Oxidized LDL can induce macrophage survival, DNA synthesis, and enhanced proliferative response to CSF-1 and GM-CSF.

Modification of low density lipoprotein (LDL), eg, by oxidation, has been proposed as being important for the formation of foam cells and therefore for the development of atherosclerotic plaques. There are a number of reports showing that macrophage-derived foam cells can proliferate in both human and animal lesions, particularly in the early phase of the disease and possibly involving macrophage-colony stimulating factor (M-CSF, or CSF-1). We studied the in vitro effects of oxidized LDL (ox-LDL) on murine bone marrow-derived macrophages (BMMs), a cell population with a high proliferative capa…

In-vitro engineering of implantable human urinary tract tissue matrices.

Due to lack or destruction of functional tissues, congenital and acquired diseases of the genitourinary tract may need, for the engineering of these tissues, biomaterials as cell carriers. Cell cultures of human urothelial and bladder smooth muscle cells were established and growth of the latter, also under mechanical stimulation was analysed. Collagen and polyesterurethane foam scaffolds were evaluated for their aptitude as cell carriers. Scaffolds made of collagens, naturally occurring extracellular matrix proteins, are biocompatible and can induce tissue regeneration and are therefore apt f

The use of acellular porcine dermis, hyaluronic acid and polynucleotides in the treatment of cutaneous ulcers: Single blind randomised clinical trial.

Reconstruction of chronic ulcers is often hampered by lack of local tissues and poor general conditions. Conservative approaches with debridement and advanced medications, such as polyurethane foam, stand as mainstays. However, the healing process is often slow, thus increasing the risk for infection or other complications. In such cases, porcine dermis (PD) and polynucleotides-added hyaluronic acid (PAHA) were previously reported to accelerate healing. The aim of the study was to compare the efficacy of PD, PAHA and polyurethane foam in chronic ulcers. Thirty patients were randomly divided in

Production of inflammatory molecules in peripheral blood mononuclear cells from severely glucose-6-phosphate dehydrogenase-deficient subjects.

OBJECTIVE: We have previously demonstrated that Mediterranean glucose-6-phosphate dehydrogenase (G6PD)-deficient peripheral blood mononuclear cells (PBMC) respond to mitogenic stimuli with a reduced cholesterol synthesis and growth. In the present study, we have investigated the release of inflammatory molecules by PBMC following a mitogenic stimulus, as well as the transformation to foam cells of monocyte-derived macrophages from severely G6PD-deficient and normal subjects. METHODS AND RESULTS: PBMC from G6PD-deficient subjects produced interleukin (IL)-1beta and IL-6 to a lower extent compa…

Nanofibrous Foams of Poly(3-hydroxybutyrate)/Cellulose Nanocrystal Composite Fabricated Using Nonsolvent-Induced Phase Separation.

In this study, we fabricated nanofibrous foams of neat poly(3-hydroxybutyrate) (PHB) and PHB/cellulose nanocrystal (CNC) nanocomposite using nonsolvent-induced phase separation (NIPS) followed by solvent extraction. Two different nonsolvents, tetrahydrofuran (THF) and 1,4-dioxane (Diox), in combination with the solvent, chloroform (CF), were used for NIPS. The parameters of NIPS-derived crystallization kinetics were calculated using Avrami analysis of time-dependent infrared spectral measurements. The lower viscosity and poorer PHB affinity of THF than those of Diox resulted in rapid crystalli

A functional polymorphism in ADAMTS7 3'-UTR abrogates miR-654-5p-mediated ADAMTS7 expression suppression and increases the incidence, short-term outcome, and recurrence of large artery atherosclerotic stroke among southern Chinese population.

BACKGROUND: A Disintegrin and Metalloproteinase with Thrombospondin motifs 7 (ADAMTS7) plays a critical role in atherosclerosis by degrading … effects. RESULTS: ADAMTS7 was highly expressed in endothelial cells, SMCs, and macrophage-derived foam cells of vulnerable human aortic plaques. Furthermore, individuals carrying the rs1045130 A allele or GA/AA genotype had a significantly higher susceptibility to LAA stroke, poorer short-term outcomes, and an increased risk of one-month recurrence. CTRP13 inhibits atherosclerosis via autophagy-lysosome-dependent degradation of CD36.

C1q/tumor necrosis factor-related protein 13 (CTRP13) is a secreted adipokine that can ameliorate abnormal glucose and lipid metabolism. However, the functional role of CTRP13 in the development of atherosclerotic plaques has yet to be described. In this study, we collected blood samples from patients of coronary artery diseases and apolipoprotein E (ApoE)-/- mice that were fed a Western diet for 12 wk to induce atherosclerosis and found that serum CTRP13 level was decreased. En face staining of aortas and aortic sinus in ApoE-/- mice showed that ectopic CTRP13 infusion in vivo dramatically de

LOX-1 in Cardiovascular Disease: A Comprehensive Molecular and Clinical Review.

LOX-1, ORL-1, or lectin-like oxidized low-density lipoprotein receptor 1 is a transmembrane glycoprotein that binds and internalizes ox-LDL in foam cells. LOX-1 is the main receptor for oxidized low-density lipoproteins (ox-LDL). The LDL comes from food intake and circulates through the bloodstream. LOX-1 belongs to scavenger receptors (SR), which are associated with various cardiovascular diseases. The most important and severe of these is the formation of atherosclerotic plaques in the intimal layer of the endothelium. These plaques can evolve into complicated thrombi with the participation …

Three-dimensional dendrite-free and anti-corrosion aluminum foam anode via hetero-metallic bismuth interface engineering for high-performance aqueous batteries.

Metallic aluminum (Al) is a promising anode material for rechargeable aqueous batteries due to its abundance, low cost, and high theoretical capacity (8056 mAh cm-3). However, the practical application of aqueous Al metal batteries faces significant challenges, including side reactions such as self-corrosion, passivation, and hydrogen evolution reaction (HER), as well as irreversible Al plating/stripping and dendrite growth. To address these issues, we propose a strategy involving the modification of conventional planar Al foil anodes into three-dimensional (3D) Al foam structures, which can e

Candida albicans Induces Foaming and Inflammation in Macrophages through FABP4: Its Implication for Atherosclerosis.

Atherosclerosis is a chronic degenerative disorder characterized by lipid-dense plaques and low-grade inflammation affecting arterial walls. Foamy macrophages are important in the formation of atherosclerotic plaques and the induction of low-grade inflammation. The presence of lipid-laden macrophages has occurred in infections caused by opportunistic pathogens. Candida albicans is the major cause of candidiasis in immunocompromised patients, including those with diabetes mellitus. However, the role played by C. albicans in macrophage foaming and the associated inflammation is poorly understood

Development and Characterisation of Gastroretentive Solid Dosage Form Based on Melt Foaming.

Dosage forms with increased gastric residence time are promising tools to increase bioavailability of drugs with narrow absorption window. Low-density floating formulations could avoid gastric emptying; therefore, sustained drug release can be achieved. Our aim was to develop a new technology to produce low-density floating formulations by melt foaming. Excipients were selected carefully, with the criteria of low gastric irritation, melting range below 70°C and well-known use in oral drug formulations. PEG 4000, Labrasol and stearic acid type 50 were used to create metronidazole dispersion whi

Differentiation factors and cytokines in the atherosclerotic plaque micro-environment as a trigger for macrophage polarisation.

The phenotype of macrophages in atherosclerotic lesions can vary dramatically, from a large lipid laden foam cell to a small inflammatory cell. Classically, the concept of macrophage heterogeneity discriminates between two extremes called either pro-inflammatory M1 macrophages or anti-inflammatory M2 macrophages. Polarisation of plaque macrophages is predominantly determined by the local micro-environment present in the atherosclerotic lesion and is rather more complex than typically described by the M1/M2 paradigm. In this review we will discuss the role of various polarising factors in regul

Design and Characterization of a Bioinspired Polyvinyl Alcohol Matrix with Structural Foam-Wall Microarchitectures for Potential Tissue Engineering Applications.

Traditional medical soft matrix used in a surgical treatment or in wound management was not good enough in both the structural support and interconnectivity to be applied in tissue engineering as a scaffold. Avian skeleton and feather rachises might be good reference objects to mimic in designing a scaffold material with good structural support and high interconnectivity because of its structural foam-wall microarchitectures and structural pneumaticity. In this study, a biomimetic airstream pore-foaming process was built up and the corresponding new medical soft matrix derived from polyvinyl a

Fabrication of uniformly cell-laden porous scaffolds using a gas-in-liquid templating technique.

Design of porous scaffolds in tissue engineering field was challenging. Uniform immobilization of cells in the scaffolds with high porosity was essential for homogeneous tissue formation. The present study was aimed at fabricating uniformly cell-laden porous scaffolds with porosity >74% using the gas-in-liquid foam templating technique. To this end, we used gelatin, microbial transglutaminase and argon gas as a scaffold material, cross-linker of the protein and porogen of scaffold, respectively. We confirmed that a porosity of >74% could be achieved by increasing the gas volume delivered to a

Recyclable Poly(urethane-urea) Covalent Adaptable Network Foams via Catalyst-, Isocyanate-, and Additive-Free Synthesis.

For decades, poly(urethane-urea) (PUU) foam production has relied on hazardous isocyanates and a complex array of external catalysts and blowing agents, resulting in chemically persistent materials that defy recycling. Herein, we break this convention by reporting the first-ever synthesis of high-performance PUU foams through an entirely autonomous, isocyanate-free, and additive-free pathway. By leveraging the ring-opening of thiazolidin-2-one with multifunctional amines, we establish a streamlined reaction that simultaneously constructs a covalent adaptable network (CAN) and triggers in situ

Extracellular vesicles and atherosclerotic peripheral arterial disease.

Atherogenesis involves a complex multifactorial process including chronic inflammation that requires the participation of several cell types and molecules. In addition to their role in vascular homeostasis, extracellular vesicles also appear to play an important role in atherogenesis, including monocyte transmigration and foam cell formation, SMC proliferation and migration, leukocyte transmigration, and thrombosis. Peripheral arterial disease, a major form of peripheral vascular disease, is characterized by structural or functional impairment of peripheral arterial supply, often secondary to
