m6A-modified Mid1 promotes sevoflurane-induced cognitive impairment in neonatal mice by ubiquitin-mediated degradation of Syngap1.

Investigating the cognitive effects of sevoflurane exposure during early development is essential due to its potential long-term neurodevelopmental impacts. This investigation systematically explored the molecular basis of sevoflurane-induced cognitive impairment, with emphasis on m6A RNA modifications and ubiquitin-dependent proteostasis involving Mid1 and Syngap1. Using integrated approaches, including methylated RNA immunoprecipitation sequencing (MeRIP-seq), transcriptomic profiling, neurobehavioural testing and molecular analyses, 2091 m6A methylation sites were identified that were diffe

tRNA modification landscapes in streptococci: shared losses and clade-specific adaptations.

tRNA modifications are central to bacterial translational control. Here, we integrated genetics, mass spectrometry, epitranscriptomics and comparative genomics to map the tRNA modification genes of the Gram-positive pathogens Streptococcus mutans and Streptococcus pneumoniae. Both species show a marked loss of modifications dependent on Fe-S enzymes, consistent with a broader trend of Fe-S enzyme reduction in Streptococcus central metabolism. In addition, the D, m1A, m7G, t6A and i6A modifications were mapped in S. pneumoniae tRNAs, and we confirmed that a unique DusB1 enzyme is responsible fo

N6-methyladenosine modification of NEU1 mediated by METTL3 exacerbates angiotensin II-induced atrial fibrillation.

Neuraminidase 1 (NEU1) regulation of atrial fibrillation (AF) progression via fibrosis remains unknown. Mice receiving AAV9-mediated NEU1 knockdown were infused with Ang II and subjected to programmed electrical stimulation to induce AF. Left atrial dilation and fibrosis were evaluated by echocardiography, histology, and fibrosis markers. Primary mouse atrial fibroblasts treated with Ang II were assessed for proliferation and migration by EdU staining and Transwell. The N6-methyladenosine (m6A) modification of NEU1 by methyltransferase-like 3 (METTL3) was confirmed through m6A quantification,

7-Deshydroxypyrogallin-4-carboxylic acid dietary phytochemical as a putative FTO inhibitor in breast cancer: an integrated computational and cytotoxicity study.

Breast cancer is the most frequently diagnosed malignancy in women globally, representing a major global health burden, with a significant subset of cases exhibiting resistance to standard therapeutic regimens. Recently, m6A RNA demethylases such as FTO have been identified as important oncogenes in breast cancer, which modify the epitranscriptome of tumor cells to favor expression of growth-promoting genes while suppressing expression of anti-apoptotic genes. In this study, we performed virtual screening of 1,000 dietary compounds targeting the FTO protein in order to discover potential thera

Associations of children's urinary metals with blood RNA N6-methyladenosine and 2'-O-methyladenosine.

BACKGROUND: Post-transcriptional RNA modifications (the epitranscriptome) regulate gene expression and represent a pathway through which environmental exposures can affect health. Two RNA modifications important for health and disease are N6-methyladenosine (m6A) and 2'-O-methyladenosine (Am). However, the effects of metals and metalloids (hence, metals), on m6A and Am are less well understood. We aimed to investigate associations between children's metals exposures and global m6A and Am in blood. METHODS: We measured essential (cobalt, copper, selenium, zinc) and nonessential (arsenic, cadmi

Prenatal cadmium exposure and male infertility in mice: A multi-generational mechanistic study.

Male infertility affects approximately one in seven couples worldwide. Prenatal cadmium (Cd) exposure has been shown to affect offspring phenotypes and increase susceptibility to diseases later in life. However, the effects of prenatal Cd exposure on multi-generational offspring fertility and the mechanisms remain unknown. A novel murine multi-generational (F1-F3 offspring) male subfertility model induced by prenatal Cd exposure was developed. The levels of testosterone and steroidogenic enzymes were also lower in these offspring's testes. The ubiquitin-dependent degradation of NR4A1, the upst

ALKBH5 inhibitors for cancer therapy: molecular docking, challenges and future prospects.

N6-Methyladenosine (m6A), the most abundant internal modification of eukaryotic mRNA, is dynamically reversed by the FeII/α-ketoglutarate-dependent dioxygenase ALKBH5, a key m6A "eraser" that regulates target mRNA fate through demethylation. ALKBH5 is aberrantly overexpressed in acute myeloid leukemia, glioblastoma, breast cancer and other malignancies, where it sustains oncogenic programs, establishing it as a promising epitranscriptomic drug target. At present, ALKBH5 inhibitor development lags behind that of FTO, with selective agents only recently emerging through covalent inhibitors such

TRMT1-mediated tRNA m22G modification drives Osimertinib resistance via the ATXN3/USP25 axis in lung adenocarcinoma.

Acquired resistance to Osimertinib, a third-generation EGFR tyrosine kinase inhibitor (TKI), remains a critical challenge in lung adenocarcinoma. Here, we identify ATXN3, a deubiquitinating enzyme, as a driver of Osimertinib resistance and a predictor of poor patient survival. Osimertinib treatment dynamically upregulates ATXN3 transcription, which stabilizes USP25 through deubiquitination, activating an ATXN3-USP25-TRMT1 signaling cascade. This axis enhances TRMT1-mediated tRNA m²2G modifications, enabling selective translation of redox-regulating enzymes (e.g., GPX4, SOD2) that scavenge reac

The expanding role of m6A RNA modification in plant-virus dynamics: friend, foe, or both?

N6-methyladenosine (m6A), the most prevalent internal mRNA modification, regulates plant development and stress responses through modulating various mRNA metabolic processes and epigenetic effects. Although well studied in animals, its roles in plant-virus interactions have only recently begun to be elucidated. Multiple plant viruses carry m6A modifications on their RNAs, validated by MeRIP-seq, LC-MS/MS, and direct RNA sequencing. Viral RNAs acquire m6A through the recruitment or relocalization of host methyltransferase complexes, which is often mediated by viral proteins. Functionally, m6A c

The RNA methyltransferase NSUN2 catalyzes 5-methylcytosine (m5C) on IL1B mRNA to promote transcript stability.

Among the plethora of RNA modifications, 5-methylcytosine (m5C) has held substantial roles in some biological processes, yet its impact on inflammation remains largely uncharted. Here, we reported an m5C-related epitranscriptomic regulatory axis in inflammatory pathogenesis. Bacterial lipopolysaccharides (LPS) challenge induces marked elevation in RNA m5C abundance as well as the expression of NOP2/Sun RNA methyltransferase 2 (NSUN2), an essential enzyme catalyzing m5C modification. Nsun2 deficient mice showed significantly reduced inflammatory response in the models of systemic inflammation a

[Primary humoral immunodeficiencies associated with enteropathies: An update].

Inflammatory enteropathies are frequent in primary immune deficiencies (PID) (up to 21%). Their pathophysiology is complex and combines immune abnormalities (loss of tolerance, deficit in mucosal IgA secretion and altered infectious response) with an abnormal environment (dysbiosis, production of pro-inflammatory endotoxins, epigenetic and epitranscriptomic alterations). This leads to a deregulated inflammatory state of the gastrointestinal tract and thus to enteropathy. These conditions are heterogeneous and manifest not only as an inflammatory bowel disease but also as in lymphocytic, autoim

RNA methylation: where to from here for hematologic malignancies?

RNA methylation and the machinery that regulates or "reads" its expression has recently been implicated in the pathogenesis of acute myeloid leukemia (AML) and other hematologic malignancies. Modulation of these epigenetic marks has started to become a reality as several companies around the world seek to leverage this knowledge therapeutically in the clinic. Although the bases of observed activity in AML have been described by numerous groups, the exact context in which these therapies will ultimately be used remains to be properly determined. While context is likely to be of great importance

NAT10 exacerbates aortic valve calcification via PCSK9 mRNA ac4C acetylation.

N4-acetylcytidine (ac4C) is an epitranscriptomic regulator involved in diverse pathological processes. This study investigates the role of N-acetyltransferase 10 (NAT10), the primary enzyme responsible for ac4C deposition, in aortic valve calcification (AVC). NAT10 expression and global ac4C levels are upregulated in calcified aortic valves from human patients and murine models. Clinically, elevated NAT10 expression correlates with increased disease severity. Functionally, NAT10 promotes human valvular interstitial cells (hVICs) osteogenic differentiation. In high-cholesterol diet (HCD)-fed Ap

Context-dependent functions of ALKBH5: a mechanistic framework linking cellular stress responses, immune regulation, viral infection, and therapeutic vulnerabilities.

N6 -methyladenosine (m6A) is a major post-transcriptional RNA modification, and the demethylase ALKBH5 has emerged as a versatile but highly context-dependent regulator of RNA fate. This review integrates current evidence showing that ALKBH5 links epitranscriptomic control to cellular stress adaptation, genome maintenance, immune-cell function, viral infection, and therapeutic response. In DNA damage and cell-cycle regulation, ALKBH5 modulates checkpoint, repair, and apoptotic pathways, thereby influencing genome stability and sensitivity to radiotherapy or chemotherapy. In immune biology, it

The regulatory role of m6A RNA methylation in insect sexual development: a review.

N6-methyladenosine (m6A) is the most prevalent internal mRNA modification in eukaryotes and serves as a key post-transcriptional regulator in insect development. This review synthesizes current knowledge on m6A in insect sexual development, beginning with its well-established role in Drosophila melanogaster, where it ensures female fate by promoting the female-specific splicing of the master switch gene Sex-lethal (Sxl). Notably, classical female-lethal genes such as Fl(2)d and Virilizer are now recognized as essential regulatory subunits of the m6A methyltransferase complex, linking classic g

Zinc finger protein ZNF33B facilitates Japanese encephalitis virus replication by regulating m6A modification on Ifih1 and Irf3 transcripts to antagonize antiviral response.

Japanese encephalitis virus (JEV), a neurotropic flavivirus, poses a significant public health threat, yet the molecular mechanisms underlying its interaction with host immunity remain poorly understood. This study reveals that zinc finger protein ZNF33B promotes JEV replication by subverting the RLR-mediated innate immune response through orchestrating m⁶A RNA modification. ZNF33B directly binds to antiviral transcripts Ifih1 (encoding MDA5), Mavs, and Irf3, recruiting the m⁶A methyltransferase METTL14 to enhance their m⁶A methylation. Concurrently, ZNF33B interacts with the nuclear m⁶A reade

Mild and ultrafast GLORI enables absolute quantification of m6A methylome from low-input samples.

Methods for absolute quantification of N6-methyladenosine (m6A) have emerged as powerful tools in epitranscriptomics. We previously reported GLORI, a chemical-assisted approach to achieve unbiased and precise m6A measurement. However, its lengthy reaction time and severe RNA degradation have limited its applicability, particularly for low-input samples. Here, we present two updated GLORI approaches that are ultrafast, mild and enable absolute m6A quantification from one to two orders of magnitude less than the RNA starting material: GLORI 2.0 is compatible with RNA from ~10,000 cells and enhan

Nat10-mediated N4-acetylcytidine modification enhances Nfatc1 translation to exacerbate osteoclastogenesis in postmenopausal osteoporosis.

Increased differentiation or activity of osteoclasts is the key pathogenic factor of postmenopausal osteoporosis (PMOP). N4-acetylcytidine (ac4C) modification, catalyzed by Nat10, is a novel posttranscriptional mRNA modification related to many diseases. However, its impact on regulating osteoclast activation in PMOP remains uncertain. Here, we initially observed that Nat10-mediated ac4C positively correlates with osteoclast differentiation of monocytes and low bone mass in PMOP. The specific knockout of Nat10 in monocytes and remodelin, a Nat10 inhibitor, alleviates ovariectomized (OVX)-induc

m6A RNA Modification Controls HTLV-1 Tax and Host Gene Expression.

Human T-cell Leukemia Virus Type 1 (HTLV-1) is a pathogenic human retrovirus that is responsible for intractable diseases such as adult T-cell leukemia-lymphoma (ATL), a malignancy with a poor patient prognosis. Although recent studies have delineated several genomic, epigenomic, and transcriptomic abnormalities associated with HTLV-1, to date the importance of epitranscriptomic modifications, particularly N6-methyladenosine (m6A), remains unclear. Here, we showed that the HTLV-1 RNA genome undergoes m6A modification, thereby suggesting that these modifications act as bidirectional regulators
