OBJECTIVE: The purpose of this study was to investigate the role of serotonin (5-HT) in the effects of oral 3,4-methylenedioxymethamphetamine (MDMA) in humans.
MATERIALS AND METHODS: The subjective and physiological effects of 1.5 mg/kg MDMA were evaluated after 20 mg fluoxetine in eight recreational MDMA users in a double-blind, placebo-controlled study. During phase 1, participants were maintained on placebo for at least 5 days and tested with MDMA and placebo on separate sessions. In phase 2, the procedure was the same except fluoxetine was administered daily for at least 5 days. During se

3,4-methylenedioxymethamphetamine (MDMA), an amphetamine derivative known as ecstasy, has stimulating and hallucinogenic properties. It has become a substance that is widely used especially by young people. Hepatotoxicity is one of the rare side effects of this substance and can be fatal. Ecstasy-induced fulminant hepatitis has been reported in case reports. The clinical course and the prognosis of the cases may differ. In this article, two cases in whom ecstasy-induced fulminant hepatic failure had developed and who were treated with liver transplantation, and one case which recovered with tr

The re-emergence of serotonergic psychedelics has introduced rapid-acting antidepressant effects and has been widely described as a paradigmatic shift in depression treatment. Recently, there has been intense debate about whether the acute subjective effects of psychedelics are necessary for their therapeutic efficacy or merely byproducts of underlying neuroplastic processes. Recent conceptualizations of "psychoplastogens" emphasize rapid induction of neuroplasticity as a potential common pathway across mechanistically diverse rapid-acting antidepressants. Here, some argue that the psychedelic

Current research into the actions of psychedelic drugs is focused on 5-HT2A receptor-coupled signaling pathways, particularly β-arrestin2 signaling. Here, we investigated the involvement of β-arrestin2 in psychedelic drug-induced mouse markers of hallucinogenic effects, molecular plasticity, and structural plasticity; specifically, the head-twitch response, cortical expression of plasticity-related genes (cFos, Egr1, Egr2, and Arc), and dendritogenesis (primary cortical neuron cultures), respectively. Psilocin-induced head-twitches, plasticity gene expression, and dendritogenesis were all bloc

BACKGROUND: Chronic use of cannabis is associated with an increased risk of psychosocial, mental and physical health impairments. Sociohealth institutions reach a very limited proportion of cannabis users in need of treatment. Using data collected from festival attendees, this study aimed to estimate the prevalence of dependent cannabis users and to characterize cannabis dependence.
METHODS: We used data from the cross-sectional OCTOPUS survey carried out at 13 music events in the French department of Loire-Atlantique between July 2017 and July 2018. 383 participants aged 18 or older underwen

Psilocybin and its active metabolite psilocin are hallucinogenic serotonergic agonists with high affinity for several serotonin receptors. In addition to underlying the hallucinogenic effects of these compounds, serotonin receptor activation also has important effects on decision-making and goal-directed behaviors. The impact of psilocybin and psilocin on these cognitive systems, however, remains unclear. This study investigated the effects of psilocybin treatment on decision-making and motivation in healthy male and female rats. We compared probability and delay discounting performance of psi

Fungi with hallucinogenic properties and neurotoxicity have been listed as prohibited drugs in recent years, but there is a lack of in situ quantification of psilocybin and analogues in these samples to avoid the decomposition of these psychoactive tryptamines in time-consuming sample preparation. In this study, matrix-assisted laser-desorption/ionization (MALDI)-Fourier transform ion cyclotron resonance (FT ICR) mass spectrometric imaging (MSI) was used to analyze the distribution of psilocybin and its analogues in hallucinogenic Psilocybe mushrooms. BACKGROUND: Visual phenomena are of many different kinds; their differential diagnosis is usually possible with directed history-taking. In this review, we describe common visual phenomena that must be distinguished from a migraine aura.
METHODS: This review is based on publications retrieved by a selective search in PubMed and the Web of Knowledge/Science, with special attention to the current classification of the International Headache Society and the recommendations of the German Migraine and Headache Society. The following search terms were used: "visual phenomena/phenomenon," "migraine

There is little in the literature about treatment of persons with problems with "club" or "party" drugs. This paper looks at the characteristics of individuals admitted to treatment for primary, secondary, or tertiary problems with club drugs such as ecstasy, gamma-hydroxybutyrate (GHB), ketamine, flunitrazepam (Rohypnol), methamphetamine, and hallucinogens (e.g., LSD) in programs funded by the Texas Commission on Alcohol and Drug Abuse. Some 38,350 unduplicated records from 1988 through 2003 of persons admitted with problems with club drugs were compared against users of alcohol or other drug…

BACKGROUND: Ketamine has been associated with a unique spectrum of subjective "psychedelic" effects in patients emerging from anesthesia. This study quantified these effects of ketamine and related them to steady-state plasma concentrations.
METHODS: Ketamine or saline was administered in a single-blinded crossover protocol to 10 psychiatrically healthy volunteers using computer-assisted continuous infusion. A stepwise series of target plasma concentrations, 0, 50, 100, 150, and 200 ng/ml were maintained for 30 min each. After 20 min at each step, the volunteers completed a visual analog (VAS

Skilled interpersonal support and safety monitoring can enhance the therapeutic potential of psychedelic compounds and reduce the potential for harm. In the anticipation of legalized psychedelic care, and the recognition of ongoing psychedelic use across a variety of contexts, numerous training programs have been established to educate psychedelic facilitators in supporting individuals who take psychedelics. However, psychedelic facilitation training in the US has not been well characterized, making it difficult to appraise the strengths, needs, and gaps in this emerging discipline, especially

Psychedelic experiences involving extreme feelings of horror, helplessness, and perceived threats can be traumatizing. Traumatic psychedelic experiences are a rare, extreme, and largely preventable form of challenging experience which can arise due to frightening psychedelic drug effects, unsafe settings, and emergence of pre-existing trauma. Some people recover quickly, but others develop prolonged anxiety, sleep disturbances, derealization, or other potentially trauma-related symptoms. This chapter discusses the causes, phenomenology, and potential outcomes of traumatic psychedelic experienc

The subacute period following the use of classic psychedelics is often marked by an "afterglow" - a state characterized by elevated mood; enhanced psychological well-being; increased emotional openness toward self, others, and nature; and sometimes a heightened sense of clarity and existential meaning. From a neurobiological perspective, subacute psychedelic states have been associated with changes in brain network connectivity, amygdala reactivity, and neuroplasticity. However, individual responses to psychedelics vary considerably, and not all users experience positive aftereffects. Some stru

Psychedelics have been studied since the 1950s as a potential treatment for alcohol use disorder (AUD), with over a dozen clinical trials of lysergic acid diethylamide (LSD), and several contemporary trials of psilocybin and ayahuasca for this indication. Herein, we characterize foundational studies from the 1950s to the present, with emphasis on key design factors that varied considerably between published studies. Critically, those design factors include pharmacological factors, such as presence or absence of a placebo control and the nature of the placebo (e.g., ephedrine, dextroamphetamine

When used clinically, psychedelics may appear unusual or even unique when compared to more familiar or long-standing medical interventions, prompting some to suggest that the ethical issues raised may likewise be exceptional. If that is correct, then perhaps psychedelics should be treated differently from other medical substances: for example, by being subjected to different ethical or evidentiary standards. Alternatively, it may be that psychedelics have more in common with various existing medical interventions than first meets the eye. We argue in favor of the latter position, drawing on pa

The authors offer a model for curriculum for education and training in substance-assisted psychotherapy (SAP), that is, psychedelic, psycholytic, and entactogen/MDMA (3,4-methylenedioxymethamphetamine)-assisted psychotherapy, addressing both the detailed contents of training and the question of experiential training. All authors of this model have an abiding interest and extensive experience in both the theory and practical aspects of SAP and questions relating to training. The model curriculum has been written through an international consensus building process and represents a consensus stat

Blackburne et al. track the electroencephalogram activity of volunteers inhaling a high dose of the powerful psychedelic 5-methoxy-N,N-dimethyltryptamine, revealing profoundly slowed-down brain activity but no significant reduction of alpha band power that is typical of other psychedelics.1.

"Psychedelic research - Going global" is a Special Issue of the Journal of Psychopharmacology that has been compiled and published as a tribute to Dr. Roland R Griffiths in recognition of his pioneering and pivotal contribution to the field of psychedelic research. Dr. Griffiths was Professor in the Departments of Psychiatry and Neurosciences at Johns Hopkins University School of Medicine, and latterly, the founding director of the Center for Psychedelic and Consciousness Research. It is not an exaggeration to state that his discoveries, indefatigable support and advocacy laid the foundations

Psychedelic use in clinical settings is emerging as a way to treat a variety of psychiatric illnesses. However, psychedelic use primarily occurs in recreational settings, and the absence of standard protocols elevates the risk of users experiencing adverse reactions or "bad trips." This article discusses the use of and risks associated with "trip killers" or "trip sitters," and key considerations for nurses when caring for patients who use any of these drugs.
