Voltage profile along the permeation pathway of an open channel.

For ion channels, the transmembrane potential plays a critical role by acting as a driving force for permeant ions. At the microscopic level, the transmembrane potential is thought to decay nonlinearly across the ion permeation pathway because of the irregular three-dimensional shape of the channel's pore. By taking advantage of the current structural and functional understanding of cyclic nucleotide-gated channels, in this study we experimentally explore the transmembrane potential's distribution across the open pore. As a readout for the voltage drop, we engineered cysteine residues along th

Motion Compensation and Adaptive Force Control via iOCT-FBG Sensor Fusion for Robotic Subretinal Injection.

Subretinal injection is a highly delicate procedure that demands micron-level precision to avoid irreversible retinal damage. Current robotic systems achieve accurate positioning but remain limited by retinal motion and the lack of tip-force feedback. We present the first adaptive tip-force compensation framework for robotic subretinal injection, fusing intraoperative optical coherence tomography (iOCT) vision with fiber Bragg grating (FBG) force sensing. Our architecture integrates a finite-state machine (FSM) for surgical phase coordination, a Long Short-Term Memory (LSTM) enhanced residual

[2+2] Cycloaddition Sensitized by CsPbX3 Nanocrystals: Cooperative Effects of Driving Force and Exciton Confinement.

Energy-transfer-induced reactions enable efficient photochemical synthesis by harnessing long-lived triplet states that enhance stereoselectivity and suppress side reactions under mild excitation. Semiconductor nanocrystals (NCs), with rapid intersystem crossing and mild photoredox potentials, serve as highly effective triplet sensitizers. In NC-sensitized systems, energy transfer efficiency is primarily governed by the driving force and exciton confinement. To probe their individual and combined effects, we conducted a systematic study using CsPbX3 and 4-vinylbenzoic acid (4VBc) as a model sy

Impact testing as a new approach to determine mechanical strength of pharmaceutical tablets.

One of the most common standardised testing of tablet strength in the pharmaceutical industry is the tablet breaking force, which records data related to diametrical compression. This method does not account for a rapid transfer of energy such as free-falling tablets hitting a solid surface, which occurs throughout manufacture, packaging and shipping. Accordingly, the test shows poor correlation with tablet defect rate. Impact fracture force was identified as a test to measure the force absorbed by the material before fracturing when applying impact energy (dynamic stress). The testing methodo

A Systematic Approach to Predicting Spring Force for Sagittal Craniosynostosis Surgery.

Spring-assisted surgery (SAS) can effectively treat scaphocephaly by reshaping crania with the appropriate spring force. However, it is difficult to accurately estimate spring force without considering biomechanical properties of tissues. This study presents and validates a reliable system to accurately predict the spring force for sagittal craniosynostosis surgery. The authors randomly chose 23 patients who underwent SAS and had been followed for at least 2 years. An elastic model was designed to characterize the biomechanical behavior of calvarial bone tissue for each individual. After simul

Application of the FlexiForce contact surface force sensor to continuous extraocular compression monitoring during craniotomy for cerebral aneurysms.

The aims of this study were to introduce our newly developed device equipped with a contact surface force sensor (FlexiForce) for monitoring extraocular compression continuously, and to illustrate its potential clinical application using this device in patients undergoing uncomplicated frontotemporal or bifrontal craniotomy for surgical clipping of unruptured anterior circulation aneurysms. In a pilot study with volunteers, we determined the critical force of 100 gf to cause painful ocular sensation. Then we performed the bilateral extraocular force measurements in 15 patients undergoing uncom

Predicting optimal electrical stimulation for repetitive human muscle activation.

Functional electrical stimulation is the use of electrical currents to activate paralyzed muscles to produce functional movements. Muscle force output must meet or exceed the external load to maintain a posture or produce movements. A mathematical force-fatigue modeling system that predicts muscle force responses during repetitive electrical stimulation has been developed in our laboratory to help identify stimulation patterns that optimize force output for individual subjects. This study tests how well this model predicts the number of contractions that can be maintained above a required forc

Toward Chemical Accuracy in Biomolecular Simulations through Data-Driven Many-Body Potentials: I. Polyalanine in the Gas Phase.

A predictive understanding of how proteins fold, misfold, and stabilize requires accurate molecular-level insights into the thermodynamic and kinetic forces shaping their backbones. While empirical force fields remain the workhorse of biomolecular simulations, their limited functional forms often fall short in capturing the complex many-body interactions that govern protein dynamics. Quantum-mechanical methods, on the other hand, offer high accuracy but are prohibitively expensive for large biomolecules. In this work, we introduce a generalized, intramolecular formulation of the data-driven ma

Muscle recruitment strategies can reduce joint loading during level walking.

Joint inflammation, with consequent cartilage damage and pain, typically reduces functionality and affects activities of daily life in a variety of musculoskeletal diseases. Since mechanical loading is an important determinant of the disease process, a possible conservative treatment is the unloading of joints. In principle, a neuromuscular rehabilitation program aimed to promote alternative muscle recruitments could reduce the loads on the lower-limb joints during walking. The extent of joint load reduction one could expect from this approach remains unknown. Furthermore, assuming significant

The fixation tines of the Micra™ leadless pacemaker are atraumatic to the tricuspid valve.

BACKGROUND: Today, there is no manufacturer-supplied retrieval tool for the Micra™ pacemaker (Medtronic, Minneapolis, MN, USA); therefore, off-the-shelf catheters have been employed for retrievals. The proximal retrieval feature of the Micra™ can be snared and the device is then retracted from the myocardium, pulling the device through the tricuspid valve. This study characterizes the potential risks of Micra™ nitinol tine engagement with the tricuspid sub-valvular apparatus. METHODS: Fresh human hearts nonviable for transplant (n = 10) were obtained from our regional organ procurement agency

Muscle strength following direct injection of doxorubicin into rabbit sternocleidomastoid muscle in situ.

Direct intramuscular injection of doxorubicin results in permanent myofiber loss. A previous phase I trial demonstrated that such injections could successfully treat blepharospasm and hemifacial spasm. Our previous in vitro study demonstrated that doxorubicin resulted in a dose-dependent reduction in isometric force generation in sternocleidomastoid muscle in rabbits. This present study examined alterations in force generation in these treated muscles in situ, i.e., with the blood and nerve supply intact. Two months after a single doxorubicin injection, functional changes in peak twitch, tetan

Direct investigations of the effects of nicardipine on calcium channels of astrocytes by Atomic Force Microscopy.

Calcium channel blockers (CCB) of astrocytes can blockade the calcium ions entry through the voltage gated calcium channels (VGCC), and is widely used in the diseases related with VGCC of astrocytes. But many aspects of the interaction mechanisms between the CCB and VGCC of astrocytes still remain unclear due to the limited resolution of the approaches. Herein the effects of the nicardipine (a type of CCB) on VGCC of astrocytes were investigated at very high spatial, force and electrical resolution by multiple modes of Atomic Force Microscopy (AFM) directly. The results reveal that after the a

Remote Monitoring System of Dynamic Compression Bracing to Correct Pectus Carinatum.

Pectus carinatum (PC) is a chest deformity caused by disproportionate growth of the costal cartilages compared with the bony thoracic skeleton, pulling the sternum forwards and leading to its protrusion. Currently, the most common non-invasive treatment is external compressive bracing, by means of an orthosis. While this treatment is widely adopted, the correct magnitude of applied compressive forces remains unknown, leading to suboptimal results. Moreover, the current orthoses are not suitable to monitor the treatment. The purpose of this study is to design a force measuring system that could

Exploring the role of ankle muscle function in gait impairments and fall risk in Parkinson's disease.

INTRODUCTION: Parkinson's disease (PD) significantly impacts mobility, with gait disturbances and muscle impairments contributing to a fall risk five times higher than similarly aged adults. Falls significantly impact the quality of life in those with PD, yet the role of ankle muscle function in gait disturbances remains underexplored. This study investigated whether deficits in ankle force and steadiness contribute to gait variability and fall risk, potentially uncovering therapeutic targets for fall prevention in individuals with PD compared with age-matched older adults (OA). METHODS: A ca

Biomechanical Study of a Tricompartmental Unloader Brace for Patellofemoral or Multicompartment Knee Osteoarthritis.

Objective: Off-loader knee braces have traditionally focused on redistributing loads away from either the medial or lateral tibiofemoral (TF) compartments. In this article, we study the potential of a novel "tricompartment unloader" (TCU) knee brace intended to simultaneously unload both the patellofemoral (PF) and TF joints during knee flexion. Three different models of the TCU brace are evaluated for their potential to unload the knee joint. Methods: A sagittal plane model of the knee was used to compute PF and TF contact forces, patellar and quadriceps tendon forces, and forces in the anter

Condensate interfacial forces reposition DNA loci and probe chromatin viscoelasticity.

Biomolecular condensates assemble in living cells through phase separation and related phase transitions. An underappreciated feature of these dynamic molecular assemblies is that they form interfaces with other cellular structures, including membranes, cytoskeleton, DNA and RNA, and other membraneless compartments. These interfaces are expected to give rise to capillary forces, but there are few ways of quantifying and harnessing these forces in living cells. Here, we introduce viscoelastic chromatin tethering and organization (VECTOR), which uses light-inducible biomolecular condensates to g

Low-frequency fatigue in isolated skeletal muscles and the effects of methylxanthines.

1. A form of skeletal muscle fatigue was examined with isolated animal and human muscle preparations. The possibility that methylxanthines could overcome this was investigated. 2. Prolonged contractile activity resulted in a long-lasting impairment of force generation at low frequencies of stimulation at times when the force at higher frequencies had substantially recovered. This was seen with both fast-twitch and slow-twitch animal muscles and with samples of isolated human muscle. 3. The decrease in low-frequency force was due to a decrease in twitch amplitude, suggesting damage to the proce

GDF15 Contributes to the Regulation of the Mechanosensitive Responses of PdL Fibroblasts through the Modulation of IL-37.

During orthodontic tooth movement (OTM), areas of compressive and tensile forces are generated in the periodontal ligament (PdL), a mechanoreactive connective tissue between the teeth and alveolar bone. Mechanically stimulated PdL fibroblasts (PdLFs), the main cell type of PdL, express significantly increased levels of growth differentiation factor 15 (GDF15). In compressed PdL areas, GDF15 plays a fundamental role in modulating relevant OTM processes, including inflammation and osteoclast activation. However, the specific function of this factor in tensile areas has not yet been investigated.
