A coupling model of transcranial magnetic stimulation induced electric fields to neural state variables.

Transcranial magnetic stimulation (TMS) is a non-invasive brain stimulation technique used to modulate neural activity, with applications in clinical treatment, diagnostics, and neuroscientific research. TMS targeting the human primary motor cortex (M1) is well studied, aided by experimental readouts from the cerebral cortex, the spinal cord, and activated muscle targets. One key readout is a series of pulses that descend the spinal cord following TMS, called DI-waves. These reflect the output of M1 to the spinal cord and are influenced by TMS parameters such as orientation, strength, and wave

Abnormal myocardial expression of SAP97 is associated with arrhythmogenic risk.

Synapse-associated protein 97 (SAP97) is a scaffolding protein crucial for the functional expression of several cardiac ion channels and therefore proper cardiac excitability. Alterations in the functional expression of SAP97 can modify the ionic currents underlying the cardiac action potential and consequently confer susceptibility for arrhythmogenesis. In this study, we generated a murine model for inducible, cardiac-targeted Sap97 ablation to investigate arrhythmia susceptibility and the underlying molecular mechanisms. Furthermore, we sought to identify human SAP97 (DLG1) variants that wer

Rapid desensitization converts prolonged glutamate release into a transient EPSC at ribbon synapses between retinal bipolar and amacrine cells.

The mechanisms underlying the conversion of prolonged glutamate release from ribbon synapses in bipolar cells to sustained and transient excitatory postsynaptic responses in identified retinal amacrine cells were studied in tiger salamander (Ambystoma tigrinum) retina. A retina slice preparation with whole cell patch recording techniques under voltage- and current-clamp conditions was used to assay the electrical properties of bipolar and amacrine cells. Amacrine cells were categorized into two basic forms: (i) transient amacrine cells that respond to a step of light with a burst of spikes onl

Synaptic mechanisms of forward suppression in rat auditory cortex.

In the auditory cortex, brief sounds elicit a powerful suppression of responsiveness that can persist for hundreds of milliseconds. This forward suppression (sometimes also called forward masking) has usually been attributed to synaptic (GABAergic) inhibition. Here we have used whole-cell recordings in vivo to assess the role of synaptic inhibition in forward suppression in auditory cortex. We measured the excitatory and inhibitory synaptic conductances elicited by pairs of brief sounds presented at intervals from tens to hundreds of milliseconds. We find that inhibitory conductances rarely la

Factors explaining heterogeneity in short-term synaptic dynamics of hippocampal glutamatergic synapses in the neonatal rat.

1. Quantal release from single hippocampal glutamatergic (CA3-CA1) synapses was examined in the neonatal rat during a 10 impulse, 50 Hz stimulus train. These synapses contain a single release site only, thus allowing for an analysis of frequency facilitation/depression at the single release site level. 2. These synapses displayed a considerable heterogeneity with respect to short-term synaptic dynamics, from a pronounced facilitation to a pronounced depression. Facilitation/depression was the same whether evaluated using the magnitude or the probability of occurrence of the postsynaptic respon

Surgical implantation of a peripheral nerve neuroma in lesioned spinal cord and brain--axonal sprouting regeneration and synaptogenesis.

Thirty-six mature female dogs weighing fifteen to twenty-five pounds, then spinal cord transected at T6 or T8. An intercostal nerve neuroma with cellular origin proximal to the transection site was dissected free to near the intervertebral foramen and the distal nerve stump was implanted in the spinal cord distal but near to the transection site and anchored with a plasma clot suture. These dogs were studied, treated and rehabilitated over a period of six months to two years or until they could stand, step and show reflex walking locomotion. Peripheral nerve distal stump neuroma, for example,

Dopamine modulation of two subthreshold currents produces phase shifts in activity of an identified motoneuron.

1. The lateral pyloric (LP) neuron is a component of the 14-neuron pyloric central pattern generator in the stomatogastric ganglion of the spiny lobster, Panulirus interruptus. In the pyloric rhythm, this neuron fires rhythmic bursts of action potentials whose phasing depends on the pattern of synaptic inhibition from other network neurons and on the intrinsic postinhibitory rebound properties of the LP cell itself. Bath-applied dopamine excites the LP cell and causes its activity to be phase advanced in the pyloric motor pattern. At least part of this modulatory effect is due to dopaminergic

Role of different types of Ca2+ channels and a reticulum-like Ca2+ pump in neurotransmitter release.

The factors controlling the Ca2+ concentration directly responsible for triggering acetylcholine (ACh) release were investigated at an identified neuro-neuronal synapse of the Aplysia buccal ganglion. The types of presynaptic voltage-gated Ca2+ channels associated with transmitter release were determined by using selective blockers such as nifedipine, omega-conotoxin and a partially purified extract from the venom of a funnel web spider (FTx). L-type, N-type and P-type Ca2+ channels are present in the presynaptic neuron. The influx of Ca2+ through both N- and P-types induces the release of ACh

Effects of systemically and locally applied cocaine on cerebrocortical neuron responsiveness to afferent synaptic inputs and glutamate.

The goal of the present study was to determine the effects of systemically or locally applied cocaine on rat somatosensory cortical neuron responsiveness to afferent synaptic inputs or putative transmitter application and to compare these results with previously observed actions of endogenous cortical monoamines on the same parameters of neuronal function. Individual cells in rat cortex were activated by stimulation of thalamocortical afferents or local iontophoretic application of glutamate. Extracellularly recorded responses to these stimuli were monitored before and after parenteral or micr

[Effect of hydrocortisone on synaptic transmission in surviving slices of the olfactory cortex from the rat brain].

A study was made of the effect of bath-applied hydrocortisone (10(-4) M) on excitatory transmission of the lateral olfactory tract (LOT) - superficial pyramidal cell synapse of the rat olfactory cortex slice. Population EPSP and IPSP were depressed for 6-12 min., then they were increased (60 min.) by hydrocortisone. Recovery of evoked potentials was long (40-60 min.) at washing of slices hy a normal incubation medium. Habituation of synaptic potentials during LOT repeating stimulation was deteriorated by hydrocortisone application in perfusion solution. The data obtained were considered to be

The channel-forming component of the Theridiidae spider venom neurotoxins.

It is known that Steatoda (Lityphantes) paykulliana and Latrodectus mactans tredecimguttatus spider venoms are toxic to mammals and insects. These venoms act presynaptically eliciting massive release of transmitters. They also form channels in bilayer lipid membranes (BLM) that are selective for cations. Venoms of both spider species were fractionated by gel filtration on a Sephadex G-100 column. The fraction obtained were tested on neuromuscular preparations of frog and locust and on BLM. A fraction of low molecular weight components (about 5000 daltons and less) was disclosed. This fraction

Potentiation and depression of synaptic transmission in the olfactory cortex of the guinea-pig.

1. The extracellular field potentials of the olfactory cortex evoked by stimulation of the lateral olfactory tract (l.o.t.) were studied in in vitro preparations from the olfactory cortex. The field potentials comprised an initial diphasic wave - the l.o.t. compound action potential - followed by a negative wave of about 10 msec duration which in turn was followed by a low amplitude positive wave of long duration (100 msec or more). In this paper, the size of the negative field potential (extracellularly recorded EPSP) has been studied during and after periods of repetitive stimulation of the

Perineuronal Nets Restrict the Induction of Long-Term Depression in the Mouse Hippocampal CA1 Region.

Long-term depression (LTD) of synaptic efficacy is widely regarded as a cellular basis of learning and memory. The magnitude of hippocampal CA1 LTD induced by low-frequency stimulation (LFS) declines with age, but the mechanisms involved remain poorly understood. Perineuronal nets (PNNs) are specialized extracellular matrix structures that function in dampening synaptic plasticity during postnatal development, suggesting that PNN formation may restrict LTD induction in the adult hippocampus. Here, we show that PNNs tightly enwrap a subpopulation of parvalbumin (PV) interneurons in the hippocam

Temporal dynamics of plastic changes in human primary somatosensory cortex after finger webbing.

The primary somatosensory cortex (SI) exhibits a detailed topographic organization of the hand and fingers, which has been found to undergo plastic changes following modifications of the sensory input. Although the spatial properties of these changes have been extensively investigated, little is known about their temporal dynamics. In this study, we adapted the paradigm of finger webbing, in which 4 fingers are temporarily webbed together, hence modifying their sensory feedback. We used magnetoencephalography, to measure changes in the hand representation in SI, before, during, and after finge

Contrasting forms of synaptic plasticity in monkey inferotemporal and primary visual cortices.

Activity-dependent synaptic plasticity was examined in vivo in two cortical areas of the adult monkey visual system, the primary visual and inferotemporal cortex, the first and late cortical stages essential for object recognition. Discontinuous high-frequency electrical stimulation of intrinsic horizontal connections in layer 2/3 caused contrasting forms of synaptic plasticity in the two areas. In the inferotemporal cortex, long-term potentiation of extracellular field potentials in layer 2/3 was induced, whereas in the same pathway of V1, identical stimulation protocol elicited long-term dep

Effects of electrical stimulation of the superior cervical ganglia on the number of "synaptic" ribbons and the activity of melatonin-forming enzymes in the rat pineal gland.

Melatonin metabolism in the mammalian pineal gland is under the clear influence of sympathetic fibers originating in the superior cervical ganglia (SCG). Previous studies suggested that pineal "synaptic" ribbons (SR) as well are regulated by the gland's sympathetic innervation. To gain more insight into the mechanisms involved, we examined the effects of sympathetic stimulation on SR number and on the activity of melatonin forming enzymes, serotonin N-acetyltransferase (NAT) and hydroxyindole-O-methyltransferase (HIOMT). The SCG in adult male rats were stimulated electrically during daytime fo

Inhibition of norepinephrine release by presynaptic alpha 2-adrenoceptors in mesenteric vasculature preparations from chronic DOCA-salt hypertensive rats.

This study investigated norepinephrine release during electrical nerve stimulation and inhibitory characteristics of presynaptic alpha 2-adrenoceptors in perfused mesenteric vasculature from deoxycorticosterone acetate (DOCA)-salt hypertensive rats (7-8 weeks after surgery). Electrical stimulation of sympathetic innervation caused a significantly greater release of endogenous norepinephrine into the mesenteric vasculature of DOCA-salt hypertensive rats than in age-matched normotensive controls. Pressor responses to electrical nerve stimulation were also enhanced in DOCA-salt hypertension. Yohi

Novel synaptic responses mediated by dopamine and gamma-aminobutyric acid in neuroendocrine cells of the intermediate pituitary.

Neuroendocrine cells of the pituitary pars intermedia contain pro-opiomelanocortin and secrete mainly alpha-melanocyte-stimulating hormone and, in lesser amounts, corticotropin and endorphin. We have recorded intracellularly from rat pars intermedia cells in vitro and analysed the synaptic inputs to these cells. Electrical stimulation of the pituitary stalk evoked a biphasic postsynaptic response in pars intermedia cells. An initial inhibitory postsynaptic potential was most likely mediated by gamma-aminobutyric acid (GABA) because there was a conductance increase, and it was blocked by bicucu
