Does physical activity and inflammation mediate the job stress-headache relationship? A sequential mediation analysis in the ELSA-Brasil study.

BACKGROUND: Evidence indicates that physical activity reduces stress and promote a myriad of health-enhancing effects through anti-inflammatory mechanisms. However, it is unknown whether these mechanisms interfere in the association between psychosocial job stress and headache disorders. OBJECTIVE: To test whether physical activity and its interplay with the systemic inflammation biomarkers high-sensitivity C-reactive protein (hs-CRP) and acute phase glycoproteins (GlycA) would mediate the associations between job stress and headache disorders. METHODS: We cross-sectionally evaluated the bas

Peripheral inflammation mediates midbrain Lrrk2 kinase activity via Rab32 expression.

Mutations that increase leucine-rich repeat kinase 2 (LRRK2) activity confer significant risk for Parkinson's disease (PD), yet incomplete disease penetrance suggest additional factors are required to manifest disease. We recently identified RAB32 Ser71Arg as a Mendelian gene for PD. Here, we establish Rab32 as a key mediator linking peripheral inflammation to Lrrk2 activation. We show that Rab32 and Rab38 expression are modestly, but inversely, correlated with their homolog Rab29. In vivo , peripheral lipopolysaccharide (LPS)-induced inflammation selectively induced Rab32 expression in midbra

Gut Microbiota-Derived Ursodeoxycholic Acid Mediates the Resistance to Colonic Inflammation in Pigs.

Microbes in the gut are crucial for host health, yet their role in disease resistance remains unclear. Using fecal microbiota transplantation from disease-resistant Min pigs to Duroc × Landrace × Yorkshire (DLY) pigs, combined with 16S rRNA sequencing and metabolomics, we investigated this relationship. The transferred microbiota alleviated lipopolysaccharide-induced intestinal inflammation and barrier damage in the DLY piglets. Key bacterial genera and bile acid metabolites have been identified, with in vitro evidence showing that the gut microbiome can convert bile acids to secondary forms,

USP43 Inhibits Intestinal Inflammation via TRAF4-mediated NF-κB Signaling.

Ulcerative colitis (UC) is a chronic inflammatory intestinal disorder; however, its exact pathophysiological mechanisms remain elusive. Through integrated RNA-seq and proteome analyses, we revealed that ubiquitin-specific protease 43 (USP43) expression is significantly decreased in dextran sulfate sodium (DSS)-induced colitis. USP43 expression was also lower in UC patients than that in healthy controls. Furthermore, USP43 deficiency in mice exacerbated DSS-induced colitis. In lipopolysaccharide (LPS)-treated RAW264.7 cells, USP43 overexpression inhibited, whereas USP43 knockdown promoted the p

Kidney Allograft Outcomes in Borderline and T Cell-Mediated Rejection With Microvascular Inflammation.

INTRODUCTION: Microvascular inflammation (MVI) is a hallmark feature of antibody-mediated rejection (ABMR), but its prognostic significance when it occurs with T-cell-mediated rejection (TCMR) in the absence of other ABMR features remains unclear. METHODS: We examined the association between pure TCMR phenotypes, defined by the presence or absence of MVI, and graft survival using restricted mean survival time (RMST), and assessed interactions with rejection involving a "v" lesion. RESULTS: Among 1614 recipients with first biopsy-proven pure TCMR (2010-2023), 85% had no MVI, 8% subthreshold M

[Mechanisms of cGAS-STING-mediated chronic inflammation and targeted interventions in brain aging].

Brain aging is a critical risk factor for various neurodegenerative diseases, with chronic neuroinflammation being one of its core features. The innate immune cGAS-STING signaling pathway, acting as a key sensor of endogenous damage, has emerged as a nexus linking cellular senescence to chronic inflammation. During brain aging, genomic damage and mitochondrial dysfunction facilitate the aberrant accumulation of cytosolic double-stranded DNA. Together with transposable element activation and intercellular propagation of cGAMP, these factors drive sustained activation of the cGAS-STING signaling

Dendritic Cell-Derived Extracellular Vesicles Mediate Inflammation in Egg Allergy Patients.

Atopic allergy is rising globally and placing a significant strain on healthcare systems, yet the understanding of the underpinning mechanisms of allergic sensitization remains incomplete. Extracellular vesicles (EVs) have recently emerged as important mediators of immune modulation, due to their diverse cargo, and therefore may play a mechanistic role in allergic sensitization development. Thus, this study investigated whether EVs released by activated dendritic cells (DCs) contribute to allergic sensitization of the common egg allergen, ovalbumin (OVA). DCs were generated from human monocyte

Adiposity and inflammation mediate altered metabolic profile in individuals with opioid use disorder.

Previous studies have linked opioid use to altered metabolic profiles, but findings have been inconsistent and mechanisms remain unclear. One potential mechanism involves increased adiposity, leading to chronic low-grade inflammation that elevates metabolic risk. Here, we examined metabolic profiles in individuals with opioid use disorder (OUD) and matched non-OUD controls, focusing on the sequential mediating roles of BMI and inflammation. Data from individuals with OUD (n = 281) and non-OUD (n = 246) were drawn from a natural history screening protocol from the National Institute on Alcohol

RADX protects against intestinal inflammation by restraining IFI16-mediated innate immunity.

Genomic instability is increased in patients with inflammatory bowel disease (IBD), yet whether it contributes directly to disease pathogenesis remains unclear. Here, we identify RADX, a structural antagonist to RAD51 and a key regulator of replication fork stability, as a critical suppressor of intestinal inflammation by limiting innate immune sensing of replication-associated DNA damage. RADX deficiency exacerbates experimental colitis, with macrophages serving as the principal mediators of this phenotype. Mechanistically, RADX competes with the DNA sensor IFI16 for binding to single-strande

Antibiotic-induced disruption of lipid mediator signaling: Implications for cardiovascular inflammation and cardiotoxicity.

Antibiotics remain the cornerstone of treatment for infectious disease, yet a growing body of evidence shows that these drugs also disrupt host inflammatory and metabolic pathways well beyond their intended antimicrobial target. One consequence that has received comparatively little attention is their effect on lipid mediator signaling. Polyunsaturated fatty acids serve as precursors to lipid mediators that keep the vasculature in balance by governing both inflammatory and resolving pathways. Antibiotic exposure disturbs this balance in two connected ways, by driving gut microbiota dysbiosis a

PCK2-Mediated PQBP1 Lactylation Promotes Asthmatic Inflammation through PRMT5 Inhibition.

Based on their immunodominant nature and ability to induce appropriate immune responses in the host, several antigens of Mycobacterium tuberculosis have shown promise of protection. However, most of the candidate vaccines developed by employing various strategies have afforded protection that is at best comparable with bacillus Calmette-Guérin (BCG) in animal models. Due to the inherent ability of BCG to prime cellular responses in the host, it has become an attractive vehicle for development of a vaccine against intracellular infections. In this study, we have cloned the genes of three immuno

HDAC9 drives intervertebral disc degeneration by potentiating STAT1-mediated senescence and inflammation.

BACKGROUND: Intervertebral disc degeneration is a leading cause of chronic low back pain, characterized by cellular senescence, extracellular matrix disintegration, and chronic inflammation. However, the key epigenetic regulators that orchestrate senescence and the resultant inflammatory cascade in disc degeneration remain poorly understood. OBJECTIVE: This study aimed to investigate the pathological role and molecular mechanism of HDAC9 in IDD. METHODS: We analyzed HDAC9 expression in aged degenerative discs. Using siRNA-mediated knockdown in primary cells, we assessed senescence, ECM, and

MZB1 Drives Periodontitis via ER Stress-Mediated Inflammation and Osteogenic Differentiation.

INTRODUCTION AND AIMS: Increasing evidence suggests that marginal zone B and B1 cell-specific protein (MZB1) plays crucial roles in inflammatory responses, endoplasmic reticulum (ER) stress, and cell survival, but its function in periodontitis is still unclear. Periodontitis is a chronic inflammatory disease. This study aimed to investigate the expression profile and functional role of MZB1 in periodontitis. METHODS: Transcriptomic analysis and RT-qPCR were performed to evaluate MZB1 expression in gingival tissues of periodontitis patients. An in vitro inflammatory model of periodontal ligame

IL-27 signaling mediates skin inflammation in experimental psoriasis and atopic dermatitis.

BACKGROUND: Psoriasis and atopic dermatitis (AD) are two prevalent inflammatory skin disorders, each characterized by distinct adaptive immune responses. However, recent evidence suggests that these diseases may share overlapping immune mechanisms, especially concerning keratinocyte function. The specific cytokines that coordinate these inflammatory pathways remain largely undefined. METHODS: The expression of IL-27 and its receptor was analyzed using data derived from GEO datasets. Imiquimod-induced psoriasis-like and MC903-induced AD-like skin inflammation models were established in wild-ty

Depletion of p75NTR in Schwann Cells Driven by Inflammation Mediates Cutaneous Pain in Psoriasis.

Skin pain is a common but poorly understood symptom of psoriasis, affecting only a subset of patients. Using imiquimod and interleukin-17A-induced psoriasiform mouse models that exhibited pain-like behaviors, we found that nerve growth factor (NGF) levels were elevated in lesional skin, activating TrkA signaling in dorsal root ganglion neurons and promoting Schwann-cell hypertrophy. Normally, Schwann cells (SCs) limit NGF signaling in cutaneous peripheral nerves through the p75NTR receptor. However, inflammation driven by interleukin-17A increased non-muscle myosin II activity and elevated NGF

Epidermal NAD+ deficiency induces IL-36-mediated skin inflammation and acanthosis.

Nicotinamide adenine dinucleotide (NAD+) is essential for cellular metabolism, DNA repair, and stress responses. NAD+ is synthesized from nicotinamide, nicotinic acid (collectively termed niacin), and tryptophan. In humans, deficiencies in these nutrients result in pellagra, marked by dermatitis, diarrhea, and dementia. The dermatitis associated with pellagra typically manifests as photodermatosis in sun-exposed areas. This study examined the effects of NAD+ deficiency on skin homeostasis using epidermis-specific Nampt-conditional KO (Nampt-cKO) mice. These mice displayed substantial NAD+ depl

Altered expression of the CD26/ADA axis in immune-mediated inflammation of infectious mononucleosis.

Infectious mononucleosis (IM), a common disease in children, was often accompanied by excessive inflammatory response and organ damage. This study aimed to explore the CD26/ADA axis in the pathogenesis of acute Epstein-Barr virus (EBV)-associated IM. Thirty children with acute IM and thirty age-matched controls from Children’s Hospital of Soochow University were enrolled in this study. The CD26 and adenosine deaminase (ADA) mRNA were detected by real-time PCR in the peripheral blood. Meanwhile, the levels of plasma interferon gamma (IFN-γ), interleukin (IL)-4 and IL-10 were measured by ELISA,

BMI trajectories are associated with NAFLD and advanced fibrosis via aging-inflammation mediation.

BACKGROUND: As the global epidemic of obesity fuels metabolic conditions, the burden of nonalcoholic fatty liver disease (NAFLD) will become enormous. Abundant studies revealed the association between high body mass index (BMI) and NAFLD but overlooked the BMI patterns across life stages. We aimed to explore how BMI trajectories over age relate to NAFLD. METHODS: Selecting 3212 participants in NHANES 2017-2020, we tracked BMI records at different ages. Using a latent class trajectory model (LCTM), we identified BMI trajectories over age. Multinomial logistic regression assessed their associat
