BACKGROUND: Extracting DNA for the purpose of identification from bone material, especially for old samples or from extreme environmental conditions, has always been a challenging task in forensic genetics. At our forensic institute with two locations between the Baltic Sea and the North Sea, found marine bones are regularly submitted for evaluation and analyzed using molecular biology methods.
METHODS: An improvement to commercially available bead-based methods routinely used for the bone investigations and often yielding poor or no DNA, was therefore aimed for. In the present study, an over

Background/Objectives: Sexual offences remain a global challenge, disproportionality affecting developing and conflict-stricken countries. Differential DNA extraction (DDE) is standardly applied to intimate swabs collected in these cases to separate and purify sperm and epithelial fractions ahead of DNA profiling. Whilst DNA purification steps using magnetic bead technology are routinely automated in general forensic workflows, the separation step within DDE workflows usually relies on centrifugation, which is challenging to automate in a high-throughput manner. This proof-of-concept study aim

Inference of genetic relationships from genomic data is central to applications in human, plant, and conservation genetics, with investigative genetic genealogy (IGG) increasingly used in forensic and population studies. While high-quality DNA samples enable robust detection of identity-by-descent (IBD) segments, low-quality or low-quantity sources such as telogen hairs pose challenges due to genotyping errors and incomplete genetic data (profiles). In this study, we 1) introduce a conditional simulation framework that generates relatives of varying degrees based on high-quality donor genotype

DNA recovery from fired cartridge cases (FCCs) found at a crime scene is complicated by the limited DNA amounts typically available for collection. While this challenge is unavoidable, recovery rates may be improved by optimizing handling and packaging processes to better preserve the available DNA. Here we compared existing methods of collecting and packaging FCCs at a crime scene and assessed the utility of a novel cartridge collection device to determine which combination best preserves DNA integrity and amounts. FCCs loaded with touch DNA or blood and some DNA-free FCCs were collected and

In this case report, we describe a sexual assault incident in which the male victim's seminal fluid contained no sperm cells, as indicated by sperm cell staining and microscopic screening, and DNA profiling results from the non-sperm cell fraction showed a major/minor DNA mixture that could be interpreted as female and male. DNA profiling of a sample from a disposable drinking cup used by the victim at the crime scene provided a single source profile, and showed a 2:1 imbalance between the heights of the X and Y chromosomes, respectively. The victim's DNA reference sample showed a similar imba

Sexual assault cases are regularly encountered in forensic laboratories. The victim's underwear and other clothes are often submitted with the intention of retrieving the offender's DNA. We examined six sexual assault scenarios; removing underwear, removing brassiere, digital penetration of the vagina from the front, grabbing breasts over the top of the brassiere, digital penetration of the vagina from the rear, and grabbing the breasts under the brassiere from the rear. For each scenario we used a dummy weighing approximately 45 kg to mimic a victim and recorded where the underwear was contac

The ability to obtain DNA profiles from trace biological evidence is routinely demonstrated with so-called 'touch DNA evidence', which is generally perceived to be the result of DNA obtained from shed skin cells transferred from a donor's hands to an object or person during direct physical contact. Current methods for the recovery of trace DNA employ swabs or adhesive tape to sample an area of interest. While of practical utility, such 'blind-swabbing' approaches will necessarily co-sample cellular material from the different individuals whose cells are present on the item, even though the ind

An increase in cutaneous and visceral leishmaniasis cases has been reported in recent years in the state of Mato Grosso do Sul, Brazil, and little is known to date about their etiological agents. An investigation into natural Leishmania infection of sand flies captured in this state between December 2003 and August 2004 was carried out. Mini-exon sequences were used as targets to identify Leishmania, and an RFLP technique was employed for those identified as belonging to the Viannia subgenus. Calculation of the minimal infection rate (MR) revealed that 1.6% of sand flies captured in the forest

In order to obtain fetal cells (e.g., for paternity cases) after abortion, the centrifugation of mountant (in our case formalin) may be tried when the DNA examination of the fixed tissue itself gives limited or no (or not enough) information. The fixed tissue was microscopically negative for fetal cells and gave no satisfactory results when examined for DNA. Centrifugation of approximately 50 mL of reddish colored formalin resulted in a cell pellet that was examined for DNA, which gave enough information to confirm a case of sexual abuse.

The distribution of the 19 currently known genospecies of Acinetobacter on human skin, i.e. forehead, forearm and toe webs, was determined. Three selective media were compared for their specificity for all genospecies of Acinetobacter. A minimal-salts agar supplemented with 1% acetate proved to be more efficient than the Leeds medium for the isolation of most genospecies in mixed culture with other bacterial species. Acinetobacter isolates were provisionally identified using biochemical tests and the DNA transformation assay of Juni. Genospecies identification was performed using amplified rib

Skin cells (keratinocytes) and biological materials originating from sweat and sebum that are transferred to surfaces through physical contact are referred to in the forensic literature as touch DNA or trace DNA. Touch DNA plays a critical role in establishing the relationship between the perpetrator, object, and scene, particularly in cases where no visible biological traces are present. However, due to its typically low quantity, degraded nature, and high susceptibility to surface characteristics, the recovery of reliable DNA profiles from touch DNA remains challenging. Consequently, determi

DNA obtained from the crime scene or in the form of trace evidence is one of the most crucial pieces of evidence to individualize an assailant. During a violent physical assault, DNA, in the form of epithelial cells or blood due to violent scratching, is deposited in the hyponychium of the fingernail. This trace DNA is recovered and extracted from the fingernail debris by nail swabbing, and genetic profiling is done from the extracted DNA. The objective of the study is to evaluate the amount of exogenous DNA that persists in fingernail debris and the difference in recovery rate of the biologic

Plastic bags, such as ziplock bags, have been used to transport illicit materials worldwide; however, very few studies have tried to optimize the recovery of DNA from these items. This study reports on the best combination of swabs and moistening solution for the greatest recovery of cellular material from ziplock bags. Five swabs, two different variations of Copan Diagnostics nylon 4N6FLOQSwabs, one Medical Wire rayon DRYSWAB, one IsoHelix rayon swab, and one Livingstone cotton swab, were evaluated with two moistening solutions, Triton X-100 in either distilled water or isopropanol. Extracting DNA from degraded human remains poses a challenge for any forensic genetics laboratory, as it requires efficient high-throughput methods. While little research has compared different techniques, silica in suspension has been identified in the literature as the best method for recovering small fragments, which are often present in these types of samples. In this study, we tested five DNA extraction protocols on 25 different degraded skeletal remains. Including the humerus, ulna, tibia, femur, and petrous bone. The five protocols were organic extraction by phenol/chloroform/isoamyl al

The authors present the case of a 61-year-old man found dead in an agricultural plot. The first investigation of the scene revealed the corpse laid face up in a spot of partially dried blood, next to an olive tree. His face, arms, legs, and abdomen showed signs of severe contusion and laceration of dogs' bite wounds. Next to the victim, an olives bin had been found overturned on the ground. A multi-disciplinary approach, including crime scene analysis, autopsy findings, veterinary animals review, odontologist bite mark study, and forensic genetics DNA correlations, was performed. Recent advances in genetic characterisation of Trichomonas vaginalis isolates show that the extensive clinical variability in trichomoniasis and its disease sequelae are matched by significant genetic diversity in the organism itself, suggesting a connection between the genetic identity of isolates and their clinical manifestations. Indeed, a high degree of genetic heterogeneity in T vaginalis isolates has been observed using multiple genotyping techniques. A unique two-type population structure that is both local and global in distribution has been identified, and there is evidence of recombi

Cryptococcosis is a fungal infection caused by yeasts of the genus Cryptococcus, with Cryptococcus neoformans and Cryptococcus gattii as the primary pathogenic species. This disease is a threat to immunocompromised patients, especially those who have AIDS. However, the disease has also been described in healthy individuals. The tests used to identify these microorganisms have limitations that make final diagnosis difficult. However, currently there are specific gene sequences that can be used to detect C. neoformans and C. gattii from clinical specimens and cultures. These sequences can be use

PURPOSE OF REVIEW: Leishmaniasis has a wider geographical distribution than before. This review focuses on molecular epidemiological studies of new foci of cutaneous leishmaniasis in South and Southeast Asia.
RECENT FINDINGS: In Pakistan, the main causative agent in southern dry areas was determined as Leishmania major followed by Leishmania tropica. There was no correlation between the skin lesion types and the causative species. In the western Indian Himalayas, L. tropica and Leishmania donovani have been reported as the causative pathogens. In Nepal, molecular methods revealed the first ca
