Pruritus is a frequent symptom in chronic cholestatic liver disease. To date, no single causative mechanism has been identified. We examined venous plasma concentrations of the known pruritogen, histamine, using a highly sensitive radioenzymatic assay in 42 patients with chronic cholestatic liver disease, and in normal controls. The mean plasma histamine level was significantly greater in chronic cholestatic liver disease patients (275 (117) pg/ml; X (SD) than in controls (140 (72) pg/ml, n = 20) (p less than 0.0001). No significant differences were found between histamine concentrations in th…

Allergic rhinitis is characterised by symptoms of sneezing, itching of the nose with watery secretions, and nasal obstruction. We have previously shown that patients can have the diagnosis of allergic rhinitis confirmed by nasal provocation tests and assessment of nasal inspiratory peak flow (NIPF) after specific allergen or hyperosmolar challenge. We now show that histamine is released into the nasal lavage fluid in response to such challenges. Saline lavage alone results in detectable histamine levels in the order of 5 ng/ml, but in the presence of allergen (HDM) there is a significant incre…

Epidemiologic evidence has shown that elevated levels of high-density lipoproteins (HDL) protect against the development of coronary heart disease (CHD). These observations have prompted the evaluation of various factors thought to affect HDL and its subspecies, HDL2 and HDL3. Numerous behavioral and physiologic factors have been shown to elevate HDL levels. These are currently being researched as potential tools in preventing CHD. Several pharmacologic agents are known to alter HDL levels. Studies show that patients with peptic ulcer disease treated with the H2-receptor antagonist cimetidine

C5a, a peptide derived from the fifth component of complement, caused significant prolonged smooth muscle contraction in isolated guinea pig trachea. Diphenhydramine, a histamine receptor antagonist of the H1 type, had no effect on either the rate, amplitude or duration of C5a induced tracheal contraction, whereas it significantly inhibited the tracheal response to exogenous histamine. Diphenhydramine also caused a significant delay in the response to compound 48/80 in normal guinea pig trachea and to antigen in actively sensitized trachea, indicating that the antihistamine was capable of inhi…

Histamine-H2-receptor antagonists (Burimamide, Metiamide, Cimetidine) may inhibit histamine-stimulated gastric secretion in various species including man; in constrast, conventional antihistaminics (H1-antagonists) do not do so. Gastric secretion as stimulated by pentagastrin, insulin, 2-deoxy-glucose or an intragastrically instilled meal is also decreased by H2-antagonists, and a final common mediator role for histamine in the gastric secretory is discussed. H2-antagonists do also inhibit pancreatic enzyme secretion, but do not affect at all bicarbonate secretion. Recent clinical trials have …

Mitogens and antigens have been the traditional ligands for activating lymphocytes in vitro for the elaboration of lymphokines. Recently, histamine, by interaction with histamine-type 2 receptors on T lymphocytes, has been found to induce the production of one lymphokine, histamine-induced suppressor factor (HSF), that inhibits lymphocyte proliferation and lymphokine production in vitro. Because the biologic effects of HSF appear to be confined to alterations in lymphocyte function, we assessed the ability of soluble products of histamine-stimulated human blood mononuclear cells to affect anot…

BACKGROUND: Allergy can be diagnosed using basophil tests. Several methods measuring basophil activation are available. This study aimed at comparing basophil activation test (BAT), histamine release assay (HR), and passive sensitization histamine release assay (passive HR) in the diagnosis of peanut allergy.
METHODS: BAT, HR, and passive HR were performed on 11 peanut-allergic and 14 nonallergic subjects. Blood was incubated with peanut extract or anti-IgE and tests were performed as follows: BAT-CD63 upregulation was assessed by flow cytometry; HR-released histamine was quantified by a glas…

UNLABELLED: It is assumed that an overproduction of gastric acid is the most important factor in the development of peptic ulcer. However, it has been also demonstrated that gastric defense mechanisms, which prevent mucosal injury, are enhanced by the same factors that increase acid secretion. The aim of this study was to examine the role of capsaicin-sensitive sensory nerves and histamine H1, H2, and H3 receptors in histamine-induced gastroprotection against stress ulcers. Studies were performed on rats with intact or ablated sensory nerves. Ablation of sensory nerves was induced by neurotoxi…

A relatively specific method for the quantitative assay of histamine H2-receptor antagonists has been developed. The method is based on the antagonism of the histamine-induced, and the spontaneous, gastric acid secretion in the stomach-perfused, urethane-anesthetized acute rat. For the induced gastric acid secretion, the animal received two consecutive equal injections of histamine (2 mg/kg, intrajugularly), the second 1.7 h after the termination of the first histamine-induced acid hypersecretion. Atropine (5 mg/kg), chlorisondamine (2 mg/kg), imipramine (10 mg/kg) and tripelennamine (5 mg/kg)…

Histamine receptors (HRH1-HRH4) are G-protein coupled receptors (GPCRs) that mediate essential physiological functions, including neurotransmission, gastric acid secretion, immune regulation, and presynaptic autoregulation. Despite their importance, systematic comparative analyses across mammalian histamine receptor sequences remain scarce. In this study, we performed a comprehensive evaluation of HRH1-HRH4 across multiple mammalian species, integrating sequence homology, invariant residue mapping, amino acid substitutions, compositional frequencies, Shannon entropy, polystring distributions, …

BACKGROUND AND AIMS: Histamine signalling is critical in heart failure (HF). Histamine N-methyltransferase (HNMT) serves as the primary enzyme responsible for histamine clearance in heart, but its role in HF remains undefined. This study investigates HNMT-mediated metabolic-epigenetic crosstalk in HF and evaluates therapeutic strategies targeting this axis.
METHODS: Cardiomyocyte-specific Hnmt-knockout and Hnmt-overexpression mice were generated. Transverse aortic constriction (TAC) was applied to induce HF in mice. RNA sequencing and targeted metabolomics were employed to explore mechanisms …

The common bed bug (Cimex lectularius L.) is a hematophagous pest species that lives in close proximity to humans. Following a blood meal, bed bugs deposit fecal material indoors. The feces contain a variety of compounds, including histamine, which serves as a component of their aggregation pheromone. Histamine is a pivotal mammalian immune modulator, and recently it was shown to be present in high concentrations in household dust from homes infested with bed bugs. To better understand the dynamics of histamine excretion, we analyzed bed bug fecal material from different life stages and popula…
