Venom from the parasitoid wasp Pimpla hypochondriaca has potent in vivo activity against insect haemocytes and disrupts host immune responses. Using hybridisation techniques, and more recently random sequence analysis, we had previously identified cDNAs encoding 10 venom proteins from this wasp and deduced their primary structures. We have now extended the random sequence analysis and discovered a further nine cDNAs encoding proteins with predicted signal sequences. The mature proteins were calculated to have masses of between 4 and 22 kDa. Post-signal sequence residues predicted from the cDNA

UNLABELLED: Most arboviruses rely on insect vectors for transmission, with salivary glands serving as a critical gateway for viral spread to new hosts. MicroRNAs (miRNAs) are key regulators of gene expression, yet their roles in salivary glands during virus infection remain poorly understood. Using the small brown planthopper (SBPH)-rice stripe virus (RSV) system, we sequenced small RNAs from salivary glands of nonviruliferous and viruliferous insects and identified 5,909 known miRNAs from 201 families. Of these, 1,143 miRNAs were differentially expressed upon RSV infection, including 1,090 up

Rift Valley fever virus (RVFV) is an emerging mosquito-borne virus causing significant morbidity and mortality in livestock and humans. Rift Valley fever is endemic in Africa, but also outside this continent outbreaks have been reported. Here we report the evaluation of two vaccine candidates based on the viral Gn and Gc envelope glycoproteins, both produced in a Drosophila insect cell expression system. Virus-like particles (VLPs) were generated by merely expressing the Gn and Gc glycoproteins. In addition, a soluble form of the Gn ectodomain was expressed and affinity-purified from the insec

The outer surface of insects is covered by the cuticle, which is derived from the apical extracellular matrix (aECM). The aECM is secreted by epidermal cells during embryogenesis. The aECM exhibits large variations in structure, function, and constituent molecules, reflecting the enormous diversity in insect appearances. To investigate the molecular principles of aECM organization and function, here we studied the role of a conserved aECM protein, the ZP domain protein Trynity, in Drosophila melanogaster. We first identified trynity as an essential gene for epidermal barrier function. trynity

As the world's population increases, traditional food sources face various constraints. This has renewed interest in the domestication and value addition of alternative food sources such as edible insects for nutrition and food security, as they are rich in proteins, fats, amino acids, flavonoids, minerals, and vitamins. Among these insects, the larvae of Rynchophorus spp (Coleoptera: Curculionidae) have a demonstrated potential as a sustainable and nutritious food source. Attempts to develop new insect farming techniques, improved harvesting methods, and creation of value-added products from

Low molecular weight compounds are typically used by insects and plants for defence against predators. They are often stored as inactive β-glucosides and kept separate from activating β-glucosidases. When the two components are mixed, the β-glucosides are hydrolysed releasing toxic aglucones. Cyanogenic plants contain cyanogenic glucosides and release hydrogen cyanide due to such a well-characterized two-component system. Some arthropods are also cyanogenic, but comparatively little is known about their system. Here, we identify a specific β-glucosidase (ZfBGD2) involved in cyanogenesis from l

The Anabrus simplex is a swarming plague orthopteran found in western North America. The genome is 15 766 bp in length and genome organization follows the ancestral insect gene arrangement. atp6 lacked any readily identifiable stop codon. Examination of mRNA secondary structure for this gene suggested a stem/loop-mediated mRNA post-transcriptional processing to liberate a mature atp6 mRNA with a complete stop codon produced by polyadenylation. Comparison of similar protein with protein gene boundaries in other insect species reveal a general mechanism for mRNA excision and provide further supp

Euroglyphus maynei is a house dust mite commonly found in homes worldwide and is the source of allergens that sensitize and induce allergic reactions in humans. It is the source of species-specific allergens as well as allergens that are cross-reactive with the allergens from house dust mites Dermatophagoides farinae and D. pteronyssinus, and the ectoparasitic scabies mite Sarcoptes scabiei. The genomics, proteomics and molecular biology of E. maynei and its allergens have not been as extensively investigated as those of D. farinae, D. pteronyssinus, and S. scabiei where natural and recombinan

Genome-scale searches for insect olfactory genes are vulnerable to rapid receptor divergence, tandem duplication, fragmented gene models, circular labels, and evolutionary leakage between training and test data. Sequence foundation models trained on DNA or proteins may extend homology-based annotation, but available evidence does not support their use as autonomous annotators or direct predictors of sensory function. This critical review organizes the problem around annotation failures rather than model catalogues. We distinguish six inference levels, from candidate locus detection to organism

Nipah virus (NiV) is an emerging high-fatality zoonotic threat lacking approved vaccines. Current virus-like particle (VLP) production methods rely on costly mammalian cell systems and non-scalable ultracentrifugation purification. We developed an alternative enveloped Nipah virus-like particle (NiVLP) expression system in Sf9 insect cells by co-expressing structural proteins F, G, and M using a single recombinant baculovirus. A novel multi-step chromatographic purification process was established using monolith convective media, integrating steric exclusion chromatography with sequential cati

To prevent the deterioration of the global environment, the reduction of chemical pesticide use and the development of eco-friendly pest control technologies are urgent issues. Our recent study revealed that the production of reactive oxygen species (ROS) by dual oxidase (Duox) plays a pivotal role in stabilizing the tracheal network by intermediating the tyrosine cross-linking of proteins that constitute trachea. Notably, the formation of dityrosine bonds by ROS can be inhibited by the intake of an antioxidant cysteine derivative N-acetyl-L-cysteine (NAC), which can suppress insect respiratio

The life cycle of Trypanosoma cruzi, the etiological agent of Chagas disease, involves different forms of the parasite, which alternates between insect and vertebrate hosts. One critical process in the parasite's life cycle is metacyclogenesis, in which the replicative non-infective forms present in the insect midgut differentiate into non-dividing vertebrate-infective forms. It is known that proline (Pro) is important for this process and that leucine (Leu) and isoleucine (Ile) can act as inhibitors of metacyclogenesis. In this study, we investigated further the role of branched-chain amino a

Influenza virus is a highly contagious virus that causes significant human mortality and morbidity annually. The most effective drugs for treating influenza are the neuraminidase inhibitors, but resistance to these inhibitors has emerged, and additional drug discovery research on neuraminidase and other targets is needed. Traditional methods of neuraminidase production from embryonated eggs are cumbersome, while insect cell derived protein is less reflective of neuraminidase produced during human infection. Herein we describe a method for producing neuraminidase from a human cell line, HEK293-

Animal venoms are complex mixtures of highly specialized toxic molecules. Cnidarians and arachnids produce pore-forming proteins (PFPs) directed against the plasma membrane of their target cells. Among PFPs from cnidarians, actinoporins stand out for their small size and molecular simplicity. While native actinoporins require only sphingomyelin for membrane binding, engineered chimeras containing a recognition antibody-derived domain fused to an actinoporin isoform can nonetheless serve as highly specific immunotoxins. Examples of such constructs targeted against malignant cells have been alre

An enzyme-linked immunosorbent assay (ELISA) useful for identifying noctuid pests parasitized by hymenopteran endoparasitoids was recently described. The ELISA employed a monoclonal antibody (MAb 9A5) that appeared highly polyspecific for parasitoid antigens, yielding banding patterns more typical of a polyclonal antiserum than of a monoclonal antibody in immunoblots of parasitoid homogenates subjected to SDS-PAGE. Although MAb 9A5 appeared capable of binding to dozens of parasitoid antigens, no cross-reactivity for noctuid antigens was evident by either immunoblotting or ELISA. In the study d

The kissing bug Rhodnius prolixus, a primary vector of Chagas disease, serves as a model for understanding insect physiology and vector-pathogen interactions. While the role of male reproductive factors in influencing female fertility is well-established in many insects, little is known about male fertility in R. prolixus. Our study examines the role of two apolipoproteins, RPRC010099 and RPRC015421, in male fertility and their potential function as carriers of eicosanoid hormones. Using RNA interference (RNAi), we silenced the genes for both apolipoproteins in male R. prolixus. The silencing

Insect respiration has long been thought to be solely dependent on an elaborate tracheal system without assistance from the circulatory system or immune cells1,2. Here we describe that Drosophila crystal cells-myeloid-like immune cells called haemocytes-control respiration by oxygenating Prophenoloxidase 2 (PPO2) proteins. Crystal cells direct the movement of haemocytes between the trachea of the larval body wall and the circulation to collect oxygen. Aided by copper and a neutral pH, oxygen is trapped in the crystalline structures of PPO2 in crystal cells. Conversely, PPO2 crystals can be dis

Plants are central to global food production, and the pursuit of sustainability aims to enhance or preserve food quality while safeguarding the environment. Due to their immobility, plants are unable to evade unfavourable climatic setups or interactions with other living creatures. Upon their interaction with insect herbivores, plants face biotic stress, which is a constant challenge for plants, causing molecular, physiological, and biochemical changes and reducing their productivity. To combat biotic stress caused by herbivores, plants have evolved intricate defence mechanisms through growth

In this project, the uptake mechanisms and localization of two lectins from Sambucus nigra, further referred to as S. nigra agglutinin (SNA)-I and SNA-II, into insect midgut CF-203 cells were studied. SNA-I is a chimeric lectin belonging to the class of ribosome-inactivating proteins, whereas SNA-II is a hololectin devoid of enzymatic activity. Internalization of the fluorescein isothiocyanate-labeled lectin was investigated using confocal microscopy. Both lectins were internalized into the cytoplasm of CF-203 cells at similar rates. Preexposure of the insect midgut cells to specific inhibitor

Anthrax is a major zoonotic disease of wildlife, and in places like West Africa, it can be caused by Bacillus anthracis in arid nonsylvatic savannahs, and by B. cereus biovar anthracis (Bcbva) in sylvatic rainforests. Bcbva-caused anthrax has been implicated in as much as 38% of mortality in rainforest ecosystems, where insects can enhance the transmission of anthrax-causing bacteria. While anthrax is well-characterized in mammals, its transmission by insects points to an unidentified anthrax-resistance mechanism in its vectors. In mammals, a secreted anthrax toxin component, 83 kDa Protective
