Discovery of molecular glue degrader for treatment of IMiDs-resistant multiple myeloma based on GLUT-mediated tumor targeting strategy.

Based on the tumor Warburg effect and the strategy of GLUT-mediated drug transport, we have successfully discovered a promising molecular glue immunomodulatory drug (IMiD) candidate that exhibits excellent dual-targeting capability in tumor selective drug uptake, immunotherapy and combating lenalidomide (LEN) and pomalidomide (POM) resistance in multiple myeloma. The structure-activity relationship study culminated in the identification of the lena- and pomalidomide-glycoconjugates (1 and 20) with potent antitumor activity in cancer cells overexpressing GLUT1, especially in the highly resistan

Role of hydrogen sulfide in bacterial antibiotic tolerance: from mechanisms to therapeutic targeting.

Hydrogen sulfide (H2S) is fundamentally a gaseous signaling molecule in bacteria. Recent studies have reported that H2S is a key mediator of bacterial antibiotic tolerance, making it a promising strategic target for overcoming tolerance, eliminating persisters, and restoring antibiotic efficacy. We systematically review recent advances in H2S-mediated multidrug tolerance mechanisms, highlighting three core pathways: iron chelation for antioxidant defense, reactive sulfur species-mediated antibiotic inactivation and efflux pump activation, and metabolic dormancy induced by respiratory inhibitio

Antimicrobial resistance pattern due to oxygen gap at high altitudes.

Over the past 10 years, microbial resistance has seriously threatened human life and health, and the treatment of multidrug-resistant bacteria remains a challenge for clinicians, pharmacists, and infectious disease physicians. Bacterial resistance is affected by a variety of factors, such as the environment, economy, and drug abuse. This review compared the differences in bacterial resistance rates between high- and low-altitude areas and explored the relevant mechanisms of bacterial resistance in the low-oxygen environment of plateaus, providing new clinical research ideas for curbing the occ

Antimicrobials in Orthopedic Infections: Overview of Clinical Perspective and Microbial Resistance.

Orthopedic infections are challenging pathologies that impose a heavy burden on patients and the healthcare system. Antimicrobial therapy is a critical component of the successful management of orthopedic infections, but its effectiveness depends on patient-, surgery-, drug-, and hospital-related factors. The dramatic increase in the emergence of multidrug-resistant microbial strains necessitates new clinical approaches in order to prevent or limit this phenomenon and to ensure a favorable therapeutic outcome. The present paper reviews the currently available antimicrobial strategies in the ma

Florfenicol administration in piglets co-selects for multiple antimicrobial resistance genes.

Antimicrobial use in food-producing animals such as pigs is a significant issue due to its association with antimicrobial resistance. Florfenicol is a broad-spectrum phenicol antibiotic used in swine for various indications; however, its effect on the swine microbiome and resistome is largely unknown. This study investigated these effects in piglets treated intramuscularly with florfenicol at 1 and 7 days of age. Fecal samples were collected from treated (n = 30) and untreated (n = 30) pigs at nine different time points up until 140 days of age, and the fecal metagenomes were sequenced. The fe

Role of the RNA binding protein IGF2BP1 in cancer multidrug resistance.

The insulin-like growth factor-2 mRNA-binding protein 1 (IGF2BP1), a member of a conserved family of single-stranded RNA-binding proteins (IGF2BP1-3), is expressed in a broad range of fetal tissues, placenta and more than sixteen cancer types but only in a limited number of normal adult tissues. IGF2BP1is required for the transport from nucleus to cytoplasm of certain mRNAs that play essential roles in embryogenesis, carcinogenesis, and multidrug resistance (MDR), by affecting their stability, translation, or localization. The purpose of this review is to gather and present information on MDR

Mapping the genomic landscape of multidrug resistance in Plasmodium falciparum and its impact on parasite fitness.

Drug-resistant Plasmodium falciparum parasites have swept across Southeast Asia and now threaten Africa. By implementing a P. falciparum genetic cross using humanized mice, we report the identification of key determinants of resistance to artemisinin (ART) and piperaquine (PPQ) in the dominant Asian KEL1/PLA1 lineage. We mapped k13 as the central mediator of ART resistance in vitro and identified secondary markers. Applying bulk segregant analysis, quantitative trait loci mapping using 34 recombinant haplotypes, and gene editing, our data reveal an epistatic interaction between mutant PfCRT an

Targeting bacterial outer-membrane remodelling to impact antimicrobial drug resistance.

The cell envelope is essential for survival and adaptation of bacteria. Bacterial membrane proteins include the major porins that mediate the influx of nutrients and several classes of antimicrobial drugs. Consequently, membrane remodelling is closely linked to antimicrobial resistance (AMR). Knowledge of bacterial membrane protein biogenesis and turnover underpins our understanding of bacterial membrane remodelling and the consequences that this process have in the evolution of AMR phenotypes. At the population level, the evolution of phenotypes is a reversible process, and we can use these i

Molecular tools for rapid identification and novel effective therapy against MDRTB/XDRTB infections.

Tuberculosis (TB) is mainly an intracellular infection of the lung alveolar macrophages, and any anti-TB agent must therefore be active at the macrophage. Among the available therapies, isoniazid and rifampicin are the most effective drugs against susceptible Mycobacterium tuberculosis, but they are ineffective against multidrug-resistant TB (MDRTB) strains. Rates of MDRTB in Portugal are the highest in Western Europe, demanding effective measures for their control. Our application of molecular techniques for the early identification of MDRTB assisted in the reduction of these rates. Further e

Epidemiology of antibiotic resistance in Haemophilus influenzae.

During the last 10 years, a continuous increase in the incidence of beta-lactamase-producing Haemophilus influenzae has been observed; in addition, beta-lactamase-negative ampicillin-resistant strains are better identified and more frequently isolated. During the same period, resistance to tetracyclines and chloramphenicol decreased to a very low level. The incidence of resistant bacteria is highly variable according to the site of infection, patient's age, type of Haemophilus, and country or region, according to local epidemiological factors. Follow-up multicenter studies are needed to monito

Key Proteins and Enzymatic Mechanisms Underlying Drug Resistance in Mycobacterium tuberculosis.

Drug resistance in Mycobacterium tuberculosis (Mtb) is a complex phenomenon resulting from multiple coordinated molecular and genetic mechanisms that allow the bacterium to evade antimicrobial agents. Intrinsic resistance primarily stems from the unique architecture of the mycobacterial cell envelope, characterized by a dense, lipid-rich barrier that substantially limits antibiotic penetration and reduces intracellular drug concentrations. Additionally, enzymatic degradation mechanisms further diminish drug efficacy by inactivating antibiotics extracellularly. Acquired resistance is predominan

Antimicrobial resistance and serotype distribution of Streptococcus pneumoniae isolates from Crete, Greece.

Susceptibility to 14 antibiotics was determined for 125 clinical isolates of Streptococcus pneumoniae collected over a 3-year period in Crete, Greece. Twenty-three isolates (18.4%) showed intermediate resistance and 15 (12%) high-level resistance to penicillin. Erythromycin, chloramphenicol, tetracycline, trimethoprim-sulphamethoxazole and sparfloxacin resistance rates were 16.8, 10.4, 19.2, 24.8 and 9.6%, respectively. Multiple resistance was observed in 22 strains. Vancomycin and levofloxacin were the most active agents tested. The most prevalent serotype among penicillin-susceptible pneumoc

Complementary supramolecular drug associates in perfecting the multidrug therapy against multidrug resistant bacteria.

The inappropriate and inconsistent use of antibiotics in combating multidrug-resistant bacteria exacerbates their drug resistance through a few distinct pathways. Firstly, these bacteria can accumulate multiple genes, each conferring resistance to a specific drug, within a single cell. This accumulation usually takes place on resistance plasmids (R). Secondly, multidrug resistance can arise from the heightened expression of genes encoding multidrug efflux pumps, which expel a broad spectrum of drugs from the bacterial cells. Additionally, bacteria can also eliminate or destroy antibiotic molec
