Bifocal hydrogel lenses: an overview.

Until the advent of bifocal hydrogel lenses, monovision or reading glasses were the modes of correction for hydrogel contact lens wearing presbyopes. Recent design innovations and improvements in the manufacturing technology of soft contact lenses have made possible new developments in bifocal hydrogel lenses. Patient selection and patient education play key roles in optimizing patient success. There are several hydrogel bifocal designs that are possible for the presbyope. The variations of these designs and their pertinent considerations are discussed.

Tripeptide-based amyloid hydrogels: phenylalanine mutation outcomes on self-assembly, hydrogelation and their cytotoxicity assays.

A tripeptide segment Leu-Val-Phe (Aβ17-19) has been judiciously selected from the core β-amyloid (Aβ1-42) peptide sequence. Seven tripeptides with the general sequence Boc-Leu-Val-X-COOH (Boc-LVX-OH), where X = L-phenylalanine, L-4-nitro-phenylalanine, L-tyrosine, L-3-nitro-tyrosine, L-tryptophan, L-valine, and γ-amino butyric acid for AP1 to AP7, respectively, have been synthesized to study their aggregation behavior, hydrogel forming abilities and corresponding cytotoxicity. Stable hydrogelation for Boc-LVF-OH (AP1) and Boc-LV(4-nF)-OH (AP2), weak hydrogelation for Boc-LVY-OH (AP3), transien

Poly(ethylene) glycol hydrogel based on oxa-Michael reaction: Precursor synthesis and hydrogel formation.

This paper reported a facile strategy for the one-pot synthesis of vinyl sulfone (VS) group terminated hydrogel precursors [poly(ethylene) glycol (PEG)-VS] and PEG hydrogels via catalytic oxa-Michael reaction. Nine potential catalysts were investigated for the reaction between PEG and divinyl sulfone, among which 4-dimethylaminopyridine (DMAP) prevailed for its high catalytic activity. DMAP produced PEG-VS with a conversion of more than 90% in 2 h under a solvent-free condition at room temperature, which significantly simplifies the synthesis of PEG-VS. The preparation of PEG hydrogels was rea…

Light-responsive liposomes for on-demand degradation of cellulose-based hydrogels.

Nanocellulose hydrogels have limited biodegradability under physiological conditions, restricting applications that require controlled scaffold degradation or removal. Here, we present a light-triggered liposomal hydrogel platform that enables localized, on-demand release of cellulase enzyme to induce controlled degradation of anionic cellulose nanofiber (aCNF) hydrogels and cellulose-polysaccharide interpenetrating polymer network (IPN) hydrogels. Cellulase was encapsulated in reactive oxygen species (ROS)-sensitive stimuli-responsive liposomes containing a photosensitizer and incorporated in…

Native Chemical Ligation-Driven Formation of Hyaluronic Acid-Based Injectable Hydrogels.

We report the development of an injectable hyaluronic acid (HA)-based hydrogel using native chemical ligation (NCL) as a biocompatible and chemoselective crosslinking strategy for potential biomedical applications. High-molecular-weight HA was pre-modified with a glycinyl thioester derivative, while a bis-cysteine peptide served as the crosslinker, enabling rapid hydrogel formation under physiological conditions. After optimization of NCL parameters, the resulting hydrogel exhibits all key attributes for biomedical hydrogel injectables: cytocompatibility, injectability, tunable rheology adapte

Deamidation-enabled fabrication of tough, stretchable, and transparent plant protein hydrogels.

Plant protein hydrogels are promising for food structuring and nutrient delivery, yet conventional soybean protein systems suffer from weak mechanical strength due to particulate aggregation. Enzymatic deamidation can dissociate dense aggregates and improve homogeneity, but limited intermolecular connectivity still restricts mechanical performance of soybean protein hydrogels. Here, a sequential strategy involving protein-glutaminase (PG)-induced deamidation and transglutaminase (TGase)-mediated crosslinking was developed. PG promoted protein unfolding and aggregate reorganization, generating …

Sea Cucumber-Inspired Double-Network Biopolymer Hydrogels for Programmable Soft Machines.

Engineering biopolymer-based hydrogels with robust load-bearing capacity, reversible mechanical programmability, and adaptive actuation remains difficult due to weak physical crosslinking and porous structure. Inspired by the mutable dermis of sea cucumbers, a collagen-alginate double-network elastomeric (CoDE) hydrogel integrates a covalently crosslinked collagen cryogel scaffold with an ionically crosslinked alginate sacrificial network. This hydrogel system is inherently biocompatible and biodegradable. Compression-induced scaffold densification and ionic locking enable broad tuning of the …

Biomanufacturing Vascular Matrix-Mimetic Hydrogels from Quantified Arterial Micromechanics and Microstructure.

The microstructural and micromechanical properties of the arterial extracellular matrix (ECM) are central to vascular cell behavior but are disrupted by disease and aging. Elastin and fibrillar collagens provide essential cues, yet the effects of disease-associated ECM remodeling on smooth muscle function remain poorly defined. Here, we imaged fibrillar ECM components across layers of healthy and hypertensive arteries in a large animal model and quantified their stiffness with submicron and piconewton resolution. These animal-derived data guided the fabrication of artery-mimetic hydrogels usin…

Nasal In Situ Hydrogels: A Boon For Anti-Emetic Drug Delivery.

Nasal route presents an alternative avenue for achieving local or systemic drug effects owing to its non-invasiveness, expansive surface area, rapid drug absorption, and circumvention of first-pass effect. It offers a promising route for brain targeting too, as it bypasses the blood-brain barrier, thus facilitating direct nose to brain drug delivery. However, traditional routes of administering anti-emetic drugs, primarily oral or parenteral, suffer from drawbacks like poor patient compliance and low drug bioavailability. The nasal route is a favorable alternative, albeit limited by the short

Tumor microenvironment-responsive injectable hydrogels for localized therapy of pancreatic cancer.

To address the clinical need for localized therapy for pancreatic cancer, an injectable, self-healing nanocomposite hydrogel (GPPA-Mn@DOX) was rationally designed and developed. The hydrogel consists of phenylboronic acid-modified gelatin (Gel-PBA) and polyvinyl alcohol (PVA), which are crosslinked through dynamic boronate ester bonds and loaded with doxorubicin (DOX)-containing hollow mesoporous MnO2 (H-MnO2) nanocarriers. This integrated design enables tumor microenvironment (TME)-responsive drug release while facilitating in situ O2 generation. The resulting network exhibits a three-dimensi

Photocrosslinkable and ultrasound-responsive composite hydrogels for osteoporotic bone defect repair.

Osteoporotic bone defects pose a threat to the lives of senior citizens, leading to prolonged hospitalization and increased cardiovascular risks due to imbalances in the bone tissue microenvironment. This study established a multifunctional composite system integrating ultrasound-responsive microsphere carriers and photocrosslinkable hydrogel substrates. The microsphere with PVP/ALG (1 : 1) group exhibited ultrasound‑responsive behavior; such feature was not detected for PVP/ALG = 3 : 1 and PVP/ALG = 1 : 3. Control over the Self-Assembly of Supramolecular Hydrogels Using Hofmeister Effect.

Supramolecular hydrogels are usually prepared in aqueous media containing diverse ions, for instance, buffer solutions, yet the influences of ions, especially the Hofmeister effect, on their self-assembly are often overlooked. Here, we systematically investigate the impacts of different anions in the Hofmeister series on the self-assembly of supramolecular hydrogels. On the basis of a hydrazone formation-mediated supramolecular gelation system, we found that the addition of kosmotropic anions can accelerate the self-assembly of gelators as determined by rheology and critical gelation concentra…

The selective permeability of mucin hydrogels is modulated by nanoplastic contaminations.

The mucosa lining all wet epithelial tissues in our body is mainly established by mucin glycoproteins and constitutes the first line of defense that controls the selective uptake of substances into sub-mucosal tissues. However, this biological barrier faces a broad range of environmental pollutants. Micro- and nanoplastics are ubiquitous contaminants that occur in our food and beverages, and they are still used as additives in cosmetics and healthcare products. Whereas there are clear indications that an exposure to microplastics can create health issues for humans, little is known about the m

Tough yet low-hysteresis conductive hydrogels for energy-efficient wearable electronics.

Conductive hydrogels combining high toughness and low hysteresis are crucial for soft electronics but remain difficult to achieve due to the intrinsic conflict between energy dissipation and elastic recovery in polymer networks. Here, we report a synergistically engineered conductive hydrogel based on a copolymer network of acrylamide (AM) and N-acryloyl tris(hydroxymethyl)aminomethane (THMA), reinforced by MXene nanosheets and a hydrophobic ionic liquid, 1-hexadecyl-3-methylimidazolium bromide ([C16mim]Br). The ionic liquid introduces dynamic hydrophobic associations and ionic interactions th…

Hydrogels as Promising Carriers for Ophthalmic Disease Treatment: A Comprehensive Review.

Ocular disorders such as keratitis, glaucoma, age-related macular degeneration (AMD), diabetic retinopathy (DR), and dry eye disease (DED) are highly prevalent worldwide and remain major causes of visual impairment and blindness. Conventional therapeutic approaches for ocular diseases, such as eye drops, surgery, and laser therapy, are frequently hampered by limited drug bioavailability, rapid clearance, and treatment-related complications, primarily due to the eye's unique anatomical and physiological barriers. Hydrogels, characterized by their three-dimensional network structure, high water …

Tailored construction and functional applications of conductive hydrogels for bioelectronic interfaces.

Conductive hydrogels have emerged as core foundational materials for bioelectronic interfaces, uniquely bridging the mechanical and electrical property gap between biological tissues and electronic devices. Unlike previous reviews that primarily focus on material classification and broad device applications, this review adopts an interface-centric perspective, specifically targeting the performance optimization of conductive hydrogels for bioelectronic interface applications. We first categorize conductive hydrogels according to their underlying conductive mechanisms, and describe the morpholo…

Ultrasound-Responsive Hydrogels for Accelerated Wound Healing through Programmable Microenvironment Modulation.

Wound healing involving persistent inflammation and fibrosis is driven by reactive oxygen species (ROS) and Yes-associated protein (YAP), often leading to scars. A polyvinyl alcohol/alginate ultrasound-responsive hydrogel was developed, containing free epigallocatechin gallate (EGCG) in the matrix and EGCG-verteporfin (VP) nanoparticles embedded in the hydrogel network. Free EGCG is released rapidly, whereas EGCG-VP nanoparticles release is triggered by ultrasound. In vitro, this system reduced ROS by ∼69%. Western blot revealed ∼71% reduction in α-smooth muscle actin and ∼47% in YAP. In vivo,
