Embryonic exposure to ambient concentrations of PFAS impairs the innate immune response in zebrafish and fathead minnow.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous environmental contaminants associated with immunotoxicity; however, their effect on innate immune function in fish at ambient concentrations remains unclear. Perfluorooctane sulfonic acid (PFOS) and perfluorohexane sulfonic acid (PFHxS) are legacy PFAS prevalent in aquatic ecosystems and have been well studied in zebrafish at high concentrations.  The present study evaluated the effects of embryonic exposure (morula through 2 days post hatch) to environmentally relevant concentrations of PFOS or PFHxS (∼0.8-800 ng/L) on the innate immun

Molecular interactions and biomarker responses to pharmaceutical contaminants in aquatic species.

Pharmaceutical residues are increasingly detected in freshwater systems, yet mechanistic links between sub-lethal exposure and organ-level injury in fish remain limited. This study assessed 21-day effects of carbamazepine (CBZ), diclofenac (DCF) and fluoxetine (FLX) on Labeo rohita using integrated hepatic and renal biomarker panels supported by molecular docking. Fish were exposed to sub-lethal concentrations under semi-static conditions, and responses were quantified using standard biochemical assays. Hepatic markers included cytochrome P4501A activity (CYP1A/EROD), uridine diphosphate-glucu

Morphological and molecular assessment of muscle metacercariae infecting tench Tinca tinca from fish farms and wild populations in Germany.

Fish can be infected with metacercariae (the final larval stage) of different species of potentially zoonotic digenetic trematodes (flukes). The fish-borne zoonotic trematodes thus compromise food safety and present a major threat for human health. Reducing the risk of human infections requires careful assessment and accurate taxonomic identification of these parasites. Here, we analysed metacercariae in muscle tissue of tench (Tinca tinca), sampled between March and September 2022 from three fish farms and three natural waterbodies in Germany. Whenever possible, we combined morphological and

m6A Methylation Mediated Autophagy and Nucleotide-Binding Oligomerization Domain-like Receptors Signaling Pathway Provides New Insight into the Mitigation of Oxidative Damage by Mulberry Leaf Polysaccharides.

m6A methylation modification is an important genetic modification involved in biological processes such as sexual maturation, antibacterial, and antiviral in aquatic animals. However, few studies have been conducted in aquatic animals on the relationship between m6A methylation modification and autophagy-inflammation induced by lipid metabolism disorders. In the present study, a high-fat (HF) group and HF-MLP group (1 g mulberry leaf polysaccharides (MLPs)/1 kg HF diet) were set up. The mid-hind intestines of Megalobrama amblycephala juveniles from the two groups were collected for MeRIP-seq a

Small intestinal flukes of the genus Metagonimus (Digenea: Heterophyidae) in Europe and the Middle East: A review of parasites with zoonotic potential.

The heterophyid trematode Metagonimus romanicus (Ciurea, 1915) (Digenea) is redescribed on the basis of type material from domestic dogs (Canis familiaris) in Romania, vouchers from experimentally infected cats (Felis catus) and adults recovered from golden hamsters (Mesocricetus auratus) infected with metacercariae from scales of chub (Squalius cephalus) and common nase (Chondrostoma nasus) (Cypriniformes: Leuciscidae) in Hungary. This trematode, endemic to Europe and neighbouring regions (northwestern Türkiye), was previously misidentified as M. yokogawai (Katsurada, 1912), a zoonotic parasi

Bioconcentration of Per- and Polyfluoroalkyl Substances and Precursors in Fathead Minnow Tissues Environmentally Exposed to Aqueous Film-Forming Foam-Contaminated Waters.

Exposure to per- and polyfluoroalkyl substances (PFAS) has been associated with toxicity in wildlife and negative health effects in humans. Decades of fire training activity at Joint Base Cape Cod (MA, USA) incorporated the use of aqueous film-forming foam (AFFF), which resulted in long-term PFAS contamination of sediments, groundwater, and hydrologically connected surface waters. To explore the bioconcentration potential of PFAS in complex environmental mixtures, a mobile laboratory was established to evaluate the bioconcentration of PFAS from AFFF-impacted groundwater by flow-through design.

Allele-specific expression variation at different ploidy levels in Squalius alburnoides.

Allopolyploid plants are long known to be subject to a homoeolog expression bias of varying degree. The same phenomenon was only much later suspected to occur also in animals based on studies of single selected genes in an allopolyploid vertebrate, the Iberian fish Squalius alburnoides. Consequently, this species became a good model for understanding the evolution of gene expression regulation in polyploid vertebrates. Here, we analyzed for the first time genome-wide allele-specific expression data from diploid and triploid hybrids of S. alburnoides and compared homoeolog expression profiles o

High prevalence of zoonotic trematodes in roach (Rutilus rutilus) in the Gulf of Finland.

The intention to increase roach (Rutilus rutilus) consumption is in focus for ecological and economic reasons in Finland. However, its safety as food has not been considered comprehensively. We collected and artificially digested 85 roach halves originating from the south-eastern coast of Finland, and found trematode metacercariae in 98.8% of the samples. Based on polymerase chain reaction (PCR) and sequencing of amplicons generated from the ITS2 gene region, zoonotic parasites of the family Opistorchiidae were identified as Pseudamphistomum truncatum and Metorchis bilis, and also non-zoonotic

Induction of viral interference by IPNV-carrier cells on target cells: A cell co-culture study.

IPNV is a salmonid birnavirus that possesses the ability to establish asymptomatic persistent infections in a number of valuable fish species. The presence of IPNV may interfere with subsequent infection by other viruses. In the present study we show that an IPNV-carrier cell line (EPCIPNV) can induce an antiviral state in fresh EPC by co-cultivating both cell types in three different ways: a "droplet" culture system, a plastic chamber setup, and a transmembrane (Transwell®) system. All three cell co-culture methods were proven useful to study donor/target cell interaction. Naïve EPC cells gro

Development and validation of species-specific molecular diagnostic tool for Opisthorchis felineus (Digenea, Opisthorchiidae) metacercariae.

Opisthorchis felineus (family Opisthorchiidae) is a parasitic flatworm representing a serious threat to humans in some countries. Opisthorchiasis occurs after consumption of raw or undercooked cyprinid fish infected by the metacercarial stage of the parasite. Due to its small size, detection of the parasite in fish fillet is time-consuming and difficult. Furthermore, isolated metacercariae can be identified to genus but not to species level using morphological features and molecular techniques are necessary. In this work, we describe the development of primers for a diagnostic PCR amplificatio

Influence of water quality on the accumulation of methyl 203mercury in gill tissue of minnow (Phoxinus phoxinus).

Effects of pH and concentrations of Cl and Ca on the uptake of methyl mercury (MeHg) in the gills of the minnow (Phoxinus phoxinus) were studied. Chloride concentration and pH in the water affect the speciation of MeHg. Ca was included because it affects the permeability of the gills and could therefore indirectly affect the amount of MeHg accumulated in the tissue. The species formed differ in hydrophobicity, as reflected in their octanol/water partition coefficients (Pow). Both a reduction in pH and an increase in Cl- concentration increased the Pow of MeHg. Ca had no effect on speciation. T

Day-night rhythms of shoaling behavior in goldfish: opioid and pineal involvement.

Intracerebroventricular (ICV) administration of the opioid peptide, beta-endorphin (5.0 pg/g) to goldfish, Carassius auratus, significantly increased the cohesiveness and duration of shoaling ('bout' length) in shoals of five fish, as well as decreasing the latency of shoal formation in response to an external disturbance, while a higher dose of beta-endorphin (15 pg/g) decreased shoaling. There were day-night rhythms in shoaling and in the extent of the facilitatory effects of beta-endorphin (5.0 pg/g) on shoaling behavior, the fish displaying significantly greater shoaling responses in the d

Inhibition of non-neuronal cell proliferation in the goldfish visual pathway affects the regenerative capacity of the retina.

Proliferating cells associated with the visual pathway were found in the present study to affect the regenerative capacity of the goldfish retina following optic nerve injury. The contribution of these cells to the process of regeneration was investigated in the goldfish visual system by reducing their proliferation in the optic tract and tecta, using X-irradiation. The regenerative ability of the retina was then evaluated by the following parameters: sprouting from retinal explants, protein synthesis in the retina and accumulation of radiolabeled transported components in the tectum. X-irradi

Structure of the mitral cell in the olfactory bulb of the goldfish (Carassius auratus).

The mitral cell in the olfactory bulb of the goldfish was examined by means of light microscopy, high-voltage electron microscopy, and conventional electron microscopy. Mitral cells are located rather diffusely throughout the glomerular and plexiform layers. They do not make their own discrete layer. The cell bodies are rounded or triangular, and are about 10-25 micrometers in diameter. In Golgi-impregnated material, thick cylindrical dendrites can be seen arising from the cell bodies and branching in the glomerular layer. Dendritic branches of some cells make two or more rather compact tufts,

Ultrastructure of the Mauthner axon collateral and its synapses in the goldfish spinal cord.

The ultrastructure of the synapses formed by the Mauthner axon collateral (MAC) was examined in the goldfish spinal cord. All such collaterals form axo-dendritic chemical synapses with dendrites of motor and interneurons. Besides these chemical synapses, contacts have been found between some MAC and axons of interneurons, which can be characterized as gap junctions with presumed electrical transmission. All cellular processes contacted by the MAC contain 'dense bands' of material in their cytoplasm. These 'dense bands' are absent from other parts of the fish C.N.S. These observations and the p

Fine structure of the olfactory bulb in the goldfish, Carassius auratus.

Ultrastructural studies have aided in identifying the principal cell types and the layers of the olfactory bulb in goldfish. The concentrically arranged four layers are: the primary olfactory nerve layer (most peripheral), the glomerular layer, the mitral cell layer, and the granule cell layer (deepest). Three types of cell bodies were differentiated by their size and cytoplasmic organelles: mitral cells, granule cells and short axon cells. The axons of the primary olfactory nerve formed synaptic contacts with mitral cell dendrites in the glomeruli. Reciprocal synapses were observed between th

Central projections and motor nuclei of the facial, glossopharyngeal, and vagus nerves in the mormyrid fish Gnathonemus petersii.

Most of the information about the anatomy of the fish's cranial nerves was collected in the first two decades of this century. Experimental analysis of the VIIth, IXth, and Xth cranial nerves by modern tract tracing techniques started about 20 years ago. Several species have been investigated to date, including one species of Agnatha (Myxinoidea), two species of elasmobranchs, and species of some orders of Teleostei like Cyprinidae, Siluriformes, Perciformes, and Gadidae.
