Motion of the center of mass in children with spastic hemiplegia: balance, energy transfer, and work performed by the affected leg vs. the unaffected leg.

Asymmetry between limbs in people with spastic hemiplegic cerebral palsy (HEMI) adversely affects limb coordination and energy generation and consumption. This study compared how the affected leg and the unaffected leg of children with HEMI would differ based on which leg trails. Full-body gait analysis data and force-plate data were analyzed for 31 children (11.9 ± 3.8 years) with HEMI and 23 children (11.1 ± 3.1 years) with typical development (TD). Results showed that peak posterior center of mass-center of pressure (COM-COP) inclination angles of HEMI were smaller than TD when the affected

Detection of lead (II) with a "turn-on" fluorescent biosensor based on energy transfer from CdSe/ZnS quantum dots to graphene oxide.

Graphene oxide (GO) sheets are mixed with the aptamer-functionalized CdSe/ZnS quantum dots (QDs). Consequently, the aptamer-conjugated QDs bind to the GO sheets to form a GO/aptamer-QD ensemble, which enables the energy transfer from the QDs to the GO sheets, quenching the fluorescence of QDs. The GO/aptamer-QD ensemble assay acts as a "turn-on" fluorescent sensor for Pb(2+) detection. When Pb(2+) ions are present in the assay, the interaction of Pb(2+) with the aptamer induces a conformational change in the aptamer, leading to the formation of a G-quadruplex/Pb(2+) complex. As a result, the Q

Trace Hydrogen Sulfide Enhances Plasmid-Mediated Antibiotic Resistance Gene Transfer under Anaerobic Sewer-Like Conditions: Associations with Contact and Energy-State Responses.

The dissemination of antibiotic resistance genes (ARGs) in sewer systems poses a potential threat to public health. However, it is currently unclear how hydrogen sulfide (H2S) in anaerobic sewers affects plasmid-mediated ARG transfer. Here, we evaluated the effects of environmentally relevant concentrations (1-100 μmol·L-1) of H2S on ARG transfer across different sewer microbial communities. The results demonstrated that H2S significantly enhanced the transfer frequency of plasmid pKJK5 in sediment-derived (2.06-3.06-fold), sewage-derived (1.50-2.81-fold), and bioaerosol-derived (2.42-4.68-fol

Enhanced through-bond energy transfer-based bioorthogonal probe enables catalytic-amplified and sensitive detection of microRNA-21 for clinical lung cancer diagnosis.

The dysregulation of microRNA (miRNA) expression is closely linked to the pathogenesis of lung cancer, rendering the quantification of trace miRNAs clinically indispensable. However, achieving ultrasensitive detection of miRNAs in complex biological matrices, including living cells, tissues, and blood serum, remains significant challenges. To address this, we developed a fluorogenic detection platform for miR-21 based on tetrazine-mediated transfer (TMT) reactions. While TMT strategies have been previously explored, we report a refined probe design optimized through computational screening. By

Stem-loop-terminated dendrimer DNA enables the ratiometric fluorescent miRNA analysis by fluorescence resonance energy transfer via the proximity/local concentration effect.

MicroRNA-21 (miR-21), a short non-coding RNA, functions as a diagnostic early biomarker for cancers, cardiovascular diseases, and inflammatory disorders. Thus, the development of a rapid, convenient, and highly sensitive method for miR-21 detection is essential for accurate disease diagnosis through point-of-care testing. Herein, we designed the stem-loop-terminated fluorescent DNA dendrimer, comprising Y-DNAa and Y-DNAb, to create a proximity/local concentration effect for detecting miR-21 through fluorescence resonance energy transfer (FRET). Both Y-DNAa and Y-DNAb consists of a dendritic co

A DNA Scaffold Approach Facilitates 5' Labeling of the SARS-CoV-2 RNA Pseudoknot for Single-Molecule Förster Resonance Energy Transfer Investigation.

Single-molecule Förster resonance energy transfer (smFRET) studies of highly structured RNA molecules are often frustrated by issues with efficient dye conjugation. Here, a DNA scaffold-based labeling strategy is developed, and applied to the frameshift-stimulating RNA pseudoknot from the SARS-CoV-2 genome. FRET-active reporters were prepared containing both Cy3 (donor) and Cy5 (acceptor) molecules and measurements conducted on freely-diffusing single molecules, enabling the evaluation of conformational heterogeneity via smFRET population distributions. Freely diffusing pseudoknots, modified a

Shoot-through proton FLASH irradiation lowers linear energy transfer in organs at risk for neurological tumors and is robust against density variations.

Objective. The goal of the study was to test the hypothesis that shoot-through FLASH proton beams would lead to lower dose-averaged LET (LETD) values in critical organs, while providing at least equal normal tissue sparing as clinical proton therapy plans.Approach. For five neurological tumor patients, pencil beam scanning (PBS) shoot-through plans were made, using the maximum energy of 227 MeV and assuming a hypothetical FLASH protective factor (FPF) of 1.5. The effect of different FPF ranging from 1.2 to 1.8 on the clinical goals were also considered. LETDwas calculated for the clinical plan

Electronic energy and electron transfer processes in photoexcited donor-acceptor dyad and triad molecular systems based on triphenylene and perylene diimide units.

We investigate the photophysical properties of organic donor-acceptor dyad and triad molecular systems based on triphenylene and perylene diimide units linked by a non-conjugated flexible bridge in solution using complementary optical spectroscopy techniques. When these molecules are diluted in dichloromethane solution, energy transfer from the triphenylene to the perylene diimide excited moieties is evidenced by time-resolved fluorescence measurements resulting in a quenching of the emission from the triphenylene moieties. Simultaneously, another quenching process that affects the emission fr

Differential detection of Trichinella papuae, T. spiralis and T. pseudospiralis by real-time fluorescence resonance energy transfer PCR and melting curve analysis.

Trichinellosis caused by nematodes of Trichinella spp. is a zoonotic foodborne disease. Three Trichinella species of the parasite including Trichinella spiralis, Trichinella papuae and Trichinella pseudospiralis, have been etiologic agents of human trichinellosis in Thailand. Definite diagnosis of this helminthiasis is based on a finding of the Trichinella larva (e) in a muscle biopsy. The parasite species or genotype can be determined using molecular methods, e.g., polymerase chain reaction (PCR). This study has utilized real-time fluorescence resonance energy transfer PCR (real-time FRET PCR

Voxel-Wise Case-Control Analysis of Dose, Linear Energy Transfer, and Symptomatic Brainstem Necrosis in Pediatric Brain Tumors Treated With Proton Therapy.

PURPOSE: Brainstem necrosis is a rare but serious toxicity following radiation therapy for pediatric brain tumors. In proton therapy, concerns have emerged around elevated linear energy transfer (LET) and the relative biological effectiveness (RBE) in relation to normal tissue damage. Although previous studies have largely focused on asymptomatic image changes in adults, we investigated voxel-wise correlations between dose, LET, and symptomatic brainstem necrosis in a pediatric case-control cohort. METHODS AND MATERIALS: From a cohort of exactly 1200 patients with central nervous system tumor

Substrate-Controlled Dearomative Cycloadditions of Perfluoronaphthalenes via Visible-Light Energy Transfer Catalysis: Switchable Access to [2+2], [4+2], and Diastereoconvergent Cycloadducts.

We report the first substrate-controlled intermolecular dearomative cycloaddition of polyfluoronapthalenes in which the substitution pattern of the olefin coupling partner governs the mode of cycloaddition. Under visible-light energy transfer (VLEnT) catalysis, monosubstituted olefins react with octafluoronaphthalene to give dual dearomative ortho [2+2] cycloadducts selectively. In comparison, 1,1-disubstituted olefins afford the corresponding para [4+2] cycloadducts with complete regiospecificity under otherwise identical conditions. Trisubstituted olefins engage in a diastereoconvergent [4+2

The Association of Linear Energy Transfer and Dose With Radiation Necrosis After Pencil Beam Scanning Proton Therapy in Pediatric Posterior Fossa Tumors.

PURPOSE: Proton therapy is the preferred treatment modality for most pediatric central nervous system tumors. The risk of radiation necrosis may be increased at the distal end of the beam because of an increase in linear energy transfer (LET) and relative biological effectiveness (RBE) dose. We report on the association of LET and dose with radiation necrosis after pencil beam scanning proton therapy in pediatric posterior fossa tumors using a case-control framework. MATERIALS AND METHODS: From 2019 to 2022, 33 patients less than or equal to 18 years of age treated with first-line proton ther

Crystallization-Driven Controlled Two-Dimensional (2D) Assemblies from Chromophore-Appended Poly(L-lactide)s: Highly Efficient Energy Transfer on a 2D Surface.

A rational approach towards precision two-dimensional (2D) assemblies by crystallization-driven self-assembly (CDSA) of poly(L-lactides) (PLLAs), end-capped with dipolar dyes like merocyanine (MC) or naphthalene monoimide (NMI) and hydrophobic pyrene (PY) or benzene (Bn) is described. PLLA chains crystallize into diamond-shaped platelets in isopropanol, which forces the terminal dyes to assemble into a 2D array on the platelet surface by either dipolar interactions or π-stacking and exhibit tunable emission. Dipolar dyes play a critical role in imparting colloidal stability and structural unif

Detection and Quantification of Biologically Active Botulinum Neurotoxin Serotypes A and B Using a Förster Resonance Energy Transfer-Based Quantum Dot Nanobiosensor.

Botulinum neurotoxin (BoNT) is the most potent toxin known. The ingestion of food contaminated with biologically active BoNT causes foodborne botulism, which can lead to respiratory paralysis, coma, and death after ingestion of as little as 70 μg for a 70 kg human. Because of its lethality and challenges associated with current detection methods, there is an urgent need for highly sensitive rapid screening techniques capable of detecting biologically active BoNT. Here, we describe a Förster resonance energy transfer-based nanobiosensor that uses quantum dots (QDs) and two specific quencher-lab

Fluorescence resonance energy transfer-based ratiometric fluorescent probe for detection of Zn(2+) using a dual-emission silica-coated quantum dots mixture.

In this work, we report the design and application of a new ratiometric fluorescent probe, which contains different-colored quantum dots (QDs) as dual fluorophores, ultrathin silica shell as spacer, and meso-tetra(4-sulfonatophenyl)porphine dihydrochloride (TSPP) as receptor, for Zn(2+) detection in aqueous solution and living cells. In the architecture of our designed probe, the silica shell plays the key roles in controlling the locations of QDs, TSPP, and Zn(2+), preventing the direct contact between QDs and Zn(2+) but affording fluorescence resonance energy transfer (FRET) from dual-color

Interplay between amplified spontaneous emission, Forster resonant energy transfer, and self-absorption in hybrid poly(9,9-dioctylfluorene)-CdSe/ZnS nanocrystal thin films.

We investigated the excitation density dependence of the photoluminescence spectra of hybrid poly(9,9-dioctylfluorene)-CdSe/ZnS nanocrystals (PF8-NCs) thin films. We demonstrate that this experiment allows the determination of the efficiency of all the CdSe/ZnS NCs excitation processes and that the presence of amplified spontaneous emission (ASE) from the PF8 leads to a strong dependence of the NC excitation processes from the laser excitation density. Below the PF8 ASE threshold only about 6% of the excitons in the NCs are due to pump laser absorption, while about 94% of the NC excitation is
