Amygdala or hippocampus damage only minimally impacts affective responding to threat.

Decades of research studying the behavioral effects of damage to structures in medial temporal lobe of rhesus monkeys have documented that s … e evaluated the responses of rhesus monkeys with bilateral ibotenic acid lesions of the amygdala or hippocampus and procedure-matched control animals to the presentation of an unfamiliar human intruder and threatening objects of varying complexity. All animals behaved as expected-calibrating their responses to the ostensible threat value of the stimuli such t …

Fetal inflammation induces acute immune tolerance in the neonatal rat hippocampus.

BACKGROUND: Infants born preterm due to chorioamnionitis are frequently affected by a fetal inflammatory response syndrome (FIRS) and then by subsequent postnatal infections. FIRS and postnatal systemic inflammatory events independently contribute to poor neurocognitive outcomes of preterm infants. Developmental integrity of the hippocampus is crucial for intact neurocognitive outcomes in preterms and hippocampally dependent behaviors are particularly vulnerable to preterm systemic inflammation. How FIRS modulates the hippocampal immune response to acute postnatal inflammatory events is not we…

Altered Effective Connectivity of Bilateral Hippocampus in Type 2 Diabetes Mellitus.

Patients with type 2 diabetes mellitus (T2DM) experience cognitive deficits but the underlying pathophysiologic mechanisms are not known. We therefore applied Granger causality analysis of resting-state functional magnetic resonance imaging to study the effective connectivity (EC) of the hippocampus in patients with T2DM. Eighty six patients with T2DM and 84 matched healthy controls (HC) were recruited. The directional EC between anatomically defined seeds in left hippocampus (LHIP) and right hippocampus (RHIP) and other brain regions was compared between T2DM and HC and Pearson correlation an…

Overshooting Subcellular Redox-Responses in Rett-Mouse Hippocampus during Neurotransmitter Stimulation.

Rett syndrome (RTT) is a neurodevelopmental disorder associated with disturbed neuronal responsiveness and impaired neuronal network function. Furthermore, mitochondrial alterations and a weakened cellular redox-homeostasis are considered part of the complex pathogenesis. So far, overshooting redox-responses of MeCP2-deficient neurons were observed during oxidant-mediated stress, hypoxia and mitochondrial inhibition. To further clarify the relevance of the fragile redox-balance for the neuronal (dys)function in RTT, we addressed more physiological stimuli and quantified the subcellular redox r

Hippocampus-driving progressive structural alterations in medication-naïve major depressive disorder.

BACKGROUND: Major depressive disorder (MDD) is associated with abnormalities in brain structure. However, structural abnormality findings ha … MDD patients and healthy controls, gray matter was greater in the bilateral amygdala, the bilateral hippocampus, the left parahippocampus, and the right fusiform, while it was lessened in the bilateral brainstem, the bilateral pallidum, and the bilateral thalamus. Selecting the hippocampus as the seed region to run further CaSCN analysis revealed that the hip …

Chronic interleukin-1β in the dorsal hippocampus impairs behavioural pattern separation.

Understanding the long-term consequences of chronic inflammation in the hippocampus may help to develop therapeutic targets for the treatment of cognitive disorders related to stress, ageing and neurodegeneration. The hippocampus is particularly vulnerable to increases in the pro-inflammatory cytokine interleukin-1β (IL-1β), a mediator of neuroinflammation, with elevated levels implicated in the aetiology of neurodegenerative diseases such as Alzheimer's and Parkinson's, and in stress-related disorders such as depression. Acute increases in hippocampal IL-1β have been shown to impair cognition…

B23/Nucleophosmin promotes reconstitution of synaptic path in hippocampus after injury.

B23, also known as nucleophosmin (NPM), is multifunctional protein directly implicated in cell proliferation, cell cycle progression, and cell survival. In the current study, in addition to confirming its anti-apoptotic function in neuronal survival, we demonstrated that the spatial-temporal expression profile of B23 during development of hippocampal neurons is high in the embryonic stage, down-regulated after birth, and preferentially localized at the tips of growing neuritis and branching points. Overexpression of B23 promotes axon growth with abundant branching points in growing hippocampal

p21-activated kinase 1 restricts tonic endocannabinoid signaling in the hippocampus.

PAK1 inhibitors are known to markedly improve social and cognitive function in several animal models of brain disorders, including autism, but the underlying mechanisms remain elusive. We show here that disruption of PAK1 in mice suppresses inhibitory neurotransmission through an increase in tonic, but not phasic, secretion of endocannabinoids (eCB). Consistently, we found elevated levels of anandamide (AEA), but not 2-arachidonoylglycerol (2-AG) following PAK1 disruption. This increased tonic AEA signaling is mediated by reduced cyclooxygenase-2 (COX-2), and COX-2 inhibitors recapitulate the

Low-level laser therapy for beta amyloid toxicity in rat hippocampus.

Beta amyloid (Aβ) is well accepted to play a central role in the pathogenesis of Alzheimer's disease (AD). The present work evaluated the therapeutic effects of low-level laser irradiation (LLI) on Aβ-induced neurotoxicity in rat hippocampus. Aβ 1-42 was injected bilaterally to the hippocampus CA1 region of adult male rats, and 2-minute daily LLI treatment was applied transcranially after Aβ injection for 5 consecutive days. LLI treatment suppressed Aβ-induced hippocampal neurodegeneration and long-term spatial and recognition memory impairments. Molecular studies revealed that LLI treatment: …

Greater glucocorticoid receptor activation in hippocampus of aged rats sensitizes microglia.

Healthy aging individuals are more likely to suffer profound memory impairments following an immune challenge than are younger adults. These challenges produce a brain inflammatory response that is exaggerated with age. Sensitized microglia found in the normal aging brain are responsible for this amplified response, which in turn interferes with processes involved in memory formation. Here, we examine factors that may lead aging to sensitize microglia. Aged rats exhibited higher corticosterone levels in the hippocampus, but not in plasma, throughout the daytime (diurnal inactive phase). These …

Time-limited involvement of dorsal hippocampus in unimodal discriminative contextual conditioning.

Converging evidence examining the effects of post-training manipulations of the hippocampus suggests that the hippocampus may play a time-limited role in the maintenance of a variety of forms of memory. In particular, either lesions or inactivation of the dorsal hippocampus results in many cases in a time-limited retrograde impairment in nondiscriminative contextual conditioning paradigms. However, the extent to which hippocampal manipulations result in a time-limited retrograde amnesia for a variety of forms of learning has recently been called into question (reviewed in Sutherland, Sparks, &…

Indoleamine 2,3 dioxygenase and quinolinic acid immunoreactivity in Alzheimer's disease hippocampus.

The present immunohistochemical study provides evidence that the kynurenine pathway is up-regulated in Alzheimer's disease (AD) brain, leading to increases in the excitotoxin quinolinic acid (QUIN). We show that the regulatory enzyme of the pathway leading to QUIN synthesis, indoleamine 2,3 dioxygenase (IDO) is abundant in AD compared with controls. In AD hippocampus, both IDO- and QUIN-immunoreactivity (-IR) was detected in cortical microglia, astrocytes and neurones, with microglial and astrocytic expression of IDO and QUIN highest in the perimeter of senile plaques. QUIN-IR was present in g…

Traumatic brain injury regulates adrenocorticosteroid receptor mRNA levels in rat hippocampus.

Glucocorticoid activation of two types of adrenocorticosteroid receptors (ACRs), the mineralocorticoid receptor (MR) and glucocorticoid receptor (GR), influences the vulnerability of hippocampal neurons to insult. To examine the potential impact of ACR activation following traumatic brain injury (TBI), the current study assesses regulation of MR and GR expression and glucocorticoid levels following controlled cortical impact (CCI). Male Sprague-Dawley rats were pretreated for 48 h with vehicle, the MR antagonist spironolactone, or the GR antagonist mifepristone (RU486). On day three, subjects

Mineralocorticoid receptors regulate bcl-2 and p53 mRNA expression in hippocampus.

The present study addressed the hypothesis that the neuronal mineralocorticoid receptor (MR) regulates genes associated with cell death, such as bax and p53, and cell viability, including bcl-2, BDNF, and NT-3. Rats were pretreated with either oil vehicle or the MR antagonist spironolactone (SPIRO) and subsequently injected with saline or kainic acid (KA). MR blockade significantly decreased basal mRNA expression of bcl-2 in CA1 of saline-treated animals and attenuated KA-induced increases in p53 mRNA levels in CA3. SPIRO pretreatment had no significant effect on expression of bax, NT-3, or BD

Pharmacologic characterization of GABAA/benzodiazepine receptor in rat hippocampus during aging.

We have characterized the pharmacologic properties of the gamma-aminobutyric acid (GABAA)/benzodiazepine receptor complex in hippocampal membranes from 3-month- and 24-month-old Wistar rats. No major changes were found in [3H]flunitrazepam or [3H]muscimol binding parameters. Neither the dissociation constant(s) nor the Bmax for either ligand seemed to be modified during aging in hippocampus. Furthermore, the allosteric interaction between the barbiturates binding site and the GABA binding site, determined by pentobarbital stimulation of [3H] muscimol binding, remained unaltered. However, there…

Histology-guided spatial lipidomics and proteomics of the trisynaptic circuit in the human hippocampus.

The human hippocampal trisynaptic circuit activity is essential for learning and memory. This canonical circuit has spatially distinct popul … of MALDI-MSI-LCMD-lipidomic and proteomic analysis to the trisynaptic circuit. Our MALDI-MSI of the hippocampus revealed that the of mass-to-charge ions of the cornu ammonis 1 (CA1) and cornu ammonis 3 (CA3) did not segment from the surrounding tissue. Thus, we developed a novel histology-guided MALDI-MS imaging-LCMD-spatial lipidomic/proteomic pipeline with …

Immune imbalance in the human hippocampus in PTSD revealed by single-nucleus transcriptomics.

INTRODUCTION: Post-traumatic stress disorder (PTSD) is increasingly recognized as a neuroimmune disorder in which disrupted neuron-glia interactions contribute to long-term cognitive and emotional dysfunction. However, the cellular and molecular basis of immune imbalance in the human hippocampus remains unclear. METHODS: We performed single-nucleus RNA sequencing on postmortem hippocampal tissues from donors with PTSD and matched controls, identifying 10 major cell types, with particular emphasis on neurovascular and glial populations. Differential expression, pathway enrichment, pseudotime t…

Hippocampus-Ventral Striatum Connectivity Is Associated With Binge Drinking Frequency in Adults.

Contextual information, such as spatial locations and social groups, are known to propagate reward-seeking behavior including binge drinking. Animal models suggest that context-evoked drug and alcohol seeking emerges from hippocampal projections to the ventral striatum (i.e., nucleus accumbens). While binge drinking has been associated with adverse structure and function of both the ventral striatum and hippocampus, the anatomical relevance of this circuit in humans has largely been inferred from volumetric studies investigating these regions separately. Using diffusion MRI tractography in adu…

The hippocampus under influence: structural effects of synthetic and natural psychotropic medications.

Normal functioning of the brain consists of a complex set of physiological interactions occurring within specific anatomical sites of the brain. One of the most important areas within the brain responsible for a cohort of these homeostatic actions is the hippocampus. The hippocampus serves as a critical nexus for memory, emotion, and cognition. However, the widespread and often long-term use of psychotropic medications, particularly benzodiazepines, has raised concern regarding their potential structural and functional effects on this vulnerable region. Chronic benzodiazepine use remains commo…
