Progress in Research on Macrophage Polarization Mechanisms and Targeted Therapies in Staphylococcus aureus Infections.

Staphylococcus aureus is a clinically prevalent, Gram-positive pathogen that can cause multiple severe infections. Macrophage polarization plays a central role in host defense and inflammatory regulation, with its M1/M2 dynamic equilibrium directly determining infection outcomes. The M1 phenotype eliminates pathogens through pro-inflammatory responses in the early phase, but excessive activation readily leads to tissue damage. In contrast, the M2 phenotype suppresses inflammation and promotes healing during the repair phase, although it may become a pathogen refuge in chronic infections. Recen

Electrocatalytic Coupling of Nitrate and Formaldehyde for Hexamethylenetetramine Synthesis via C-N Bond Construction and Ring Formation.

Hexamethylenetetramine (HMTA) is extensively used in the defense industry, medicines, food, plastics, rubber, and other applications. Traditional organic synthesis of HMTA relies on ammonia derived from the Haber process at high temperatures and pressures. In contrast, electrochemical methods enable a safe and green one-pot synthesis of HMTA from waste NO3-. However, HMTA synthesis through the electrochemical method is challenging owing to the complex reaction pathways involving C-N bond construction and ring formation. In this study, HMTA was efficiently synthesized over electrochemical oxida

Targeting oxidative stress in preeclampsia.

Preeclampsia is a complex condition characterized by elevated blood pressure and organ damage involving kidneys or liver, resulting in significant morbidity and mortality for both the mother and the fetus. Increasing evidence suggests that oxidative stress, often caused by mitochondrial dysfunction within fetal trophoblast cells may play a major role in the development and progression of preeclampsia. Oxidative stress occurs as a result of an imbalance between the production of reactive oxygen species (ROS) and the capacity of antioxidant defenses, which can lead to placental cellular damage a

Direct antimicrobial effects of chemokines on Cryptococcus spp, with special emphasis on a 'CXC' chemokine.

Cryptococcus species are ingenious human pathogens that are widespread globally. They continue to cause over 200,000 deaths per year. Presently due to the rise in resistance and therapy failure, it is necessary to shift the focus to an alternate therapeutic strategy against this pathogen. One promising approach is to emphasize the host defense system in order to develop more precise and customized treatment strategies. In this regard, research has revealed that interferon-γ-inducible CXCL10 chemokine, amongst other chemokines spanning both CXC and CC categories, has a direct killing effect in

Antifungal Peptides of the AFP Family Revisited: Are These Cannibal Toxins?

The emergence and spread of pathogenic fungi resistant to currently used antifungal drugs represents a serious challenge for medicine and agriculture. The use of smart antimicrobials, so-called "dirty drugs" which affect multiple cellular targets, is one strategy to prevent resistance. Of special interest is the exploitation of the AFP family of antimicrobial peptides, which include its founding member AFP from Aspergillus giganteus. This latter is a highly potent inhibitor of chitin synthesis and affects plasma membrane integrity in many human and plant pathogenic fungi. A transcriptomic meta

Confirmed malaria cases among active component U.S. Army personnel, January-September 2012.

Of 26 cases of malaria reported among active component U.S. Army personnel during January through September 2012, 16 were laboratory-confirmed according to electronic medical records. Medical records and responses on postdeployment health assessment questionnaires were used to assess demographic and clinical characteristics, adherence to malaria prevention measures, and compliance with prescriptions for chemoprophylaxis. All but two cases were confirmed by peripheral blood smears. Twelve cases were caused by Plasmodium vivax, one by P. falciparum, and three unspecified diagnoses were deemed li

Crosstalk in Silence: PIWIL/piRNA in ncRNA Hubs for Immune Gene Regulation and Epigenetic Antiviral Defense.

Recent studies indicate that PIWI (P-element-induced WImpy testis) like proteins and their cognate PIWI-interacting RNAs (piRNAs), initially identified in germline cells for protecting genomic stability by transposon silencing, also exert multiple regulatory functions in somatic and immune systems. Additionally, PIWIL/piRNA pathways are linked to innate antiviral defense and immune signaling pathway regulation. This review systematically compiles the reports on the immunological functions of noncoding RNAs (ncRNAs) and delineates the potential molecular processes and functional characteristics

Environmental adaptation and functional diversity of calcin peptides from the venom of East Asian scorpions.

Toxins as naturally occurring hazardous substances, have evolved as a defense mechanism in organisms for survival. The expression of toxins is profoundly influenced by environmental factors, and the investigation of their correlation holds significant implications for understanding toxin-induced harm and developing protective measures. Scorpions are a significant venomous species, and the relationship between the variability in scorpion toxin expression and environmental factors remains unclear. By conducting transcriptome profiling of scorpion samples collected from eight distinct climatic an

Detoxifying Effects of a Phenolic-Rich Thunbergia laurifolia Water Extract Against Chlorpyrifos-Induced Oxidative Stress: Evidence from an Integrated In Vitro and In Vivo Study.

Thunbergia laurifolia has long been used in traditional medicine for detoxification; however, the biological basis of its detoxifying effects remains poorly understood. This study investigated the detoxifying effects of a phenolic-rich aqueous extract of T. laurifolia using integrated in vitro and in vivo approaches. Caffeic acid (CA) and rosmarinic acid (RA) were used as marker compounds for extract standardization. The aqueous extract was evaluated for antioxidant, acetylcholinesterase, and apoptosis-modulating activities in vitro, followed by evaluation in a chlorpyrifos-induced oxidative s

Molecular Pathways Implicated in Radioresistance of Glioblastoma Multiforme: What Is the Role of Extracellular Vesicles?

Glioblastoma multiforme (GBM) is a primary brain tumor that is very aggressive, resistant to treatment, and characterized by a high degree of anaplasia and proliferation. Routine treatment includes ablative surgery, chemotherapy, and radiotherapy. However, GMB rapidly relapses and develops radioresistance. Here, we briefly review the mechanisms underpinning radioresistance and discuss research to stop it and install anti-tumor defenses. Factors that participate in radioresistance are varied and include stem cells, tumor heterogeneity, tumor microenvironment, hypoxia, metabolic reprogramming, t

Potential of natural products in the treatment of myocardial infarction: focus on molecular mechanisms.

Although conventional drugs are widely used in the prevention and treatment of cardiovascular disease (CVD), they are being used less frequently due to concerns about possible side effects over the long term. There has been a renewed research interest in medicinal plant products, and their role in protecting the cardiovascular system and treating CVD, which are now being considered as potential alternatives to modern drugs. The most important mechanism causing damage to the myocardium after heart attack and reperfusion, is increased levels of free radicals and oxidative stress. Therefore, trea

[Anti-inflammatory drugs in ORL].

The idea of anti-inflammatory therapy in medicine arose in the 1950's with the development of corticosteroids. Prior to this time the treatment of phlogistic pathologies was aimed at achieving analgesic, antipyretic and diaphoretic effects. Cortisone and its derivatives have spurred the setting up of a series of pharmacological tests to study their anti-inflammatory activity and to verify their clinical efficacy in rheumatology. The side effects of corticosteroids have promoted research into non steroid anti-inflammatory drugs (NADs). A new pharmacological class has been created which includes

Hippocratic ideal, Faustian bargain and Damocles' sword: erosion of patient autonomy in obstetrics.

Respect for patient autonomy remains a foundational principle guiding the ethical practice of medicine-a mission first articulated by Hippocrates. Damocles, another figure from ancient Greece, provides a useful parable for describing performance under distress: Damocles loses his desire for opulence and power when he notices a sword dangling precariously above his head. Contemporary obstetricians deciding whether to forestall or impose major abdominal surgery on parturients entrusted to their care struggle valiantly in the chasm dividing Hippocratic idealism from the economic realism driven by
