Elucidation of the hetero-dimeric binding activity of LasR and RhlR proteins with the promoter DNA and the role of a specific Phe residue during the biosynthesis of HCN synthase from opportunistic pathogen Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic human pathogen. It causes secondary infections in patients suffering from cancer and other immunological disorders. The pathogenicity of the organism is dependent on the ability of the organism to code for hydrogen cyanide (HCN), the synthesis of which is mediated by HCN synthase enzyme. HCN synthase is encoded by hcnABC operon. The transcription of the operon is controlled by a complex interplay between the proteins LasR and RhlR. Till date, there is no report that deals with the binding interactions of the RhlR-LasR heterodimer with the promoter DNA

Dynamics of ex vivo cytokine transcription during experimental Toxocara canis infection in Balb/c mice.

The cytokine microenvironment is crucial in generating and polarizing the immune response. A means of monitoring this environment would be of great value for better understanding Toxocara canis immune modulation. The aim of this study was to analyze the dynamics of cytokine transcription ex vivo, during early (24-48 hours) and late (15-30 days) times post-infection, in the mesenteric lymph nodes, spleen and intestinal mucosa of Balb/c mice experimentally infected with T. canis larvae. Mice in the treated group were infected with 100 third-stage larvae (L3), whereas mice in the control group we

Colony size and initial conditions combine to shape colony reunification dynamics.

Group cohesion and collective decision-making are important for many social animals, like social insects, whose societies depend on the coordinated action of individuals to complete collective tasks. A useful model for understanding collective, consensus-driven decision-making is the fluid nest selection dynamics of ant colonies. Certain ant species oscillate between occupying multiple nests simultaneously (polydomy) and reuniting at a single location (monodomy), but little is known about how colonies achieve a consensus around these dynamics. To investigate the factors underpinning the splitt

Pre-existing mutations related to tenofovir in chronic hepatitis B patients with long-term nucleos(t)ide analogue drugs treatment by ultra-deep pyrosequencing.

AIMS: The dynamics of resistance-associated mutations under combination therapy were explored. METHODS: A total of 46 patients were classified into adefovir (n=14) and entecavir (n=32) groups. In the adefovir (ADV) group, six patients receiving combined therapy were DNA-positive after more than 3 years of therapy. Ultra-deep pyrosequencing was used to analyze the dynamics of multi-drugs resistance mutations. RESULTS: At baseline, all 46 treatment-naïve patients harbored rtA181V/T substitutions (1.2%-4.6%) and rtN236T substitutions (1.6%-6.1%). In the ADV group, eight patients with long-term

Comparative Study of Premixed Versus Sequential Administration of Hyperbaric Bupivacaine and Fentanyl for Subarachnoid Block in Caesarean Section: A Randomized Controlled Trial.

BACKGROUND: Spinal anaesthesia is the standard technique for elective caesarean section due to its simplicity, rapid onset, and maternal safety profile. While combining hyperbaric bupivacaine with intrathecal fentanyl enhances anaesthetic efficacy, the sequential drug administration may influence the spread, block dynamics, and haemodynamic stability. METHODS: In this prospective, randomised controlled study, 180 ASA I-II parturients were allocated to either Group M (premixed bupivacaine 9 mg + fentanyl 15 mcg) or Group S (sequential administration of the same drugs). Primary outcomes were ti

Cognitive-behavioral group therapy versus group psychotherapy for social anxiety disorder among college students: a randomized controlled trial.

OBJECTIVE: In this randomized controlled trial, cognitive-behavioral group therapy (CBGT) for social anxiety disorder (SAD) was compared to group psychotherapy (GPT), a credible, structurally equivalent control condition that included only nonspecific factors of group treatment (such as group dynamics). METHODS: Participants were 45 college students at the University of Colorado with a primary diagnosis of SAD. Each treatment condition comprised eight group sessions lasting 2 hr each. Independent assessors (blind to treatment assignment) assessed participants at baseline and posttreatment wit…

Quantitative radio-isotope scanning of the sacroiliac joints in ankylosing spondylitis.

A method for applying 99mTc-MDP for dynamic and static quantitative radioisotope scanning (QRS) of the sacroiliac joints (SI) in early progressive sacroiliitis in ankylosing spondylitis (AS) is described. In a prospective study, 2 groups of male AS patients were investigated, one with increased elevated erythrocytic sedimentation rate (ESR) (group A, n = 7) and one with normal ESR (group B, n = 8). In both groups an increased uptake of the radiotracer was found in the static part of the study versus a control group C (n = 9). An increased uptake versus group C was also found for group A in the

Conformational properties of penicillins: quantum chemical calculations and molecular dynamics simulations of benzylpenicillin.

Herein, we present theoretical results on the conformational properties of benzylpenicillin, which are characterized by means of quantum chemical calculations (MP2/6-31G* and B3LYP/6-31G*) and classical molecular dynamics simulations (5 ns) both in the gas phase and in aqueous solution. In the gas phase, the benzylpenicillin conformer in which the thiazolidine ring has the carboxylate group oriented axially is the most favored one. Both intramolecular CH. O and dispersion interactions contribute to stabilize the axial conformer with respect to the equatorial one. In aqueous solution, a molecul

On the critical behaviour of simple epidemics.

We show how ideas and models which were originally introduced to gain an understanding of critical phenomena can be used to interpret the dynamics of epidemics of communicable disease in real populations. Specifically, we present an analysis of the dynamics of disease outbreaks for three common communicable infections from a small isolated island population. The strongly fluctuating nature of the temporal incidence of disease is captured by the model, and comparisons between exponents calculated from the data and from simulations are made. A forest-fire model with sparks is used to classify th

[Dynamic alterations in regional brain activity associated with bipolar disorder and smoking status].

OBJECTIVES: Patients with bipolar disorder (BD) have a high prevalence of smoking. Smoking status may interact with disease-related brain functional abnormalities and jointly influence resting-state brain activity. This study aims to investigate the independent and interactive effects of BD diagnosis and smoking status on the dynamic instability of regional brain activity. Based on resting-state functional magnetic resonance imaging (fMRI) data, dynamic amplitude of low-frequency fluctuation (dALFF), dynamic fractional amplitude of low-frequency fluctuation (dfALFF), and dynamic regional homog

Sex-specific biological susceptibility and sexual mixing patterns in herpes simplex virus type 2 transmission: a mathematical modeling study.

BACKGROUND: Herpes simplex virus type 2 (HSV-2) infection is a lifelong sexually transmitted infection with substantial disease and economic burdens. Despite its global impact, key features of its transmission dynamics-including sex-specific biological susceptibility (the inherent likelihood of acquiring infection upon exposure, independent of behavioral factors) and sexual mixing patterns (how individuals form sexual partnerships across age and risk groups)-remain poorly quantified. METHODS: A population-level mathematical model of HSV-2 transmission dynamics was applied to heterosexual tran

Neuronal degeneration and cognitive impairment can be prevented via the normalization of mitochondrial dynamics.

Neuronal cells possess a certain degree of plasticity to recover from cell damage. When the stress levels are higher than their plasticity capabilities, neuronal degeneration is triggered. However, the factors correlated to the plasticity capabilities need to be investigated. In this study, we generated a novel mouse model that able to express in an inducible manner a dominant-negative form of MFN2, a mitochondrial fusion factor. We then compared the phenotype of the mice continuously expressing the mutated MFN2 with that of the mice only transiently expressing it. Remarkably, the phenotypes o

Reciprocal escalation of violent extremism: Experimental and longitudinal evidence from Denmark.

Recent surges in intergroup conflict across racial, religious, and national lines highlight how mutual perceptions of threat can lead to escalating cycles of hostility. These cycles are believed to be driven by interconnected psychological, sociopolitical, and cultural mechanisms, with each group responding aggressively to perceived hostility from the other. Yet, systematic exploration of the psychological mechanisms behind reciprocally escalating violence and negativity remains limited. This research uniquely integrates experimental and longitudinal methodologies to provide novel, causal insi
