Isolation and characterization of ACC deaminase-producing fluorescent pseudomonads, to alleviate salinity stress on canola (Brassica napus L.) growth.

Salinity stress is of great importance in arid and semi-arid areas of the world due to its impact in reducing crop yield. Under salinity stress, the amount of 1-aminocyclopropane-1-carboxylate (ACC), a precursor for ethylene production in plants, increases. Here, we conducted research under the hypothesis that isolated ACC deaminase-producing Pseudomonas fluorescens and Pseudomonas putida can alleviate the stressful effects of salinity on canola (Brassica napus L.) growth. The experiments were conducted in the Soil and Water Research Institute, Tehran, Iran. Seven experimental stages were cond

A Chinese cabbage (Brassica campetris subsp. Chinensis) τ-type glutathione-S-transferase stimulates Arabidopsis development and primes against abiotic and biotic stress.

The beneficial root-colonizing fungus Piriformospora indica stimulates root development of Chinese cabbage (Brassica campestris subsp. Chinensis) and this is accompanied by the up-regulation of a τ-class glutathione (GSH)-S-transferase gene (BcGSTU) (Lee et al. 2011) in the roots. BcGSTU expression is further promoted by osmotic (salt and PEG) and heat stress. Ectopic expression of BcGSTU in Arabidopsis under the control of the 35S promoter results in the promotion of root and shoot growth as well as better performance of the plants under abiotic (150 mM NaCl, PEG, 42 °C) and biotic (Alternari

Vitamin D deficiency in early life regulates gut microbiome composition and leads to impaired glucose tolerance in adult and offspring rats.

Vitamin D has been found to be involved in glucose metabolism in recent years. Its deficiency is very common, especially in children. Whether vitamin D deficiency in early life affects adult diabetes risk is unknown. In this study, a rat model of early life vitamin D deficiency (F1 Early-VDD) was established by depriving it of vitamin D from the 0 to the 8th week. Further, some rats were switched to normal feeding conditions and sacrificed at the 18th week. Other rats were mated randomly to generate offspring rats (F2 Early-VDD), and F2 rats were fed under normal conditions and sacrificed at t

Transcriptome Analysis of Pyrus betulaefolia Seedling Root Responses to Short-Term Potassium Deficiency.

Potassium (K) plays a crucial role in multiple physiological and developmental processes in plants. Its deficiency is a common abiotic stress that inhibits plant growth and reduces crop productivity. A better understanding of the mechanisms involved in plant responses to low K could help to improve the efficiency of K use in plants. However, such responses remain poorly characterized in fruit tree species such as pears (Pyrus sp). We analyzed the physiological and transcriptome responses of a commonly used pear rootstock, Pyrus betulaefolia, to K-deficiency stress (0 mM). Potassium deprivation

Early-life microbial contact with adult hens modulates cecal microbiota and mitigates host responses to lipopolysaccharide-induced stress in broilers.

BACKGROUND: In commercial production, the absence of adult hens disrupts natural gut colonization, forcing chicks to rely on environmental microbes and potentially compromising the ecological succession essential for health. This study investigated whether early-life adult hen microbial contact could optimize the intestinal community and metabolite profiles, thereby mitigating lipopolysaccharide (LPS)-induced immunological stress in broilers. Accordingly, 128 one-day-old broilers were assigned to control or adult hen microbial contact groups. Hens were removed on D 10, and LPS was administered

Auxin-Activated Caspase 9 System (AuxiCasp9), a Suicide Switch for Engineered Cells.

Engineered cells incorporating synthetic gene circuits have been developed for therapeutic and diagnostic applications. However, cell proliferation and long-term engraftment may pose a risk of unpredictable toxicities. To address this issue, a suicide switch that can be selectively activated by exogenous stimuli has been developed as a regulatory system for engineered cells. However, the variety of reliable suicide switches is limited and insufficient for the orthogonal integration of multiple functions in engineered cells. This study developed auxin-activated caspase 9 (AuxiCasp9), a novel au

CaMACPF1 and CaMACPF6 positively regulate hypoxia tolerance through modulating jasmonate and auxin metabolic pathways.

Submergence and flooding impose severe crop losses through hypoxic stress. The Membrane Attack Complex/Perforin (MACPF) superfamily proteins function as pore-forming effectors in development and immunity across eukaryotic organisms, yet their role in plant hypoxia responses remains largely unexplored. Here, we identify two MACPF proteins in pepper (Capsicum annuum), CaMACPF1 and CaMACPF6, as positive regulators of hypoxia tolerance. CaMACPF1 and CaMACPF6 localize to endoplasmic reticulum-plasma membrane contact sites (EPCS) and the Golgi apparatus, with CaMACPF1 additionally detected in the nu

Co-option of a conserved lateral-root development program by symbiotic signals.

Nod factors (NFs) are microbial signals originally identified for their key role in the nitrogen-fixing root nodule symbiosis in legumes. Beyond symbiosis, NFs also possess a conserved capacity to induce lateral-root formation across diverse plant species, including non-legumes. It is now well established that the nodule organogenesis program has co-opted several molecular mechanisms involved in root development, which raises the question of the developmental pathway controlled by NFs to trigger lateral-root formation and how it overlaps with nodule organogenesis in legumes. In Medicago trunca

Beneficial microbes isolated from the strawberry rhizosphere: development of growth-promoting and anthracnose-suppressive SynComs.

Soil rhizosphere microorganisms play crucial roles in the regulation of plant growth and disease resistance. This study aimed to construct stable growth-promoting and anthracnose-suppressive synthetic microbial communities (SynComs) using beneficial microbes isolated from the strawberry rhizosphere. Reanalysis of published high-throughput sequencing data confirmed that the core rhizosphere microbial communities of strawberry across different substrates were dominated by phyla such as Proteobacteria, Actinobacteria, and Bacteroidetes, and genera Pseudomonas and Streptomyces (typical plant growt

Auxin-induced ARF transcription factor degradation defines tissue boundaries.

How organs partition themselves into discrete domains with distinct functions is a fundamental question in biology. The gynoecium of flowering plants provides an excellent system to address this question. Here, we show that the boundary between the stigma and style at the gynoecium apex is established by the complementary distribution of the phytohormone auxin and the Auxin Response Factor (ARF), ETTIN (ETT). Mechanistically, auxin induces ETT protein destabilization via the ubiquitin-proteasome pathway. A short sequence motif within an intrinsically disordered region is required for this auxi

Combined effect of microbial and non-microbial biostimulants on wheat growth, yield, and grain nutritional quality under sandy soil conditions.

BACKGROUND: The agricultural use of sandy soils has expanded worldwide to meet the increasing demand for food and other agricultural commodities resulting from rapid population growth and urbanization. However, the inherent limitations of these soils, particularly their low capacity to retain water and nutrients, pose major challenges to sustainable crop production. Accordingly, field experiments were conducted to assess the effectiveness of integrated microbial and non-microbial biostimulants in improving the performance of wheat (Triticum aestivum L. cv. Sakha-94). Wheat grains were co-inocu

Loss of galactan synthesis in pea (Pisum sativum) causes defects in organ expansion and is associated with increased extensin content.

Pectic galactan is a rhamnogalacturonan-I side chain, composed of β-(1→4)-linked D-galactose residues. Its multifunctional role in cell wall assembly and properties has been proposed in various developmental contexts. In this study, we investigated the role of galactan in pea (Pisum sativum syn. Lathyrus oleraceus). First, we used glycan profiling to examine stems with different auxin-induced elongation rates for changes in cell wall composition. The galactan epitopes, recognised with the LM5 monoclonal antibody, were more abundant in the convex, more elongated part of the pea stem. This findi

Auxin Gradients Determine Reproductive Development in Pea (Pisum sativum).

Auxins are involved in the regulation of fruit set and development; however, the role of IAA is unclear in pea (Pisum sativum) since the endogenous auxin 4-Cl-IAA appears to be the auxin stimulating ovary (pericarp) growth. To further understand the role of auxins during fruit development, auxin localization, quantitation, transport, and gene expression activity were assessed in this model legume species. IAA levels and auxin activity (DR5::β-Glucuronidase [GUS] staining and enzyme activity) were substantially reduced in the pericarp vascular tissues, pedicels, and peduncles of fruit upon seed

The Metabolite Indole-3-Acetic Acid of Bacteroides Ovatus Improves Atherosclerosis by Restoring the Polarisation Balance of M1/M2 Macrophages and Inhibiting Inflammation.

Emerging research has highlighted the significant role of the gut microbiota in atherosclerosis (AS), with microbiota-targeted interventions offering promising therapeutic potential. A central component of this process is gut-derived metabolites, which play a crucial role in mediating the distal functioning of the microbiota. In this study, a comprehensive microbiome-metabolite analysis using fecal and serum samples from patients with atherosclerotic cardiovascular disease and volunteers with risk factors for coronary heart disease and culture histology is performed, and identified the core st

Harnessing PGPRs from Asparagus officinalis to Increase the Growth and Yield of Zea mays L.

Microbial biotechnology employs techniques that rely on the natural interactions that occur in ecosystems. Bacteria, including rhizobacteria, play an important role in plant growth, providing crops with an alternative that can mitigate the negative effects of abiotic stress, such as those caused by saline environments, and increase the excessive use of chemical fertilizers. The present study examined the promoting potential of bacterial isolates obtained from the rhizospheric soil and roots of the Asparagus officinalis cultivar UF-157 F2 in Viru, la Libertad, Peru. This region has high soil sa
