slovakia. Q fever, manifested as a flu-like illness with atypical pneumonia and hepatic involvement, was diagnosed using four serological tests (microimmunofluorescence, microagglutination, complement fixation, and enzyme immunoassay). Aborting goats were proven as a source of infection. During a 4-year follow-up study, no chronic form of Q fever could be demonstrated, either clinically or by tests to detect phase I Coxiella burnetii

A group of 800 human sera, obtained from syphilitic and nonsyphilitic individuals were tested in the counterimmunoelectrophoresis (CIE) technique with Reiter protein antigen. The sera were assayed for comparison in the following treponemal and nontreponemal tests: standard serologic tests for syphilis with cardiolipin; complement fixation (Kolmer 1/5 vol) with Reiter protein antigen (RPCF); T. pallidium immobilization (TPI) test; haemoagglutination test for T. pallidum antibodies (TPHA). The sensitivity and specificity of CIE test, its simplicity, the low cost, the possibility of practical app

This is a case report of a 53-year-old woman involved in an outbreak of Q fever, in whom Q fever endocarditis was diagnosed 18 months after acute Q fever infection. At the time of diagnosis, she was completely asymptomatic and without screening for chronic Q fever, this severe potentially life-threatening infection would probably not have been recognised until significant valvular destruction had taken place. Early diagnosis enabled prompt, potentially curative medical treatment to start without the need for valvular heart surgery. The authors advocate that serological monitoring should be car

A dot enzyme-linked immunosorbent assay (dot ELISA) was developed for diagnosis of glanders in equines. The test was based on the detection of IgG antibodies to Pseudomonas mallei antigens bound to nitrocellulose coated on plastic strips (dipsticks), the reaction being amplified by an avidin-biotin system with biotinylated anti-horse IgG and horseradish peroxidase-avidin D. Sera from 810 normal, six naturally infected and 48 sensitized equines were tested by this assay, and results were compared with complement fixation, indirect haemagglutination and counter-immunoelectrophoresis tests. The sensitivities and specificities of 17 antibody detection tests for brucellosis in goats were estimated. Tests evaluated included the U.S … ensis antigens (SATA and SATM), serum enzyme-linked immunosorbent assay (ELISA), USDA cold-fixation complement fixation tests with B. abortus and B. melitensis antigens (CFA and CFM), USDA and Mexican milk ring tests (UBRT and MBRT), and a milk ELISA. Test sensitivity was evaluated by using two groups of 10 goats experimentally infected with B. melitensis or B. abortus and …

The incidence of coccidioidomycosis continues to increase. The diagnosis frequently relies on non-invasive diagnostic testing with immunodiffusion and complement fixation (CF) testing the current gold standard. A direct comparison of quantitative immunodiffusion and CF for IgG antibodies has not been previously reported. In a comparison of 368 samples, there was close concordance observed (360/368 = 97.8%) (P-value < .001). These tests can be considerably interchangeable in the reference laboratory setting.

The substance in syphilitic serum which is responsible for the Wassermann reaction, like that which determines the diagnostic flocculation tests, is associated with the globulin fraction of serum. Every positive Wassermann is accompanied by microscopic (or submicroscopic) aggregation, which is not an essential feature of the reaction; conversely, after every positive flocculation test, the washed precipitate will fix complement. An excess of antigen removes both flocculating and complement-fixing substances completely (>95 per cent). Heating the lipoid-reagin precipitate to 100 degrees for 1 m

In the studies of complement fixation described in this paper, the antigens were prepared from (a) normal monkey red cells, (b) parasitized red cells of monkeys dying with Plasmodium knowlesi infection, (c) the spleens of monkeys dying with Plasmodium knowlesi infection; the sera came from (a) normal human beings, (b) patients with syphilis, (c) patients with paresis who were receiving malaria therapy with Plasmodium knowlesi, Plasmodium vivax, or Plasmodium falciparum, and (d) patients with malaria alone. The malarial antigens gave negative complement fixation reactions with 70 to 80 per cent

The diagnosis of coccidioidomycosis is often achieved serologically by the detection of antibodies against fungal antigens. While several serologic tests are available for coccidioidomycosis, all of them are performed in a laboratory setting causing delays in diagnosis and therapeutic intervention. Point-of-care testing offers the ability to provide a shorter time to result by avoiding specimen send-out, minimizing processing steps, and employing expeditious immunochemical techniques. A preliminary trial of a rapid anti-coccidioidal antibody lateral flow assay (LFA) using fingerstick blood was

Of 495 patients reported in a large urban histoplasmosis outbreak, we studied 276 whose serologic tests were done in a single laboratory. Serologic test results were positive in 96% of these patients (compared with less than 5% of controls from an endemic area), cultures were positive in 22%, and special stains in 19%. The immunodiffusion test results were negative in 13% of patients who had positive findings by complement fixation, and 1% had positive results only by immunodiffusion. The complement fixation test was almost twice as sensitive as the immunodiffusion test in patients with subcli

Serologic tests are valuable in the management of patients with suspected or confirmed coccidioidomycosis. Qualitative tests (immunodiffusion, enzyme immunoassay, or latex particle agglutination) permit detection in the serum of the major antibody responses-coccidioidal IgM in early coccidioidomycosis, and complement fixing (CF) IgG, which appears later and is more persistent. Quantitation of the level (titer) of coccidioidal IgG is useful in prognosis and diagnosis. The preferable antigen for these tests is coccidioidin from the hyphal phase of Coccidioides immitis. When coccidioidal disease
