A Comprehensive Review on Strategies of Topical Niosomes and Their Synergistic Effect for Enhanced Therapeutic Outcomes.

The review aims to assess the potential of niosomes-nonionic surfactant-based vesicular systems-as carriers for topical and transdermal drug delivery. Niosomes enable targeted and controlled drug release while minimizing systemic toxicity. The investigation centers on their structure, stability, and capacity to entrap both hydrophilic and lipophilic drugs, as well as their use in managing various dermatological and systemic disorders. Recent studies have examined the formulation of niosomes, particularly highlighting the roles of nonionic surfactants and cholesterol in enhancing the stability

Ferrous lactate-loaded hydrogels induce iron-dependent non-canonical ferroptosis in Cutibacterium acnes: a novel therapeutic strategy for acne vulgaris.

Acne vulgaris is a common inflammatory skin condition that affects up to 85% of adolescents and is primarily driven by Cutibacterium acnes (anaerobic bacteria, formerly Propionibacterium acnes). With the rise of antibiotic resistance, research on alternative antimicrobial agents that can address anaerobic bacteria resistance is becoming increasingly important, especially for patients who cannot use systemic antimicrobial therapy. In this study, a hyaluronic acid (HA)-based hydrogel incorporating ferrous lactate (FeLac) was developed as a non-antibiotic antimicrobial strategy targeting anaerobi

A Molecular-Dynamics-Guided Screening Pipeline Identifies Topical Glucocorticoid Receptor Modulators with Selective Epidermal Activation.

INTRODUCTION: Topical corticosteroids remain the mainstay of psoriasis management but are limited by skin atrophy and barrier impairment. Developing safer, steroid-sparing therapy represents an unmet need in topical pharmacology. The glucocorticoid receptor (GR) plays a central role in epidermal homeostasis, yet selective activation by natural phytochemicals has not been systematically explored. METHODS: A molecular modeling framework integrating docking, molecular dynamics simulations, and pharmacophore conservation was established to evaluate GR engagement by four phytochemicals - sakuranet

Recent Advances in the Local Drug Delivery Systems for Diabetic Wound Healing: A Comprehensive Review.

Wound management in diabetic patients holds significant importance in both clinical and social contexts due to the delayed and compromised healing that these individuals experience. Diabetic wounds exhibit slow and incomplete healing, increasing patients' susceptibility to infections. Managing wounds in diabetic patients, particularly when complicated by diabetic foot infection or diabetic foot ulcer, becomes challenging. The ideal drug delivery systems for treating diabetic wounds should integrate diverse drugs and/or biological factors, offering advantages such as sustained and localized rel

Dual-enhanced transdermal delivery: Magnolol-borneol nanoparticle hydrogel for topical treatment of diabetic peripheral neuropathy.

Diabetic peripheral neuropathy (DPN) remains a prevalent and challenging complication of diabetes with limited therapeutic options. Magnolol (Mag), a natural multi-target agent, demonstrates significant potential in managing DPN through synergistic anti-inflammatory, antioxidant, and neurorestorative properties. However, its poor oral bioavailability and limited skin penetration hinder clinical application. To address these challenges, we developed a novel topical nanogel system for enhanced transdermal delivery of Mag. Borneol (Bor), a potent penetration enhancer, was co-assembled with Mag vi

Association between Scabies Treatment and Parkinson's Disease: A Nationwide, Population-Based Study.

BACKGROUND: Scabies is typically treated with scabicides like lindane, which poses a risk for acute neural toxicity. Lindane's prolonged use, particularly in agriculture, is linked to neurodegenerative diseases, including Parkinson's disease (PD), the second most common neurodegenerative disorder. This study aimed to evaluate whether scabies patients, particularly those treated with topical lindane, are at increased risk of developing PD. METHODS: A nationwide population-based cohort study was conducted using data from Taiwan's National Health Research Institutes claims database from 2000 to

Amphotericin B Loaded Polymeric Nanoparticles for Treatment of Leishmania Infections.

Fungal infections in immune-compromised patients are an important cause of mortality and morbidity. Amphotericin B (Amp B) is considered a powerful fungicidal drug but its clinical usage has certain limitations when administered intravenously due to its toxicity and poor solubility. In consideration of such challenges, in cutaneous leishmaniasis, the topical application of Amp B can be a safer option in many aspects. Thus, herein, biopolymer of polycaprolactone (PCL) nanoparticles (NPs) were developed with the loading of Amp B by nanoprecipitation for the treatment of topical leishmanial infec

Design of a Topically Applied Gel Spray Formulation with Ivermectin Using a Novel Low Molecular Weight Gelling Agent, Palmitoyl-Glycine-Histidine, to Treat Scabies.

Palmitoyl-glycine-histidine (Pal-GH) is a new low molecular weight gelling agent. It exhibits thixotropic behavior, low viscosity, and high dissolving properties for a wide range of hydrophilic to lipophilic drugs. Orally administered ivermectin (IVM) is used to treat scabies. However, this treatment is associated with well-known side effects, thus a study is awaited to search for alternative routes of administration. Although a topical formulation of IVM could be a candidate, it requires whole body application except the head and face for several hours on a daily basis. Therefore, in this stu

Unexpected rewards induce dopamine-dependent positive emotion-like state changes in bumblebees.

Whether invertebrates exhibit positive emotion-like states and what mechanisms underlie such states remain poorly understood. We demonstrate that bumblebees exhibit dopamine-dependent positive emotion-like states across behavioral contexts. After training with one rewarding and one unrewarding cue, bees that received pretest sucrose responded in a positive manner toward ambiguous cues. In a second experiment, pretest consumption of sucrose solution resulted in a shorter time to reinitiate foraging after a simulated predator attack. These behavioral changes were abolished with topical applicati
