Laser Powder Bed Fusion (LPBF) WE43 Magnesium Scaffold with EGCG/PDA-Functionalized Silk Fibroin Membrane for Antibacterial Osteoinductive Guided Bone Regeneration.

Guided bone regeneration (GBR) is a prominent focus in biomedical materials research, yet few studies address practical clinical needs. GBR membranes must fulfill the "PASS" principles to be effective in surgery, but existing membranes often fall short in balancing antibacterial activity, controlled degradation, osteoinductive potential, and mechanical support. In this study, we employed laser powder bed fusion (LPBF) to fabricate a porous WE43 magnesium alloy scaffold suitable for large alveolar bone defects. We then used electrospinning and layer-by-layer (LBL) self-assembly to create a silk

Silk fibroin peptide self-assembled nanofibers delivered naringenin to alleviate cisplatin-induced acute kidney injury by inhibiting mtDNA-cGAS-STING pathway.

Silk fibroin (SF) has excellent biocompatibility and biodegradability as a biomaterial. The purity and molecular weight distribution of silk fibroin peptide (SFP) make it more suitable for medical application. In this study, SFP nanofibers (molecular weight ∼30kD) were prepared through CaCl2/H2O/C2H5OH solution decomposition and dialysis, and adsorbed naringenin (NGN) to obtain SFP/NGN NFs. In vitro results showed that SFP/NGN NFs increased the antioxidant activity of NGN and protected HK-2 cells from cisplatin-induced damage. In vivo results also showed that SFP/NGN NFs protected mice from ci

Dual drug delivery system of teicoplanin and phenamil based on pH-sensitive silk fibroin/sodium alginate hydrogel scaffold for treating chronic bone infection.

For effective treatment of infected bone, it is essential to use local drug delivery systems with the ability to deliver both antibiotics and osteoinductive factors. Herein, a pH-sensitive silk fibroin (SF)/sodium alginate (SA) hydrogel scaffolds containing teicoplanin (TEC) and phenamil (PM) loaded SF nanoparticles (PMSFNPS) are introduced for treating chronic osteomyelitis. The TEC and PM showed a sustained- and pH-sensitive release behavior from SF/SA hydrogel. The higher release rate was seen in an alkaline pH in comparison to neutral and acidic pH during 10 days. The eluted TEC maintained

Tuning properties of fibroin-based materials: effect of freezing cycles on shape memory of hydrated cryogels and a comparative analysis of drying techniques on dried material performance.

This study addresses the development of hydrated silk fibroin cryogels and their subsequent drying using a sustainable approach in which water is the only solvent, offering a simple and environmentally friendly processing route. The effects of key parameters, including the number of freeze-thaw cycles (1, 2, and 3) and freezing temperatures (-20 °C and -80 °C), on the shape memory behavior of hydrated cryogels were systematically investigated. The influence of two drying techniques, supercritical CO₂ drying and freeze-drying, on the structural, thermal, and mechanical properties of the dried m

Protective effects of silk fibroin against 6-OHDA in SH-SY5Y human neuroblastoma cells and comparative study with its release from gelatin films.

In this study, we analyzed the neuroprotective action of silk fibroin (SF) regenerated with calcium chloride (CaCl2), distinguishing the effects of CaCl2 and SF, and subsequently fabricating a neuroprotective hybrid material based on SF gelatin film. Cytotoxicity induced by 6-hydroxydopamine (6-OHDA) on the human neuroblastoma SH-SY5Y cell line showed that SF had a significant shielding power against 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in SH-SY5Y neuroblastoma cells, as assessed by the CCK-8 assay, cell imaging and cell cycle using flow cytometer. Specifically, the concurrent trea

In vitro study of cartilage tissue engineering using human adipose-derived stem cells induced by platelet-rich plasma and cultured on silk fibroin scaffold.

BACKGROUND: Cartilage tissue engineering is a promising technique for repairing cartilage defect. Due to the limitation of cell number and proliferation, mesenchymal stem cells (MSCs) have been developed as a substitute to chondrocytes as a cartilage cell-source. This study aimed to develop cartilage tissue from human adipose-derived stem cells (ADSCs) cultured on a Bombyx mori silk fibroin scaffold and supplemented with 10% platelet-rich plasma (PRP). METHODS: Human ADSCs and PRP were characterized. A silk fibroin scaffold with 500 μm pore size was fabricated through salt leaching. ADSCs wer

Silk fibroin nanoparticles coated with secretome-derived extracellular vesicles via GMP-ready microfluidics: a biomimetic carrier-in-carrier platform with preferential tumor cell internalization.

The integration of synthetic nanocarriers with biologically derived membranes represents a promising strategy to engineer biomimetic systems for precision medicine. Here, we report a hybrid nanoplatform in which silk fibroin nanoparticles (SFNs) are coated with the vesicular fraction of a GMP-compliant lyosecretome (herein referred to as secretome-derived EVs), in a hierarchical carrier-in-carrier architecture, enabling the combination of tunable material properties with intrinsic biological functionality. A key advancement of this work is the development of a microfluidic, GMP-compatible asse

Surface modification of vascular endothelial growth factor-loaded silk fibroin to improve biological performance of ultra-high-molecular-weight polyethylene via promoting angiogenesis.

Ultra-high-molecular-weight polyethylene (UHMWPE) has been applied in orthopedics, as the materials of joint prosthesis, artificial ligaments, and sutures due to its advantages such as high tensile strength, good wear resistance, and chemical stability. However, postoperative osteolysis induced by UHMWPE wear particles and poor bone-implant healing interface due to scarcity of osseointegration is a significant problem and should be solved imperatively. In order to enhance its affinity to bone tissue, vascular endothelial growth factor (VEGF) was loaded on the surface of materials, the loading

Three-dimensional-printed silk fibroin scaffolds loaded with adipose-derived stem cells prevent post endoscopic submucosal dissection esophageal stricture in a porcine model.

Endoscopic submucosal dissection (ESD) is the preferred treatment for early esophageal cancer. However, extensive mucosal dissection frequently results in esophageal stricture. This study aimed to evaluate the efficacy of Three-dimensional (3D)-printed silk fibroin scaffolds (SFS) loaded with adipose-derived stem cells (ADSCs) in preventing post-ESD esophageal stricture, and to explore the underlying mechanisms. A near-circumferential ESD model was established in pigs, three groups were set: Control, SFS and ADSCs-SFS. The dynamic change of postoperative esophageal healing and stricture format

Sustainable fabrication of superhydrophilic polypropylene nonwovens via tannic acid/silk fibroin coating and confined in-situ zinc oxide synthesis for robust oil/water separation.

To address the pressing issue of oily wastewater pollution, superhydrophilic membranes with favorable oleophobic properties have garnered considerable attention for their potential in oil/water separation. This study proposed a green strategy to fabricate superhydrophilic/underwater superoleophobic membranes from waste polypropylene nonwovens (PP) for efficient oil/water separation. The design leverages hydrogen-bond-driven co-assembly of silk fibroin (SF) and tannic acid (TA), whereby TA's catechol/o-benzenetriol groups form robust hydrogen bonds with SF's carbonyl residues, ensuring robust a
