Pharmacokinetics, toxicity, and activity of intravenous dextran sulfate in human immunodeficiency virus infection.

Polysulfated polysaccharides are attractive candidates for antiviral drug development because of their potent in vitro activities against human immunodeficiency virus (HIV), herpesviruses, and other enveloped viruses. To determine the potential anti-HIV activity of a prototypical polysulfated polysaccharide, we administered the maximally tolerated dose of dextran sulfate by continuous intravenous infusion to 10 subjects with symptomatic HIV infection for up to 14 days. Since parenteral dextran sulfate is an anticoagulant, the infusion was adjusted to produce the greatest acceptable increase in

Concentration of Babesia bigemina-infected erythrocytes using a dextran sulphate affinity column.

Babesia bigemina-infected erythrocytes preferentially bind to dextran sulphate affinity columns. Subsequent elution yields suspensions containing up to 95% infected erythrocytes. Preliminary immunoblotting studies indicate a parasite antigen of 35,000 mol. wt might be implicated in the binding.

Perspectives on clinical trials for hypertonic saline/dextran solutions for the treatment of traumatic shock.

Animal studies with hypertonic solutions suggest that they can achieve resuscitation of hypovolemic shock with extremely small volumes. Such small volume resuscitation might be ideal in the field treatment of injured patients. Our studies to date, with 60 patients entered into a prospective, randomized, placebo-controlled, and double-blind clinical trial, suggest that the use of a 7.5% NaCl/Dextran 70 solution increases blood pressures during transport. The solutions have been safe, and we have encountered no adverse side effects from their use. Survival rates to date favor use of the solution

The effect of a new intraocular irrigating solution containing dextran on rabbit corneal endothelium.

This study investigated the effect of a balanced salt solution for intraocular irrigation containing 2% dextran 40. The effect of this solution was compared to that of a normal saline solution (0.9%) and balanced salt solution (Alcon). Following incubation of rabbit corneas for up to 72 h at 4 degrees C, the incubated corneas were studied by staining with Nitroblue Tetrazolium and by scanning electron microscopy. Insignificant changes occurred during the first 24 h of incubation. However, corneas stored in the dextran containing solution showed slightly smaller endothelial cells with tight jun

Enhancement of survival in acute experimental fowl typhoid in chicks by the administration of iron dextran.

Chicks aged 15 days were infected orally with 10(6-4) living Salmonella gallinarum. Iron from iron dextran or ferric ammonium citrate (DFe and CFe respectively), in doses of 2-0 or 0-4 mg/kg given intramuscularly at the time of infection, had no effect on subsequent mortality compared with infected controls. Increasing the amount of CFe to the maximum (20 mg/kg) that was non-toxic intramuscularly was also ineffective but when the dose of DFe was increased to 20 or 50 mg/kg the survival rate rose sharply. If given at other times in relation to the time of infection, or more frequently, 50 mg/kg

Effect of dextran and hyaluronic acid on the development of postoperative peritoneal adhesions in experimental animals.

The development of postoperative peritoneal adhesions was studied in rats and rabbits, the frequency of adhesions in the experimental model used being very high. In the development of an adhesion, fibrin seems to be an important contributor to the bridge between different tissues. Dextran, which modifies the fibrin network and makes it more susceptible to lysis, was used as a possible prophylactic agent, but we found no difference between treated and control groups. It is concluded that the stimulus for fibrin formation in the peritoneal adhesions using this atraumatic model was too strong to

Studies on passive transfer of anti-dextran and anti-egg albumin reactivity in the rat.

We were unable, passively, to transfer reactivity against dextran to the skin or peritoneal mast cells of non-dextran-reactive rats, by using serum or material eluted at pH 3 from mast cells of spontaneously dextran-reactive rats. When the dextran-reactive donor rats had also been immunized against egg albumin (EA) with pertussis vaccine (inducing IgE anti-EA antibody), passive sensitivity against EA (but not against dextran) could easily be transferred. The results indicated that the anti-dextran reactivity is not due to IgE antibody. Systemic reactions against dextran and EA differed in patt

The Influence of Synthesis Parameters on the Properties of Dextran-Based Hydrogels for Colon-Targeted Antitumor Drug Delivery Part I: Room Temperature Synthesis of Dextran/Inulin Hydrogels for Colon-Targeted Antitumor Drug Delivery.

This research successfully developed novel hydrogels composed of methacrylated dextran and inulin for targeted drug delivery in colorectal cancer therapy. The formulation exploits the natural degradation of both biopolymers by the large intestine's microflora. A key achievement was the development of a room-temperature free radical polymerization synthesis method. The study thoroughly investigated how varying inulin content (10 and 20 wt%) influenced the hydrogels' properties. The formulation with 20 wt% inulin exhibited the highest swelling ability at both pH 3 and pH 6, and consequently the

Sustained elution of amikacin, clindamycin, and vancomycin from a biodegradable cross-linked dextran gel.

Osteomyelitis, surgical site infections, implant infections, abscesses, and other localized infections can be difficult to treat due to poor vascularity, biofilm formation, and the presence of antibiotic-resistant bacteria. These infections result in significant morbidity and mortality in human and veterinary patients. Local delivery of high concentrations of antibiotics that are released over a prolonged period of time can overcome these issues, and avoid side effects of systemic antibiotics. In vitro release kinetics of amikacin, clindamycin, and vancomycin from a cross-linked dextran (CLD)

Optimization of dextran sulfate sodium-induced colitis and associated mortality in BALB/c mice.

Dextran sulfate sodium (DSS)-induced colitis in BALB/c mice has revealed underlying mechanisms of colitis pathology. This study aimed to optimize the DSS dosage to improve reproducibility while ensuring high survival rates. Mice were treated with 2 and 3% DSS solutions in drinking water daily for 7 days (days 1-7), followed by water (8-14 days). Disease activity index (DAI) and survival percentage were monitored over 14 consecutive days. On the last day, intestinal permeability was assessed by Evans blue assay, and colon length was measured. After sacrifice, colon samples were collected to eva

Molecular and Cellular Mechanisms of Fibrosis of Abdominal Adhesions under the Influence of Oxidized Dextran in the Experiment.

Formation of abdominal adhesions is accompanied by the synthesis of collagen monomers by fibroblasts and deposition of type I and III collagens. This process is regulated by various factors, including TIMP-1, MMP-1, TGFβ, IL-10, and IFNγ. Oxidized dextran prevents fibrosis, adhesion formation, affecting the functional state of macrophages and fibroblasts. Collagen formation and fibroblasts in abdominal adhesions of Wistar rats were studied against the background of intraperitoneal administration of oxidized dextran. Electron and light microscopy and immunohistochemical method (collagen I, TIMP

Reducing dextran sodium sulfate interference with gene expression quantification in a mouse model of colitis.

Gene expression analysis via reverse transcription quantitative real-time polymerase chain reaction (qPCR) can be inhibited by various substances, including dextran sodium sulfate (DSS), a chemical commonly used to induce intestinal inflammation in animal models. Ensuring elimination and reduction of qPCR interference in tissues from laboratory animals following oral administration of DSS is critical and may improve the power of experimental tests. While methods for DSS removal have been reported, their effectiveness varies depending on the animal model, extraction techniques, DSS concentratio

Oleic Acid Integrated Acetalated Dextran Nanoparticles for Enhanced Chemotherapeutic Delivery to the Bone Marrow.

Bone marrow targeted delivery of chemotherapeutics remains critical in the treatment of hematological malignancies such as acute lymphoblastic leukemia (ALL). Current drug delivery platforms to treat ALL do not specialize in enhancing drug accumulation in the bone marrow, often leading to suboptimal therapeutic responses and off-target side effects. Here, we developed a polymeric lipid hybrid nanoparticle (PLHP) platform through integration of a pH-responsive acetalated dextran (Ac-Dex) polymer and oleic acid (OA), an endogenous bone marrow-rich fatty acid, for enhanced accumulation of therape

Genistein-loaded, quaternized dextran/κ-carrageenan gels for bone regeneration in osteoporotic conditions.

Osteoporosis is a metabolic bone disease marked by the decreased bone mass from impaired bone homeostasis, increasing risk of fractures, particularly in postmenopausal women. The underlying cause of postmenopausal osteoporosis is a sharp decline in estrogen levels, crucial for the maintenance of bone health. Current treatment modalities are associated with drawbacks leading to growing need for alternative approaches for bone tissue regeneration. Various strategies have been explored but they fail to completely address the challenge of oxidative stress, hormonal imbalance, and inflammation. Bio
